Tørkeperioder kan gjøre bakken hardere og frastøte seg vann, dette fører til økt fare for flom og jorderosjon hvis det skulle komme styrtregn. Sammenfallende hendelser hvor en tørkeperiode ender i styrtregn kan derfor forårsake større skader på infrastruktur og avlinger enn enkelthendelser med kun tørke eller styrtregn. Global oppvarming har forårsaket at både tørkeperioder og styrtregn intensiveres og skjer oftere, som også kan føre til at sammenfallende hendelser med tørke etterfulgt av styrtregn også kan intensiveres og skje oftere. 
Selv om det er funnet en økning i tørkeperioder og ekstremnedbør med global oppvarming også i klimamodellene, er det også en stor spredning mellom modellene på hvor mye og i hvilke områder som opplever mer tørke og nedbør. Prosjektet vil undersøke faktiske historiske hendelser hvor tørkeperioder endte i styrtregn i værobservasjoner og værmodelldata for å finne passende indekser som kan brukes til å analysere hvordan de forekommer i klimamodellene. Modeller på finere skala gjør en bedre jobb med å modellere nedbør, prosjektet vil derfor bruke finskala høytoppløselige klimamodeller og globale klimamodeller for å se hvordan tørkeperioder avsluttet med styrtregn forekommer i disse. De modellene som gjør det godt opp mot observasjoner vil bli identifisert.  Videre vil prosjektet studere endringene i hyppighet og intensitet av sammenfallende hendelser med tørke og styrtregn med global oppvarming.
En av grunnene man vet til hvorfor det er stor usikkerhet knyttet til hvordan klimamodellene gjenskaper nedbørsmønstre er hvordan fordamping varierer over skog- og andre arealer. I prosjektet vil vi se på effekten av avskoging og økt skogbruk i globale modeller, og gjøre egne beregninger over mindre areal på finskala for å for å se hvordan fordamping blir påvirket. Resultatene av prosjektet er viktig får å unngå eller minimere skadene slike sammenfallende med tørke etterfulgt av styrtregn kan forårsake.

Global warming intensifies drought and extreme precipitation. Compound events with droughts ending in intensified precipitation - DRIP can cause large damages but are extremely understudied. This project aims to estimate the change in frequency and intensity of DRIP events with global warming due to changes in the water-cycle. We will identify actual historical compound DRIP events and find indices that are suitable to identify these events in model data. As global climate models show a large spread in precipitation for both historical and the different future climate projections, there is expected to be a spread in model projections of DRIP events as well. We will constrain this spread by identifying global climate models that do a good statistical representation of historical DRIP events and by analyzing large ensembles to evaluate the natural variability. As higher resolution modelling shows a better representation of the water cycle, the project will analyze regional downscaled global models in identified regions of interest.  We will also perform in-house regional simulations of identified global models down to convection permitting scale to study the change in DRIP events with model resolution. The project will also target a highlighted uncertainty connected to the water-cycle by exploring how evapotranspiration and the occurrence of DRIP events found in climate models respond to land use change. For this analysis we will extend the regional downscaling to include sensitivity studies which contain deforestation and forestation. Results gained from DRIP will make a large contribution to the knowledge needed on future mitigation measures due to the increase in compound events.

DRIP - DRought ending in Intensified Precipitation

Renewable energy on the high seas – science-informed and indigenous knowledge-based recommendations for regulatory and governance challenges

The high seas are those marine areas under no State's jurisdiction. So far, all marine renewable energy (RE) activity is being conducted under State's jurisdiction. However, recent advances in technology are raising the prospect of carrying out RE activity on the high seas. Currently, only seabed mining activities in the Area have been regulated. The Area is the seabed and ocean floor beyond the limits of State's jurisdiction. From the legal viewpoint, the main gap is a lack of an international regulatory framework, informed by participatory mechanisms and supported by scientific evidence, for RE activities on the high seas, especially regarding their environmental and social impacts. 
The primary project objective (O) is to provide the scientific community and policymakers with science-informed and indigenous knowledge-based recommendations in support of the development of  RE regulation on the high seas. 
Two specific Os are set to complement the primary O, namely:
O1 To advance the use of systematic consultation processes that facilitates research at the crossroads of marine sciences and law. 
O2 To advance critical knowledge and understanding of environmental pressing issues comparable to the regulation of RE on the high seas, such as deep-sea mining in the Area.
A multistep innovative methodology will be adopted, by integrating marine scientific knowledge, consultative processes, and advanced critical research in the subject matter. A case study (interdisciplinary and participatory) involving marine scientists, indigenous and coastal communities and policy makers will be conducted onboard the Research Vessel KronPrins Haakon at the beginning of the project. The case study outcomes will be analysed through critical legal analysis to build a knowledge base for policy recommendations. 
The project will advance the PI’s career in an international research environment, allowing her to generate independent, high-quality, and reproducible results.




Prosjektet går ut på å invitere forskere fra norske eller utenlandske universiteter til å presentere sin forskning om økonomi og klima ved Handelshøyskolen ved Universitetet i Stavanger (HHUiS).

Dette skjer i form av et seminar som vanligvis finner sted en gang i uken. En kjent forsker fra et annet universitet kommer til Stavanger og holder et 60-minutters seminar og har en rekke møter med forskere i Stavanger. Vi prøver å ha minst 6-8 møter per gjest. I tillegg spiser gjesten lunsj og middag med UiS-ansatte.

Disse seminarene er en del av en eksisterende seminarserie som også tar for seg andre klimaspørsmål. En stor del av seminarene er imidlertid fokusert på klimaendringer og økonomi, og det er kun til disse seminarene vi søker midler.

En aktiv seminarrekke er hjertet i et forskningsuniversitet, og svært viktig for at forskerne ved universitetet skal kunne utvikle seg og være en aktiv del av det globale forskningsmiljøet. Dessverre må vi av budsjettmessige årsaker kutte ned på denne aktiviteten i år og fremover, og derfor sender vi denne søknaden for å be om midler til denne aktiviteten.

UiS har et meget energisk forskerteam innen økonomi og klima. For eksempel ble Dora Simons forskning nylig nevnt i The Economist, Anders Åkermans forskning ble denne uken nevnt av Det hvite hus i den amerikanske presidentens årlige økonomiske rapport, og Torfinn Harding skriver jevnlig artikler i Dagens Næringsliv og opptrer på norsk TV. For at denne gruppen skal overleve, er det svært viktig å kunne arrangere seminarer om klimaendringer på høyeste internasjonale nivå.

Seminarer med internasjonale forskere om økonomi og klima ved Universitetet i Stavanger

Improving tropical forest conservation in Africa

The importance of non-timber forest products (NTFPs) for better balancing tropical forest conservation and poverty alleviation goals is increasingly recognised. Yet, in Africa, poor understanding of the impact of harvesting on species’ populations and forest carbon stocks, together with little information on the traditional institutions which have informally regulated NTFPs extraction in the past -and could continue to do so in the future-, hamper the use of NTFPs in both forest conservation and development initiatives. 
AFRI-FOR will gather the knowledge needed to determine (1) the key enablers of sustainable harvesting for groups of plant or wild meat species, (2) which types of NTFP harvesting can have synergies with carbon storage, and (3) how traditional institutions can be integrated into formal conservation. To achieve these objectives, AFRI-FOR provide new methods, including (i) a new interdisciplinary approach for investigating NTFPs harvesting impacts, combining traditional ecological knowledge (TEK, from local communities), surveys of plants and wildlife, market surveys and modelling techniques; and (ii) a novel standardised protocol for gathering information about traditional institutions and compliance towards them. AFRI-FOR will gather data and knowledge from 20 socio-ecological forest contexts in five countries (Sierra Leone, Cameroon, DR Congo, Uganda and Kenya), which will allow insights to transcend single sites and provide a much-needed general understanding. This high-risk and high-gain project will provide a step-change in our ability to inform forest conservation in Africa, by identifying synergies between biodiversity conservation, carbon storage and poverty alleviation goals.


Evaluation of thermohydraulic characteristic of printed circuit heat exchangers in pseudocritical region for supercritical CO2 cycle

Currently, 28.2% of the total EU-28 greenhouse gas emissions come from the power sector, a large contributor to greenhouse gas emissions. Consequently, the emphasis of the research in power generation has swung towards assessing highly efficient and greener power cycles. In this reference, the novel supercritical carbon dioxide Brayton cycle (sCO2-BC) is an ideal choice that outstrips other formally well-known power cycles (Brayton & Rankine cycles). In sCO2-BC, the role of the pre-cooler is critical. It serves as a sink to the power cycle and regulates the conditions at the compressor's inlet. The compressor's inlet temperature is intended to be maintained close to the critical temperature of carbon dioxide (CO2) to achieve greater cycle efficiencies. However, exceptionally higher values of the specific heat capacity of CO2 near its critical point (up to 40 times higher than water) require exceedingly high water flow rates on the cold side to achieve the desired exit temperatures of CO2. Consequently, the pre-cooler's pumping power requirements become high enough to deteriorate the cycle's performance. This problem can only be mitigated by exploring new channel geometries with enhanced thermohydraulic characteristics. Therefore, the proposed study plan to characterize the complex thermohydraulic characteristics in the pseudocritical region of CO2 using a multifaceted technique that includes, experimental, numerical, and machine learning techniques. The proposed work will provide a step forward to the success of sCO2-BC technologies that, in turn, will facilitate its integration with the green energy resources (generation-IV nuclear reactors and solar concentrated plants), helping to meet the EU's 2030 climate and energy framework goals of achieving at least 32% share for renewable energy.

Potetskrell, et biprodukt fra industriproduksjon av potet (pommes frites, flak, chips mm.) er en råstoff ressurs som kan bli bedre utnyttet til nye formål. Skrellet er nemlig en rik kilde på antioksidanter, stoffer som planter naturlig har i seg for å beskytte mot skadelige UV-stråler og reaktive forbindelser som oksygen radikaler. Antioksidanter behøver vi også å tilsette i matvarer for å beskytte og ta vare på umettet fett så det ikke blir harskt og vondt i kontakt med luft. I dag brukes flere ulike syntetiske stoffer i matvarer for å hindre oksidasjon av fett og oljer. Men det er reist bekymring rundt sikkerheten for bruken av dem. Matvareindustrien, myndigheter og forbruker ønsker derfor å fjerne flere syntetiske antioksidanter og erstatte dem med naturlig alternativer. Det er imidlertid svært få slike produkter som er godkjent og tilgjengelig på markedet. Prosjektet Peel Preserve, som har et klart bærekraft mål, skal vise om et uttrekk av antioksidanter laget på industrielt potetskrell råstoff kan fungere som konserveringsmiddel og erstatte noen av de stoffene som brukes i dag. Tre næringsmiddelbedrifter skal teste potetskrell-ekstraktet i noen av sine produkter og utføre både interne og eksterne analyser på effekten. Men en viktig forutsetning for at potetantioksidant kan tas i bruk som nytt tilsetningsstoff i mat er at det blir godkjent av den Europeiske matsikkerhets myndigheten (EFSA) og gitt et E-nummer. Prosjektet skal igangsette denne lengre flertrinns godkjennings prosessen. Parallelt skal det utvikles en forretningsmodell og nødvendig beskyttelse rundt teknologien. Dersom industrien etter testingen av potetantioksidanten, vurderer denne som er god erstatning til konkurransedyktig pris, og en EFSA-godkjenning kan oppnås på sikt, åpner det for å kommersialisere et naturlig plante-basert produkt til beste for forbruker og for miljøet. Bak prosjektet Peel Preserve står Høgskolen i Innlandet i samarbeid med Nofima og Klosser Innovasjon.

Preserving foods with potato peels: Industry-relevant validation and first regulatory assessment of a new natural antioxidant

Sjøisen i Arktis har minket drastisk de siste tiårene, og Polhavet forventes å være isfritt om sommeren mot slutten av århundret. Denne nedgangen har utsatt store havområder for vind, noe som har resultert i at det oftere dannes store bølger i Polhavet. Disse bølgene kan forplante seg over flere titalls til hundrevis av kilometer i sjøisen, knekke den og akselerere smeltingen. Disse fysiske prosessene – bølger som knekker sjøis og effekten dette har på sjøissmelting og vekst – mangler i modellene vi bruker for å forutsi klimautviklingen. Dette bidrar til store usikkerheter i klimamodeller. Effekten av bølger på sjøisen er heller ikke tatt med i prognoser for havets og sjøisens forhold, som brukes av skip som opererer i islagte farvann. I ForWArd ønsker vi å vurdere bølgenes innvirkning på sjøisens utvikling i det stadig røffere Polhavet. I dette arbeidet brukes en svært avansert modell som inkluderer sjøis, hav, bølger og deres interaksjoner. Vi skal først vurdere hvor godt den representerer forholdene i Polhavet sammenlignet med de få tilgjengelige observasjonene. Vi vil så bruke modellen til å undersøke hvordan bølgehøyden og arealet av ødelagt sjøis har utviklet seg de siste tre tiårene. Deretter vil vi kombinere modellen vår med forholdene i det fremtidige Polhavet gitt av klimafremskrivninger, for å undersøke hvordan betydningen av interaksjoner mellom bølger og is vil utvikle seg ettersom sjøis avtar i det 21. århundre. Vår modells estimering av "bølgepåvirkningen" vil tillate polarvitenskapsmiljøet å vurdere hvor usikre deres klimaspådommer uten bølger er. Til syvende og sist vil våre modellresultater og utviklinger legge grunnlaget for å legge til bølgeeffekter i hav- og sjøisvarsler i Arktis. Dette vil gjøre varslene mer nøyaktige og bidra til sikkerheten for menneskelig aktivitet i denne regionen.

Arctic sea ice decline under climate change has allowed waves to form and grow more easily in the Arctic Ocean. These waves propagate into the sea ice over tens to hundreds of kilometres, fragmenting it into small floes. This area of broken ice, known as the Marginal Ice Zone (MIZ), is a highly complex region where many interactions occur between the ocean, sea ice, and atmosphere. The action of waves strongly affects these interactions, with rapid feedbacks on the sea ice. However, climate models do not account for these wave-ice interaction processes, which likely contributes to the strong biases in their estimation of modelled sea ice extent and volume and their inability to capture short-term sea ice extent variability. Given the lack of observations available, numerical models remain the best way to estimate how wave-ice interaction processes can impact Arctic sea ice, but most wave-ice coupled models are still prototypes. In ForWArd, we will use one of the most advanced of these models to assess this impact in the context of climate change. To this aim, we will use the latest observations to calibrate the modelled wave-affected area and produce a 30-year-long hindcast of the MIZ extent evolution to analyse how it responded to larger wave height, delayed refreezing, and thinner ice. Then, we will assess the relative importance of the wave-ice interactions suspected to affect sea ice in the MIZ using well-documented events. Finally, we will estimate how waves will contribute to sea ice extent and volume variability in the coming decades, as large areas of open water will remain open until late in the autumn. The flexibility of our setup will allow us to test the sensitivity of our results to model resolution and assumptions, giving invaluable insights into the biases induced by misrepresenting wave-ice interactions. ForWArd will also pave the way for the integration of wave effects in sea ice forecasts, a requirement for safer operations in ice-infested waters.

Forecasting Wave impact as Arctic sea ice declines

RETRACE-prosjektet, som strekker seg over tre år, tar for seg den økende trusselen fra naturkatastrofer, som har rammet over 2,3 milliarder mennesker siden 2000-tallet. RETRACE fokuserer på lokalsamfunn i Arktis og Stillehavsområdet, som marquisene i Fransk Polynesia, samene og kvenene i Norge og ahtnaene i Alaska, og har som mål å styrke motstandskraften mot klimarisiko. Disse lokalsamfunnene, som er dypt sammenvevd med omgivelsene sine, gir uvurderlig innsikt i hvordan man overlever og trives i en krevende miljøsituasjon.

RETRACE tar utgangspunkt i hypotesen om at urfolks- og lokalkunnskapssystemer, særlig fra lokalsamfunn i polar- og stillehavsområdene, er uunnværlige for å utvikle effektive strategier for motstandskraft. Prosjektets metode innebærer å samle inn et rikt datasett som omfatter vitnesbyrd, kulturelle praksiser og levde erfaringer, i tillegg til kvantitative analyser. Målet er å skape et omfattende rammeverk som tar hensyn til lokale kontekster og fremmer tilpasningsdyktige og inkluderende resilienspraksiser.

Et internasjonalt forskerteam fra Frankrike, Norge og USA samler en rekke ulike ferdigheter for å bygge bro mellom vitenskapelig forskning og virkeligheten i lokalsamfunnene. Målet er å utvikle et romlig beslutningsstøttesystem som både bidrar til å visualisere resiliensdata og fungerer som et verktøy for samfunnsbasert beslutningstaking, noe som forbedrer evnen til å håndtere risiko og utnytte tradisjonell kunnskap på en effektiv måte.

Ved å kombinere lokal innsikt med vitenskapelig innovasjon skal RETRACE bane vei for resiliensstrategier som både er bærekraftige og tar hensyn til de unike behovene til sårbare lokalsamfunn. Dette prosjektet setter ikke bare søkelyset på urfolks- og lokalkunnskapens avgjørende rolle når det gjelder å håndtere klimaendringene, men har også som mål å sette lokalsamfunn over hele verden i stand til å håndtere de utfordringene som et klima i endring fører med seg.

The three-year RETRACE project responds to the increasing impact of natural disasters on over 2.3 billion people since the beginning of the millennium. It has a particular focus on Arctic and Pacific communities vulnerable to climate risks, specifically, the Marquesan community in French Polynesia, the Sámi and Kven communities in Norway, and the Ahtna people in Alaska. The project aims to address the resilience(s) of communities, in order to develop more inclusive and equitable approaches to developing and implementing resilience strategies.

Five hypotheses guide the research, based on the assumption that indigenous communities such as the ones living in Polar zones and Pacific islands, developed resilience knowledge and strategies to cope with climate-related events. The project aims to co-develop a spatial decision-support system for producing and visualizing qualitative and quantitative data on the different forms of local, community, intrinsic and traditional resiliences in those communities.

Utilizing a narrative approach, the consortium collects testimonials, cultures, memories, experiences, sensitivities and expertise from both Arctic and Pacific communities on forms of resilience and their visions of climate risks, alongside quantitative open-source data from traditional assessments. This data will be cross-referenced to build an interdisciplinary framework for a nuanced understanding of local needs, suitable to assess resilience in local contexts. 

Comprising researchers from France, Norway and the USA with diverse expertise, the consortium aims to contribute in developing risk management and resilience strategies by integrating local and indigenous knowledge into an adaptable and sustainable spatial decision-making tool. The tool aims to bridge the gap between international scientific research and local community realities, potentially informing strategies in other vulnerable territories in the future.

ResilienceS to climate risks: lessons from Arctic and Pacific communities

SASCHA har som mål å utvikle praktiske verktøy og metoder for å vurdere og håndtere kulturminner når klimaendringer utgjør en risikofaktor. Prosjektet ledes av University College London (UCL), og har Norsk institutt for kulturminneforskning (NIKU), Arizona State University (USA), NHAZCA (Italia) og North Carolina State University (USA) med som partnere. ICCROM (International Centre for the Study of the Preservation and Restoration of Cultural Property), English Heritage Trust (Storbritannia) og Ventotene kommune (Italia) deltar i prosjektet som samarbeidspartnere. 

 Hovedmål for prosjektet:  
- Utvikling av indikatorer og målemetoder for måling av klimaendringers virkning på kulturarv. 
- Etablere metoder for å involvere relevante aktører i identifiseringen og målingen av disse virkningene. Metodene skal kunne tilpasses lokale forhold og kulturell kontekst. 
- Utvikle verktøy for å hjelpe beslutningstakere til å vurdere og anvende målinger på ulike nivåer. 
- Utforske utfordringer og muligheter knyttet til bruk av nye metoder for vurdering klimarelaterte risikofaktorer for kulturminner. 

Ved å oppnå disse målene skal SASCHA ruste lokalsamfunn og kulturminneforvaltningen til å bedre forstå og håndtere klimarisiko. I tillegg, gjennom samarbeid med organisasjoner som UNESCO og ICCROM, skal SASCHA bidra til globale diskusjoner om hvordan klimaproblematikken kan integreres i kulturminneforvaltningens praksis. Alt i alt vil SASCHA legge til rette for politikkutvikling og styrke kulturminneforvaltningen ved å tilby verktøy og metoder for å effektivt vurdere, tolke og formidle klimarisiko for kulturarvsteder over hele verden.

SASCHA proposes operationalisation for transferable risk assessment and a value-based decision support tool. Supported by this approach, the overarching aim of SASCHA is therefore to develop innovative methods for understanding risks of climate change for heritage that allow for multiscale analysis and scalable approaches, bridging the gaps between local knowledge and global data, and from local vulnerability to global risks. To this end, SASCHA will: a) develop a set of operationalised indicators/metrics for relevant determinants of climate change; b) develop a locally adaptable protocol for stakeholders' engagement in the identification of relevant determinants of climate impacts that acknowledge and celebrate diversity in cultural contexts; c) develop a climate adaptation decision support tool to evaluate relevance of assessment approaches for different spatial scales; d) explore the challenges and opportunities of an operationalisation approach to climate change--heritage risk. Addressing a complex, wide and research-intensive subject, SASCHA unfolds several organisational, scientific, technical, technological and cultural heritage-oriented elements. SASCHA includes four research partners, together bringing significant experience in climate change and heritage and requisite expertise in physical sciences (UCL), social sciences and humanities (NIKU, NCSU/ASU) and technology (NHAZCA). The consortium can significantly contribute to safeguarding and protecting their region's coastal heritage but also to the global discussion of the integration of climate impacts assessment on cultural heritage through the close collaboration with UNESCO's Advisory body, ICCROM and case study stakeholders. SASCHA will develop methods for assessing, interpreting, and communicating climate change risks for heritage, while also supporting effective policy development and heritage management.

Multiscale approaches and scalability within climate change-heritage risk assessments

AGREE tar for seg klimaendringers innvirkning på kulturarv gjennom tverrfaglige metoder og samarbeid mellom ulike samfunnsaktører. Prosjektet tar særlig for seg urbane områder som påvirkes av flom. Målet er å forstå det komplekse forholdet mellom forvaltning av kulturarv, klimatilpasning og samfunnets sårbarhet, og å se på tiltak for å minimere risiko for tap og ødeleggelse som følge av klimaendringer. Sentrale delmål inkluderer integrering av strategier for bevaring av kulturarv med strategier for klimatilpasning, spesielt gjennom konseptet Historisk Urban Landskap (HUL). 

AGREE vil utvikle en beslutningsmodell forankret i HUL-konseptet som sammenligner dagens forvaltningspraksis med historiske data for å belyse sårbarhet og klimatilpasning i en urban kontekst. AGREE vil fremme transformative strategier for klimatilpasning ved å evaluere beslutningstakernes forståelse av disse samspillene. 

Prosjektet ledes av University of Leeds (Storbritannia), og har Norsk institutt for kulturminneforskning (NIKU), Politecnico di Torino (Italia) med som partnere. ICCROM (International Centre for the Study of the Preservation and Restoration of Cultural Property), British Council (Storbritannia), UK’s Department for Culture, Media and Sport (Storbritannia), Hull City Council (Storbritannia) og Innlandet fylke (Norge) deltar som samarbeidspartnere. Gjennom dette samarbeidet skal prosjektet bidra til å forme politikk og klimastrategier, både lokalt og globalt.  AGREE vil også engasjere publikum for å øke bevisstheten om klimatilpasning og mobilisere til felles handling.  Gjennom bred involvering skal AGREE bidra til å sikre at kulturarven forblir beskyttet og motstandsdyktig i møte med klimaendringer.

Alt i alt vil AGREE utforske klimaendringers påvirkning på kulturarv med en praksisorientert tilnærming, og med fokus på samarbeid, databasert beslutningstaking og samfunnsengasjement for å sikre vår felles kulturarv for fremtidige generasjoner.

AGREE champions transformative sustainability in response, emphasizing interdisciplinary methodologies and societal shifts. It explores the intricate link between cultural heritage governance, climate adaptation, and community resilience, rooted in responses to flooding in urban contexts. AGREE promotes the Historic Urban Landscape concept for integrative decision-making in climate adaptation, considering community resilience amidst environmental changes. An interactive Geographic Information System platform will illuminate this interplay over time. An innovative AGREE's contribution is a decision-making model grounded in the HUL paradigm. This model juxtaposes current national and local policies enabling cultural heritage integration in climate adaptation with historical data sources revealing urban resilience lessons and changes in the built environment over time. AGREE employs transformative governance concepts to evaluate decision-makers comprehension of these synergies and their perspectives. It advances transformative climate adaptation by uncovering potentials and barriers within heritage governance in case studies from the UK, Norway, and Italy. Beyond research, AGREE will shape policies with multi-scalar and cross-sectorial governance, interpreting climate intricacies through cultural heritage. It will strengthen global, national, and local heritage-focused climate strategies through partnerships with the British Council, ICCROM, and the UK's Department for Culture, Media, and Sport and local stakeholders, such as Hull City Council (UK) and Innlandet Region (Norway). AGREE also engages the public, raising climate adaptation awareness and mobilizing collective action, benefiting governmental climate efforts.

Graphene-based revolutions in ICT and beyond

The proposed  Framework Partnership Agreement aims to take graphene and related layered materials from a state of raw potential to a point where they can revolutionize multiple industries – from flexible, wearable and transparent electronics, to high performance computing and spintronics. A large European consortium of world-leading academics and key industries, is cooperating  closely to create a new disruptive technology. The scope and level of ambition of this endeavor is such that it can only be realized through a large scale coordinated effort that integrates the entire value chain from materials to components and systems. The rewards of the Graphene Flagship will impact the lives of Europeans with increased standard of living through new products, job opportunities, and economic growth.  In the ten-year time frame, disruptive technologies based on graphene and other two-dimensional materials extend to new areas such as biomedical applications, with the aim of becoming new platform materials, as ubiquitous as steel or plastics. This progression from scientific excellence to industrial exploitation and societal impacts is the guiding principle of the Graphene Flagship.

Frigjøring av sporer og fragmenter fra 2 og 8 uker gamle soppkulturer av Aspergillus fumigatus (AF), Aspergillus versicolor (AV) og Penicillium chrysogenum (PC) ble studert med to generatorer, tre medier og to lufthastigheter. Det frigjøres sub-? fragmenter fra soppkulturene i tillegg til sporer, sporeaggregater og andre større partikler. Flest sub-um fragmentene stammer fra mycelium (trådformet nettverk av soppceller). I forsøkene med Aspergillus artene ble det også funnet sporefragmenter. Det ble funnet fragmenter i samme størrelse og form som sporer. Det er sannsynlig at tidligere laboratoriestudier har overvurdert frigjøring av sporer. Observasjon av fragmenterte sporer tyder også på at tidligere publiserte funn kan være basert på artefakter. To artikler er publisert i Appl Environ Microbiol og Aerosol Sci&Technol. 
Deteksjon av fragmenter fra mycelium samt sub-um partikler i miljøprøver krever spesifikk merking fordi disse partiklene kan mangle karakteristisk morfologi. En metode ble utviklet basert på immuno-gull merking med IgY antistoffer mot AV sporer og deteksjon med field emission scanning elektronmikroskopi i under utenlandsoppholdet i USA. Metoden er testet ut på miljøprøver tatt under renovering av fuktskader og beskrevet i en artikkel i Appl Environ Microbiol. Metoden er en viktig forutsetning for å kunne studere forekomst av mycelium fragmenter som forventes å gi allergiske effekter i luftveiene. 
Kandidaten forsvarte sin avhandling ved NMBU 22.5.2015 For de toksikologiske studiene er mengden av sporer fra AF og PC som kan genereres er tilstrekkelig, men ikke for AV og Stachybotrys chartarum (SC). Sporer fra AV og SC ble fremstilt ved vasking av kulturer. Også fragmenter ble generert i for små mengder og ble fremstilt ved å høste mycelium før sporedannelse, og tørke og knuse det. Finfraksjonen ble deretter samlet opp med en respirabel syklon. 
Hyfefragmenter og sporer ble undersøkt i hus med synlig soppvekst. I inneluft var 53% av sopp partiklene hyfefragmenter, mens det var 21% i uteluft. Kun 3% var småfragmenter (manuskript).

Sporer og hyfefragmenter fra AF, PC, AV og SC kulturer ble samlet inn, inaktivert og karakterisert mhp antall og partikkelform, og mykotoksin, b-glukan og proteaseinnhold/aktivitet. Proinflammatoriske egenskaper ble undersøkt i humane THP-1 monocytter, THP-1 makrofager og bronkiale epitelceller (BEAS-2B). Preparatene viste seg å være rene spore- eller hyfepreparater, bortsett fra sporer fra PC og SC som hadde en viss andel hyfer. Hver muggsoppart produserte hovedsakelig én type protease. Mykotoksin-nivåene var lave. ?-glukaner og proteaser ble hovedsakelig funnet i hyfene. THP-1 makrofager var mest sensitive og responderte med cytokinutskillelse på meget lave doser av AF hyfe (10 ng/cm2). Det var store potens forskjeller mellom ulike sopparter og vekststadier mhp proinflammatoriske responser, men forskjellene kunne ikke enkelt relateres til en enkelt av karakteristikaene over. AF and PC hyfer var mye mer potente en korresponderende sporer, noe som er i tråd med nåværende konsensus. Interessant nok gjaldt imidlertid ikke dette sporer og hyfer fra AV og SC. En artikkel publisert i Indoor Air 2017.
En sammenligning av AF sporer og hyfer som var inaktiverte ved røntgen eller varme, versus ubehandlede viste at ingen av inaktiveringsmetodene ødela cytokin responsene. Studier av mekanismer for hvordan sporer og hyfer fra AF og AV induserer cytokinresponser i ulike makrofager THP-1, primære blod monocytt-deriverte (MDM), og lungemakrofager, er ferdigstilt. AF var klart den mest potente; og de ulike makrofagmodellene responderte kvantitativt og kvalitativt forskjellig. Responsene i MDM synes mest relevante. I MDM synes responsene fra sporer og hyfer fra AF å involvere for det meste toll-like reseptor (TLR)4 og i mindre grad TLR2 De ulike cytokinresponsene gikk alt vesentlig via samme reseptor. Manuskriptet blir innsendt i 2018.
I en studie basert på sporer og hyfer fra AF og PC foretas en karakterisering av immunresponsene i lungeepitelceller. Ubehandlede AF sporer ble omdannet til hyfer raskere i nærvær med epitelceller enn uten, og førte til responser som lignet på de fra inaktiverte hyfer. Interessant nok ser responsen fra både AF og PC hyfer å gå via TLR2, og ikke via TLR4 som i makrofager. Manuskriptet blir innsendt i 2018.
Kombinert eksponering av mineralpartikler og organiske komponenter (AF hyfe) førte til en synergistisk utskillelse av IL-1?. AF hyfer førte til økt syntese og produksjon pro-IL-1?, og til en viss grad også utskillelse av IL-1?. Mineralpartikler førte til en synergistisk effekt ved å kløyve proformen indusert av AF hyfer, noe som førte til en ytterligere utskillelse av IL-1?. Denne effekten ble karakterisert i THP-1 makrofager, og ble bekreftet i de andre makrofagmodellene og i lunge epitelceller. Manuskriptet er innsendt.
PhD-sammendraget basert på 4 artikler er påbegynt og blir ferdigstilt i februar 2019.

Fungal spores have obtained widespread attention as possible causal agents for adverse indoor air associated health problems. It has been reported that fungi may adversely affect human health through allergic and non-allergic airway inflammation, by infec tion and by the toxicity of mycotoxins. However, it is still not clear if spores inhaled in indoor environments really have all the adverse health implications suggested, as the indoor levels of airborne spores are often found to be too low to explain ass ociations with health effects other than allergy.
Recent studies have drawn attention to fungal hyphae and smaller-sized fungal fragments as tentative causes of indoor health problems.  Furthermore, experimental studies have suggested that the inflammator y properties of fungal spores are modulated by their viability showing allergic responses to viable spores and non-allergic responses to non-viable spores. However, since no method for measurement of fungal fragments exists, the airborne levels indoor and  outdoor, and their toxic properties are largely unknown. Thus, possible health implications of fungal particles besides spores are still very unclear.
The goals for this project are therefore to i) develop measurement methods for viable and non-viable sp ores, hyphae and fragments, ii) assess their airborne levels in a small proof-of-principle study in common indoor environments and in moisture-damaged buildings, iii) characterize these fungal particles by chemical and biological methods, and iv) study th eir toxic properties in vitro and in vivo. For this purpose, viable and non-viable spores, hyphae and fragments will be prepared from pure cultures of fungal species associated with moisture-damage in buildings. 
The results are expected to provide new me thods for exposure assessment of fungal particles to be used in future epidemiological studies, and new insights in the possible role that fungal growth in indoor environments may have on respiratory health.

Fungal particles in indoor air: occurrence and toxic properties of viable and non-viable spores, hyphae and fragments

Phthalates and bisphenol A are commercially important classes of industrial chemicals used in a wide variety of consumer products. Many of these compounds have been identified as endocrine disruptors causing functional and structural impairment of reprodu ction and development. Furthermore, epidemiological associations have been observed between exposure to these chemicals and disease outcomes like allergy/asthma and diabetes. However, the routes whereby people become exposed to phthalates and BPA are not  yet fully understood.

The overall aim of the project is to assess the pathways for human exposure to phthalates and BPA by comparing estimates of exposure from indoor environment and food with excretion of urinary metabolites as integrated biomarkers of  exposure. Results from this study will give valuable inputs to the risk assessment of these compound classes and to the development of control strategies to minimise human exposure.

A cohort of about 40 households with children at age 6 to 10 years will  be established. Indoor air and dust samples and urine samples will be collected both from adults and children as well as information on dietary habits and indoor variables. Selected basic foodstuffs will be analysed for the estimation of the dietary intak e. 

The exposure of vulnerable subpopulations such as the children from the cohort mentioned above and pregnant women will be assessed and compared with average exposures in general populations. Finally, associations between exposure to phthalates and BP A and immunological response and inflammation reactions will be investigated in cell cultures and experimental animals.

This project is based on a collaboration of national and international scientists from a variety of fields. The combination of analyti cal and environmental chemists, nutritionists and toxicologists forms the needed framework for progress in the understanding of the exposure to these chemicals and the implications for humans.

Internal exposure to bisphenol A and phthalates - Sources, pathways and relationship to effects

Selv om sammenhengen mellom utendørs luftforurensning og diverse helsetilstander har vært studert i mange undersøkelser, er det fortsatt usikkerhet om mange forhold. Dette gjelder ikke minst om luftforurensing under svangerskapet kan ha betydning for mor og barns helse.  I denne undersøkelsen vil en bruke opplysninger fra ca. 20 000 deltagere i Den norske mor barn undersøkelse (MoBa) som under svangerskapet var bosatt i Oslo og Bergen og områdene rundt disse byene. Det planlegges å studere om det er noen sammenheng mellom bl.a. svangerskapsutfall, preeclampsia, og luftveisplager i tidlig levealder og matematisk beregnende luftforurensningsnivåer på deres bostedsadresser. Fremgangsmåten for luftforurensingsberegningene er basert på metoder som nylige ble benyttet i en EU-undersøkelse (ESCAPE) som omfattet 29 forskningssentre rundt omkring i Europa. Det er også planlagt å undersøke om er sammenheng mellom indikatorer for luftforurensningseksponering og grad av DNA methylering for et underutvalg. Om noe forsinket så er prosjektet i hovedsak gjennomført etter planen. eksponeringsmålinger har vært gjort og eksponeringsmodeller har blitt bygget og deltagerne har fått tilskrevet luftforurensingsestimater. Analyser av sammenhengene mellom eksponering og de beskrevne helserelaterte utfall er gjennomført og artikler er under ferdigstilling (innsendt eller i manusform), men ingen resultater er publisert offentlig per i dag.

This project is based on the Norwegian Mother and Child Cohort Study (MoBa). Pregnancy is a critical time period determining disease susceptibility not only in childhood, but also later in life. Air pollution exposure during pregnancy has been associated  with adverse birth outcomes. Early exposure to outdoor air pollution even in pregnancy may also have an effect upon the development of respiratory diseases, such as asthma related phenotypes. The aim of this project is to build up new knowledge on the pat hogenesis of respiratory diseases by focusing on fetal exposure to air pollution during pregnancy. There is a great need to develop a new paradigm of disease pathogenesis that takes advantages of applied molecular approaches to asthma as it occurs in huma ns at different stages of development. The MoBa cohort has a large sample size and will be able to address more sensitive groups due to the use of genetic factors that influence individual susceptibility to air pollution. In addition, we are participating  in the EU project European Study of Cohorts for Air Pollution Effects (ESCAPE) and this enables us to have better spatial and temporal understanding of the air pollution exposure, thus being able to address more trimester-specific exposure for each pregn ancy. The international collaboration will enhance Norway's position in playing a key role in epigenetics and research into respiratory diseases by building national competence in these fields. This study will advance knowledge of the mechanisms whereby o utdoor air pollution exposure during pregnancy influences gene expression and risk of atopic diseases. Further, we will examine whether methylation of these genes is associated with the development of asthma-related phenotypes, and this could provide us w ith great insights in how environmental factors in pregnancy and early childhood can influence methylation and DNA regulation. Results from this study will form the basis of the project and future studies.

Air pollution, birth outcomes and early respiratory health - taking gene environmental interactions into account

I de siste tiårene har det kommet stadig nye beviser for at miljøkjemikalier som ftalater og bisfenol A (BPA) kan føre til nevronal utviklingsskade. Man er derfor bekymret for effektene disse stoffene kan ha på nervesystemet hos fostre, spedbarn og barn. I tråd med dette har EU-kommisjonen og deres Scientific Committee on Emerging and Newly Identified Health Risks (SCENIHR) i februar i år (2015) kommet med en uttalelse vedrørende sikkerheten knyttet til bruk av bisfenol A i medisinsk utstyr, der de konkluderer med at det er en viss risiko for alvorlige effekter når BPA er direkte tilgjengelig for systemisk eksponering [?], særlig for nyfødte i intensivavdelinger og spedbarn som gjennomgår langvarig medisinsk behandling.
 Videre la EFSA, EUs organ for matsikkerhet, i januar 2014 frem en foreløpig rapport fra sin nyeste risikovurdering av BPA. Her anbefaler de at det nåværende nivået for tolerabelt daglig inntak (TDI) settes ned fra 50 µg/kg kroppsvekt/dag til 5 µg/kg kroppsvekt/dag. Dette er hovedsakelig basert på skadelige effekter i lever og nyre, men det understrekes i rapporten at det fortsatt er stor usikkerhet knyttet til andre helsefarer, bl.a. nevrotoksisitet. Som følge av dette er det et akutt behov for bedre dyremodeller til å studere slike effekter.
 I dette prosjektet har vi jobbet med utviklingen av en kyllingmodell der vi eksponerer kyllingembryo in ovo (i egget) for å mimikere eksponering in utero (i mors liv) og i den tidligste fasen etter fødsel. I tillegg til denne kyllingmodellen har vi benyttet museavkom fra musemødre som har fått BPA i drikkevannet. Musemødrene fikk BPA i vannet både før parring, under graviditet og mens de diet. Målet med begge disse modellene er å studere effektene av ftalat- og bisfenol A-eksponering på hjernen under utvikling.
Sentrale prosesser under nervesystemutviklingen er cellevekst (celledeling) og en regulert celledød som kalles apoptose, samt at cellene modnes og forflytter seg til de riktige områdene av hjernen og nervesystemet for øvrig.  I lillehjernen er nivået av et bestemt protein som heter Pax6 avgjørende for korrekt utvikling av en type nerveceller som kalles kornceller. Vår forskningsgruppe har publisert et arbeid der vi viser at vi kan gjenskape den reduksjonen i Pax6-nivåer som normalt skjer i lillehjernen under utvikling i kyllingkornceller når vi dyrker dem i laboratoriet. Videre viser vi at eksponering av kyllingembryoene for BPA mens de er i egget gir endringer i disse korncellene som manifesterte seg over tid i form av endringer i Pax6-uttrykket. 
Vi hevder derfor at kyllingmodellen er en godt egnet modell for å oppdage kjemikalieinduserte forstyrrelser i lillehjernens utvikling, og har benyttet denne modellen også i videre studier.
Vi har sett at også ftalatene di-isononylftalat (DINP) og dibutylftalat (DBP) forstyrrer Pax6-uttrykket i lillehjernen hos kyllingembryo. Disse funnene blir nå skrevet sammen for publisering. Dataene har også blitt presentert av K. Rakkestad på to ulike internasjonale konferanser; Neuroscience 2013 i San Diego, USA og Eurotox 2014 i Edinburgh, Skottland.
Utviklingen av kyllingmodellen har også vært viktig for forskningsgruppens arbeid innen sikkerhetsfarmakologi, og flere arbeider som omhandler mulige skadelig effekter på utviklingen av nervesystemet hos fosteret ved bruk av bl.a. glukokortikoider (en form for steroider) og epilepsimedisin under svangerskapet er nå på vei til å bli publisert. Sammen med studiene av miljøgifter har dette dessuten bidratt til en økt grunnleggende forståelse av Pax 6 sin rolle under hjernens utvikling.
I samarbeid med Folkehelseinstituttet har vi også arbeidet med nanosilikapartikler. Vi har benyttet en såkalt cellelinje (feokromocytomcellelinjen PC12) til å studere effekten av eksponering for både SiNP10 og SiNP50 på aktiviteten av ulike transkripsjonsfaktorer (proteiner) som er viktige for betennelsesreaksjoner og for differensiering av nevroner. Deler av dette arbeidet vil bli publisert i form av en masteroppgave våren 2015.
VI har også hatt et prosjekt der vi har benyttet PC12 celler til studier av BPA og ftalaters effekt på glutationproduksjon. Glutation er den viktigste av cellenes egne antioksidanter. Disse studiene er en oppfølging av et arbeid av Rakkestad og Sørvik som ble publisert i 2014. Her viste vi at 17?-østradiol, som vanligvis anses som en beskyttende effekt på nerveceller, nedregulerer glutationsyntesen i celler som er stresset ved hjelp av  endrede vekstbetingelser (serumdeprivasjon). Våre foreløpige funn tyder på at BPA og DBP i lave konsentrasjoner kan ha en lignende effekt. Deler av dette arbeidet er publisert i en bacheloroppgave av en Erasmus-student fra Østerrike (Reumann D, University of Applied Sciences, FH Campus Wien) som Rakkestad var veileder for.
Totalt er det så langt publisert to vitenskapelige artikler fra dette prosjektet, og fire nye artikler er under utarbeidelse i det prosjektperioden utløper.

The present research proposal is in toxicology. It is a cooperation between the Norwegian Institute of Public Health and the School of Pharmacy, University of Oslo. It is focused on the the toxicity of the environmental chemicals/indoor air pollutants bis phenol A (BPA) and phthalates on fetal brain development. Humans are widely exposed to BPA (an additive in the production of polycarbonate plastics and other types of plastic) and phthalates (a class of chemicals used as softeners in polyvinyl chloride pl astics used in a variety of consumer products). EU has recently (april 2008) updated the risk assessment report of BPA, and 50 mg/kg/day was suggested as NOAEL-value (no observed adverse effect level). The scandinavian Pollution Control Authorities was in volved in the discussion of this revised risk assessment report, but did not approve that the NOAEL-value covered neurotoxic effects on the developing nervous system. In risk assessments published by The U.S. National Toxicology Program, Health Canada and  Environment Canada (april 2008) some concern is expressed for the effects of BPA on the nervous system and behavior of fetus, babies and children. Phthalates readily crosses the placenta and accumulates in the fetus, and it was recently reported that pht halates may affect the neurodevelopment of the human brain. The aim of this project is to evaluate the neurotoxicity of phthalates and BPA on the developing brain. Both in vivo and in vitro studies will be performed. Cell division rates, apoptosis rates a nd neuronal morphology will be addressed.

Neurodevelopmental toxicity caused by environmental exposure to phthalates and Bisphenol A

En parallell økning i overvekt/fedme og ulike krefttyper er sett de siste tre tiår. Fedme antas å øke risikoen for kreft, inkludert tarmkreft, men mekanismene er uklare. Samtidig har vi blitt utsatt for mer forurensende stoffer i miljøet (miljøkontaminanter). Vi har studert sammenhengen mellom fedme, forårsaket av en genfeil (mutasjon) eller av fettrik kost, og tarmkreft. Dette er gjort bl.a. i en ny dyremodell laget ved å krysse en veletablert modell for tarmkreft (Min-mus) med en mus som utvikler fedme fordi den mangler hormonet leptin som styrer matinntak og energiforbruk (ob-mus). Prosjektet hadde tre problemstillinger: 1) Øker fedme i voksne dyr spontan (arvet) tarmkreft og tarmkreft forårsaket av stekemutagenet PhIP, som dannes i stekt kjøtt og fisk. 2) Påvirker fedme tidlig i livet utvikling av fedme og følsomhet for miljøkontaminanter som voksen. Dette ble studert i avkom av Min-mus hunner som fikk fettrikt fôr under ulike perioder i livet. 3) Ved eksponering under fosterlivet, vil miljøkontaminantene PFOA eller PFOS, som bl.a. fins i matemballasje, øke fedme og følsomhet for andre stoffer som voksne. I 1) fant vi at mus med mutasjoner i begge kopier av leptin-genet og som totalt mangler leptin, utviklet kraftig fedme. Fedmen økte enda mer med fettrikt fôr. Musene uten leptin med fedme hadde også høyere antall svulster i tynntarmen, og antallet svulster økte med fett-rikt fôr. Blodsukkerreguleringen var forstyrret og insulin i plasma var økt i musene med fedme, og dette ble forverret med fett-rikt fôr. Det var en klar sammenheng mellom fedme forårsaket genetisk og via kosten, forstyrret blodsukkerregulering og tarmkreft. Musene med fedme hadde også økt plasma-nivå av TNFalfa, en markør for betennelse. Mus hvor bare en kopi av leptin-genet var skadet, og som produserte noe leptin, hadde litt økt kroppsvekt, men normalt blodsukker og antall tynntarmsvulster. I 2) fant vi at mus som ble utsatt for fettrikt fôr via mor tidlig i livet, dvs. som foster og i dieperioden, hadde økt kroppsvekt og flere svulster i tynntarmen som voksne. Blodsukkeret var ikke påvirket av denne tidlige eksponeringen, slik som ved eksponering som voksne. I 3) fant vi at eksponering for PFOA eller PFOS under fosterlivet ikke gav økt kroppsvekt, forstyrret blodsukker eller flere svulster i tynntarmen hos musene som voksne.

The prevalence of both obesity and colorectal cancer has increased substantially in the last decades. Simultaneously have humans been exposed to increasing amounts of environmental contaminants. Three fundamental questions related to the interplay between  environmental contaminants, genes and health outcomes, obesity and intestinal cancer, will be studied in this project. The question whether genetical or diet-induced obesity can affect food mutagen-induced or spontaneous, via mutation in the tumor suppre ssor gene adenomatous polyposis coli (Apc), intestinal tumorigenesis in adult mice, will be studied in a new double mutant Apc x ob mouse model. The obesity will be induced genetically via mutation in the ob (obese) gene coding for the hormone/cytokine le ptin, or through administration of an obesogenic diet. The next question is whether obesogenic conditions in utero and in early life can affect the risk of developing obesity as well as increase the susceptibility for environmental contaminants causing in testinal cancer in the offspring. The third question which will be studied is whether in utero exposure for environmental contaminants, i.e. endocrine disruptors such as perfluorooctanoic acid and bisphenol A, can increase the risk of developing obesity a s well as increase the susceptibility for other environmental contaminants in the offspring. Mechanisms for how obesity affects intestinal tumorigenesis and how environmental contaminants may contribute to obesity will be studied along two main hypotheses , via insulin resistance/hyperinsulinemia and/or inflammation. The information obtained in this project will increase the understanding of the mutual causal relationship between obesity and susceptibility for environmental contaminants, causing intestinal  cancer. The results obtained will also be very useful in human risk assessment of substances such as perfluorooctanoic acid, for which data on toxic effects and dose-response relationships are still lacking.

Researcher project: Interplay between environmental contaminants, genes and diet in obesity and intestinal cancer

This project is based on the Norwegian Mother and Child Cohort Study (MoBa). While limited human evidence is available on the association of childhood atopic diseases with maternal diet during pregnancy and gene methylation in the offspring, new murine st udies from our collaborators at the National Institute of Environmental Health Sciences (NIEHS) in the US have identified effects of high intake of methyl donors during gestation on gene methylation in relation to the development of an allergic asthmatic- type response in the offspring. In this proposal, which includes the first phase of the project, we will investigate if maternal intake of methyl donors in pregnancy affects risk of asthma and atopy at age 3 years. When additional funding is granted, futu re studies based on this project will examine whether mothers with high levels of biomarkers of methyl donor intake give birth to offspring with increased methylation in the genes identified in the mice studies and in additional human atopic disease candi date genes. Further, we will examine whether methylation of these genes is associated with the development of asthma phenotypes. This study will advance knowledge of the mechanisms whereby diet, and other environmental factors, influences gene expression and risk of atopic disease. Analyses of maternal methyl donor status, B-vitamin metabolism and methylation patterns will give great insights in how environmental factors in pregnancy and childhood can influence methylation and DNA regulation. The current study will address associations between childhood respiratory and atopic diseases and maternal b-vitamin status and SNPs in B-vitamin metabolism. Results from this study will form the basis of the project and the future studies.

Nutrients, genes and epigenetic modification of DNA in relation to asthma and atopy.

Ambient particulate matter (PM) contribute to the development of adverse health effects in the lung and cardiovascular systems. PM has a complex composition and may contain a variety of different organic compounds and metals. We will investigate the possi ble role of PAHs and metals in PM-induced inflammation and cell death, and whether differences in biological activity between PM from different sources might be attributed to differences in PAH and metal content. In particular, it will be examined whether  different metals and PAHs induce differential patterns of cytokine release. Selected metals and PAHs will be used to study the initial signalling mechanisms and pathways leading to cytokine synthesis and cell death in cell culture models. Furthermore, we  will study how the balance between mechanisms involved in cell death, cell survival and inflammation may modulate the outcome of PAH and metal exposure. This may elucidate whether cytokine production and cell death are triggered by similar particle prope rties but at different doses, or whether completely different particle-properties and signalling pathways may be responsible for the two outcomes. Possible cooperative effects between different metals and PAHs will be examined to better understand the tox icology of PMs. In parallel to study the components, we will examine the potential of different combustion particles (CPs) to trigger cellular responses, and in particular chemokines. We will include wood-burning particles produced at different conditions , different biodiesel CPs, diesel CPs produced in absence and presence of particle filters and extracts from selected CPs. Our intention is to identify component groups responsible for the observed effects of CP and sensitive endpoints for mechanistic stu dies. In a cooperating, parallel project, prof. Sandström, Univ. of Umeå will test the same CPs by exposure of human volunteers in inhalation chambers, which opens for comparison of in vitro and in vivo effects.

Importance of components and sources in particle-induced lung inflammation: Role of PAHs and metals

Although marine food has long been known to have health properties, its roles and impacts on insulin sensitivity and metabolic diseases (e.g. type 2 diabetes, obesity, hypertension, coronary heart diseases etc.) remain still unclear and worldwide debated.  Beside its potential health effects, recent reports highlight that marine food may contain a potentially harmful level of contaminants. Evaluating the balance between the risks and benefits of marine food consumption is very challenging and as a conseque nce, the public faces conflicting reports on the risks and benefits of marine food intake resulting in controversy and confusion over the role of marine food consumption. The present project will generate new knowledge regarding the health benefits and ri sks of marine food (salmon oil) intake.

Salmon and contaminants: impacts on insulin sensitivity and metabolic diseases

Persistent pollutants in Arctic areas have received much attention. It is well known that these contaminants accumulate in food webs and potentially cause severe physiological damages to top level predators including humans. People in Arctic regions tend to be more exposed to these pollutants due to their high intake of wild fish and animals. The project will conduct an interdisciplinary survey of environmental sensitivity and human vulnerability to mercury (Hg) in a population group in Arctic Norway. Amo ng the wide range of contaminants found in food items of that region, the presence of Hg frequently sets the consumption limits for fish. Surprisingly little knowledge exists on the role of Hg in the Norwegian Arctic ecosystems and people. Elevated concen trations of Hg in both sediments and fish, related to long-term natural accumulation and recent long range transported pollution, have been found. Studies from other Arctic countries have shown partly high concentrations of Hg in certain population groups . Most studies dealing with the impact of Hg in the environment limit their scope to certain aspects of the biogeochemical cycle, often neglecting to put in perspective their behavior at the ecosystem level, and disregarding the role that humans do play i n these cycles. This project will integrate knowledge from the natural sciences, health sciences and social sciences to assess the specific environmental sensitivity to the presence of Hg and the human vulnerability of an exposed population in the Norwegi an Arctic. The project team comprises national expertise in the relevant fields and in addition will draw on leading international expertise.

Environmental mercury and human health in Arctic Norway

Grains are commonly infected with toxigenic fungi, particularly with species of the Fusarium and Alternaria genera. Fusarium fungi are producers of a range of well known mycotoxins as well as less known bioactive metabolites for which little toxicological  information exists. Food and feed containing naturally infected cereals have often more pronounced effects in animals than if the corresponding levels of pure trichothecenes are added to the feed. This may be due to the presence of other fungal metabolit es. The aim of this project is to provide a toxicological characterization of selected cytotoxic fungal metabolites other than trichothecenes present in Norwegian grains, and to assess their contribution to the toxicity of grains contaminated with toxigen ic field fungi. Selected metabolites from Fusarium will be purified for further studies. The selected cytotoxic metabolites will be further studied in appropriate cell models(s) to provide information about toxicity and cellular targets, and the effects w ill be compared to those of trichothecenes. The information obtained will also be study the combined effect of selected cytotoxic compounds. The effects of these fungal metabolites on gene expression will be studied by microarray technology and the effect s on gene expression will be related to observations from the cellular systems. An in vivo study will be performed to sustain that the in vitro findings also apply to intact organisms. The information generated on mechanisms of action will be used to clar ify the role of different metabolites in the toxicity of Fusarium infected grain and thereby strengthen the hazard identification and hazard characterization to improve the risk assessment of mycotoxins in cereals.

Toxicological characterization of selected secondary fungal metabolites in Norwegian grain

Asthma, allergy and diabetes increased during the last decades. These diseases are caused by dysfunction of the immune system. Diabetes and allergic diseases could share common underlying immunological mechanisms and environmental causes. Our hypothesis i s that the two groups of chemicals, phthalates and bisphenol A (BPA), contribute to the development of diabetes, asthma and allergy. Exposure occurs broadly through both the indoor environment and food. Recent data have indicated an association between as thma and allergic symptoms in children and phthalates in residential dust. Also for BPA are there data indicating a link between exposure and diabetes as well as allergy. Through two experimental models we will investigate the effects of phthalates and BP A in relation to allergy, autoimmunity and inflammatory responses in lung cells. 
In vivo we will study the effects of phthalate and bisphenol A exposure in female mice with regard to development of insulin dependent (autoimmune) diabetes and allergy in o ffspring (as well as in the mother mice). Mice will be exposed perorally. Development of diabetes will be analyzed in mothers and the offspring together with specific IgE response to allergen. A number of relevant ex vivo investigations on cells and cytok ines will be conducted.  
In vitro we will examine the effects of the same substances on intracellular signal transduction pathways pertinent to airway inflammatory responses/asthma in different lung cell types from rats and humans. Also cells from the in  vivo-model will be studied. The study will examine if the substances mediate their effects through receptor binding or more unspecific toxic effects. Release of  inflammatory mediators, expression, activation and localization of components in relevant si gnal transduction pathways as well as cell death will be studied by immunologic, molecular biological  as well as morphological techniques.

Environmental exposure to phthalates and Bisphenol A and the development of diabetes and asthma - an in vivo and in vitro study

We here present evidence that environmental exposure to immunoadjuvants in particular mineral oils, may precipitate autoimmune diseases in susceptible individuals, and that a novel class of genes encoding leukocyte regulatory receptors contribute to this susceptibility. The evidence is based on the structural identification of the rat Oia2-gene, a quantitative trait locus conferring susceptibility to autoimmune arthritis elicited in susceptible rat strains by skin exposure to small doses of mineral oil. W e show that the trait is linked to a novel gene complex that we have called APLEC, encoding regulatory leukocyte receptors expressed by professional antigen presenting cells. The functional role of these receptors, their cellular expression patterns, and the finding of genetic association between a gene in homologous human gene complex and rheumatoid arthritis (RA) constitute evidence that the model is valid for the study of human autoimmune diseases. We argue that oil induced arthritis represent a search light model of a multifactorial trait where genes and environmental factors interact in 
the precipitation of disease. We present project plans for
