Effektene av fysisk og seksuell vold på ofre og deres familier kan være ødeleggende og vedvarende, med potensielt langvarige konsekvenser for fysisk og mental helse, utdanning, sysselsetting og bruk av velferdstjenester. Mens strafferettssystemet anses som en viktig del av offerets gjenopprettingsprosess, er det begrenset empirisk forskning på hvorvidt strafferettslige prosesser hjelper og/eller hindrer bedring langs disse ulike dimensjonene. I dette prosjektet vil vi adressere dette kunnskapshullet ved å bruke en kombinasjon av kvantitative og kvalitative forskningsmetoder. I de kvantitative analysene vil vi bruke administrative registerdata som dekker hele befolkningen sammen med kvasi-eksperimentelle metoder for å kunne trekke kausale slutninger. Vi vil også benytte maskinlæringsteknikker for prediksjon. De kvalitative analysene vil supplere de kvantitative analysene ved å gi kontekst til et komplekst felt av saker og ofre og ved å identifisere potensielle mekanismer mellom strafferettslige prosesser og utfall for ofrene. Samlet vil disse metodene gi en mer robust, representativ og nyansert forståelse av hvordan kontakt med strafferettssystemet påvirker offeret på tvers av flere viktige livsområder både på kort og lang sikt. Prosjektet ledes av Folkehelseinstituttet (FHI) med Frischsenteret og Universitetet i Oslo som partnere. Prosjektet har et rådgivende utvalg med medlemmer fra Nasjonalt kunnskapssenter om vold og traumatisk stress (NKVTS), ETH Zurich, Universitetet i Oslo, Frischsenteret, Universitetet i Bergen, og OsloMet. Det er også tilknyttet en brukergruppe bestående av personer og organisasjoner som er i kontakt med ofre for vold og overgrep.

Victims of sexual and physical violence is one of the most vulnerable groups in our society. The criminal justice system (CJS) serves a critical role in providing assistance to victims of these crimes, enabling them to seek justice, protection, compensation, and closure. Yet, knowledge about whether the CJS helps or harms these victims is extremely limited. This project aims to remedy this by providing a comprehensive understanding of the role of the CJS in supporting and providing restoration for victims of physical and sexual violence in Norway. We will utilize a combination of quantitative and qualitative research methods across disciplines to study outcomes across multiple important life domains, both in the short and long run. For the quantitative analyses we will use exceptionally rich longitudinal administrative register data with full population coverage combined with state-of-the-art quasi-experimental methods for credible causal inference and predictive machine learning techniques. The qualitative analysis will be based on narrative interviews with victims, providing context and insights into how interactions with the CJS shape life stories. The results will yield a robust and nuanced understanding of how the different stages of the CJS impacts victims. This is particularly urgent given how the current emphasis on legal responses to assaults has caused a sense of obligation and civic duty for especially women to report incidents, while the chances of legal closure remain small. In addition to advancing the methods for quasi-experimental designs and machine learning within quantitative criminology, this project will provide valuable insights for policymakers, stakeholders, and practitioners working in the field of victim support and rehabilitation and may inform current legal reform efforts aimed at addressing these issues.

Helpful or harmful: The effects of criminal justice system contact on victims of sexual and physical violence

Hovedmålet med kreftbehandling er å eliminere kreftceller og dette er tradisjonelt oppnådd ved kirurgi, bruk av giftige medikamenter (cellegift) og stråling. En ny og betydningsfull tilnærming til kreftbehandling kalt immunterapi, har derimot fått mye oppmerksomhet det siste tiåret; Her forsøker leger og forskere å utnytte immunsystemet vårt for å målrettet  ødelegge kreftceller. En nøkkelaktør i denne banebrytende tilnærmingen er en gruppe «drapsmenn» i immunsystemet kjent som cytotoxiske T-lymfocytter (CTL-er). Disse cellene har en bemerkelsesverdig evne til å oppdage kreftceller ved å gjenkjenne unormale molekyler på overflaten av kreftcellene - molekyler som friske celler ikke har. Når CTL-er gjenkjenner kreftcellene ødelegger de dem ved å frigjøre giftige molekyler som initierer kreftcellenes selvdestruksjon. 
 
Selv om betydelige innsats har blitt dedikert til å forstå hvordan CTL-er aktiveres og hvordan de gjenkjenner kreftceller, vet vi mindre om de påfølgende hendelsene inne i kreftcellene som fører til deres død. Fra før vet vi at CTL-er kan initiere to forskjellige typer celledød i kreft: den første, kalt apoptose, er en stille og ordnet prosess der kreftcellene krymper og brytes opp uten å varsle immunsystemet; den andre, kalt pyroptose, er en mer dramatisk prosess som involverer dannelse av åpninger i cellemembranen, noe som fører til at cellene svulmer opp og sprekker. Under denne cellebristen blir innholdet inne i cellene frigitt i omgivelsene, og dette kan fungere som 'faretegn' som kan stimulere en sterkere immunrespons mot kreftcellene. Betydningen av denne cellebristen for å forsterke anti-tumorresponsene vil bli undersøkt i dette prosjektet. 
 
Videre er vi fascinert av muligheten for å avdekke ytterligere, ennå ukjente, former for celledød indusert av CTL-er. Utforsking av disse dødsrelaterte prosessene i kreftcellene kan potensielt øke effektiviteten av immunterapi og bidra betydelig i den pågående kampen mot kreft.

The primary goal of most cancer therapies is to induce cancer cell death. Immunotherapy relies on the cytotoxic activity of lymphocytes such as natural killer cells (NKs) and cytotoxic T lymphocytes (CTLs) to achieve this. While a significant amount of research in immunotherapy focuses on the events within cytotoxic lymphocytes or at the interface between cytotoxic lymphocytes and cancer cells, less is known about the molecular mechanisms that orchestrate cancer cell death, particularly in the case of CTL-mediated death.
CTLs are known to induce two types of cell death in cancer: non-inflammatory apoptosis and proinflammatory pyroptosis. Pyroptotic cancer cells release proinflammatory signals that attract immune cells to the tumor site, thereby enhancing the anti-tumor response. Cell lysis is a common outcome of pyroptotic cell death. It is unclear whether lysis can be decoupled from pyroptosis in cancer cells and whether the proinflammatory properties of pyroptotic cancer cell death depend on lysis. Moreover, it remains unknown whether CTLs can induce other forms of cell death in cancer, apart from apoptosis and pyroptosis.
Studying the mechanisms of CTL-mediated cancer cell death is challenging because CTLs selectively target cells expressing specific antigens. However, given the critical role of CTLs in immunotherapy, it is essential to deepen our understanding of CTL-induced cancer cell death.
This project aims to develop novel methodologies to investigate the death mechanisms that are initiated by CTLs. In addition, we will identify new pathways and mediators involved in CTL-induced cancer cell death. Furthermore, we will assess the contribution of proinflammatory cell death and lysis in promoting anti-tumor responses in vivo. Overall, this research will improve our knowledge of the molecular mechanisms of CTL-mediated cancer cell death and potentially reveal novel targets to enhance the efficacy of cancer immunotherapy.

T-cell-mediated proinflammatory cell death and lysis in cancer

Norge har under Parisavtalen påtatt seg en forpliktelse til å redusere klimautslippene sine med minst 55 prosent innen 2030 sammenlignet med nivået i 1990. Hittil har vi kun oppnådd 5 prosent av dette målet; vi har følgelig kun syv år på å redusere de resterende 50 prosent for å imøtekomme målene. For 2050 har vi satt oss enda større mål: å redusere klimautslippene med hele 90 prosent. Bakgrunnen for målene er at vi prøver å unngå de verste scenarioene FNs klimautvalg skisserer opp i sine rapporter. I tillegg har energiforbruket økt, og prognosene fra Statnett for ulike scenarier viser at energiforbruket skal fortsette å øke. Vi trenger følgelig mer energi. Dette har bidrat til at diskusjoner rundt kjernekraft i Norge er høyaktuelle og pågående. Norsk Kjernekraft AS har blitt opprettet med formål om å bygge Norges første kjernekraftverk. I tillegg ble det i fjor også bevilget 200 millioner kroner fra Regjeringen til et nytt senter som skal drive med nukleær forskning. Målet er å styrke den norske kunnskapsberedskapen på det nukleære området. Dette betyr at vi beveger oss i en retning hvor kjernekraft en dag kan bli aktuelt i Norge, og noen mener at dette er den eneste veien å gå hvis vi skal nå målene våre for 2050. Men i dag sitter vi med flere spørsmål enn svar, blant annet: Hvilken teknologi og leverandør burde vi velge? Er regelverket for snevert og strengt? Hvordan kan dette gjøres og samtidig ivareta sikkerhet? Hvordan tjener et kjernekraftverk penger?

Dette prosjektet skal svare på samtlige av disse spørsmålene i en detaljert og omfattende analyse. Her bruker vi grunnlagsdata til å vurdere dagens forskning opp mot hvordan industrien i andre land opererer, og ut ifra dette forsøke estimere hva risikoen knyttet til å bygge kjernekraft som industri i Norge er. Prosjektet sikter på å gi et så helhetlig bilde som mulig slik at vi kan gjøre oss opp en mening basert på fakta; om kjernekraft passer inn i miksen av det som utgjør norsk energi.

Denne prosjektbeskrivelsen skisserer forskningen Vala Maria Valsdottir (omtales herunder som stipendiaten) skal utføre innenfor sin doktorgradsavhandling. Prosjektet skal være en del av Senter for nukleær forskning (SNF) som fikk midler vinteren 2023.
Ph.d.-prosjektet er initiert av Inmeta Consulting AS, som igjennom denne ph.d.-stillingen vil bli en del av SNF. Inmeta planlegger en prosjektperiode på fire år, hvor stipendiaten arbeider 75 % med ph.d.-prosjektet og 25 % med vanlig virksomhet hos Inmeta. 
Prosjektet skal undersøke muligheten for kjernekraft som industri i Norge. Dette er både en samfunnsaktuell problemstilling samt en som medlemmene i SNF er interessert i å utforske. Stipendiaten kommer derfor til å samarbeide med alle aktørene som er involvert i senteret og benytte forskningsdata fra dem. Dette gjelder da spesielt Institutt for energiteknikk (IFE) som har mest grunnlagsdata som er relevant for dette prosjektet.
Kandidaten ferdigstilte en masteroppgave høsten 2020 i kjernefysikk på Universitetet i Oslo (UiO) og har derfor gode og relevante grunnkunnskaper for å ta på seg et slikt prosjekt. I tillegg har kandidaten tre års erfaring innenfor energibransjen, og er derfor oppdatert på industrien og forstår deres problemstillinger godt.

Er kjernekraft aktuelt for Norge?

Applied Biocatalysis is the development through science and engineering of useful biological catalysts and their commercial applications. The BNCM 2024 conference has an intention to gather approx. 100 international and national scientists from academia and industry in addition to PhD students from the international community. BNCM 2024 will be the 7th in this series of meetings with attendees from both academia and industry presenting and discussing the newest research in enzyme catalysed reactions in other media than water. The conference series was started in 1986 by professors Hans Tramper and Urs von Stockar, University of Wageningen, the Netherlands, and has later been chaired in Russia, Italy and Germany before Elisabeth Jacobsen at NTNU was give the honor of chairing the conference.  Collaboration in biocatalysis research has been supported by EU Frameworks as three subsequent COST actions. The first action, COST Action D25 "Applied Biocatalysis. Stereoselective and environmentally friendly reactions catalysed by enzymes " (2001-2007) was initiated and chaired by professor Thorleif Anthonsen, NTNU, the founder of the Biocatalysis Group at NTNU, which the project leader of this conference now chair. The strategy of applied biocatalysis fits well under the definitions of white biotechnology and Sustainable chemistry (Suschem) and fits also well with the EU Green Deal Strategy.

Sustainable cultivation of polychaetes as feed in aquaculture

I denne randomiserte kontrollerte studien har vi som mål å optimalisere behandlingen av pasienter med juvenil idiopatisk artritt (JIA) som er i stabil remisjon. Å finne riktig behandlingsnivå hvor risikoen for sykdomsoppbluss er balansert med byrden av å ta medisiner og deres bivirkninger er svært viktig for pasienter, foreldre og helsepersonell.

JIA er den vanligste kroniske revmatiske sykdommen hos barn og ungdom, og rammer flest jenter. Sykdommen er karakterisert av leddbetennelser, og kan føre til leddskader og funksjonstap. Heldigvis, takket være moderne medisiner og behandlingsstrategier, kan mange JIA-pasienter oppnå "vedvarende remisjon" - uten symptomer på sykdommen.

Men fortsatt vet vi ikke: hvordan bør JIA-pasienter som er i remisjon best behandles? Det finnes ikke kontrollerte studier som veileder oss, og det foreligger ingen anbefalinger om hvorvidt behandlingen bør trappes ned eller avsluttes.

Derfor introduserer vi "MOVE-JIA": en nasjonal studie som vil inkludere JIA-pasienter i vedvarende remisjon behandlet med tumor nekrosefaktor-hemmere (TNFi) og metotreksat. De vil bli tilfeldig delt i tre grupper: A) Gradvis seponering av TNFi, B) Gradvis seponering av metotreksat, eller C) Fortsatt stabil behandling med både TNFi og metotreksat. Studiens hovedmål er å måle hvor mange pasienter i hver gruppe som opplever sykdomsoppbluss i løpet av et år.

MOVE-JIA studien vil bli den første studien som undersøker om å redusere metotreksat fører til færre sykdomsoppbluss sammenlignet med å redusere TNFi hos JIA-pasienter i remisjon. Studien vil også vurdere om behandling med kun TNFi eller kun metotreksat er like effektivt som kombinasjonsbehandling.

MOVE-JIA studien er et nasjonalt samarbeid som inkluderer alle barnerevmatologiske avdelinger i Norge og aktiv deltakelse fra pasientene. Dette sikrer rask overføring av resultater til klinisk praksis. Funnene vil gi etterlengtet kunnskap om hvordan vi best skal håndtere barn og ungdommer med JIA i remisjon.

Juvenile idiopathic arthritis (JIA) is the most common chronic rheumatic disease in children and adolescents and occurs 3-6 times more often in girls. It is characterized by joint inflammation with a potential for joint destruction and disability if not treated. Sustained remission (no sign of active disease) is an achievable goal in JIA after the introduction of biological agents such as inhibitors of tumor necrosis factor (TNFi) and treat-to-target strategies. However, strategies to best treat JIA patients in remission remains unclear. No controlled studies have addressed the effect of tapering strategies for JIA in remission, and there are no current recommendations for stepdown or optimizing maintenance treatment. Given the risks, side effects and costs of JIA medications, treatment withdrawal can give patients and families relief and improve their quality of life. 
The MyJIA-Ahead trial is a national, investigator initiated, multicenter, 36 months, RCT of JIA patients in sustained remission on TNF-inhibitor and methotrexate. 150 JIA patients will be randomized 1:1:1 at baseline to A) Stepwise withdrawal of TNFi, B) Stepwise withdrawal of MTX, or C) Continued stable treatment with TNFi and MTX. The primary endpoint is proportion of patients with disease flare within 12 months. Key secondary endpoints are time to flare and time to regain remission after flare. If a disease flare occurs, the patient will return to the full dose study medication (the treatment the patient was receiving at baseline). Exploration of risk factors for disease flare includes patients reported health status and preferences, clinical characteristics, immunogenicity, molecular signalling (transcriptional, cellular and genetic risk) and ultrasound assessment of synovitis.

While evidence-based advice for starting therapies in active disease is available, The MyJIA-Ahead will provide much needed knowledge about treatment continuation in children and young adults with JIA in remission.

PhosBoard er en ny miljøvennlig fiberplate med lav tetthet utviklet av lavverdige restmaterialer fra treindustrien. Nye komponentkombinasjoner og prosesseringsmetoder for å produsere PhosBoards med forskjellige attributter og potensielle bruksområder vil bli utforsket i dette prosjektet.
Konvensjonell fiberplateproduksjon er sterkt avhengig av fossilt avledet lim, inkludert formaldehyd og energikrevende prosesser, som bidrar til miljøforringelse og klimagassutslipp. Dette prosjektet prioriterer bruk av bærekraftige og fornybare materialer i en lavenergiprosess for å minimere miljøpåvirkninger knyttet til konvensjonell fiberplateproduksjon. I fravær av formaldehydutslipp vil PhosBoard bidra til forbedret inneluftkvalitet og trygt arbeidsmiljø.
Utvalget av komponentene som brukes i PhosBoard tilbyr flere fordelaktige egenskaper, inkludert fysisk, mekanisk og brannhemmende. I tillegg er alle komponentene gunstige for økosystemet og dermed vil kasserte produkter ikke ha noen potensiell negativ innvirkning på miljøet. Prosjektet omfavner prinsippet om sirkulær økonomi ved å inkorporere resirkulerte trefiber fra industrielt og etterforbruksavfall for å fremme ressurseffektivitet og materialsirkularitet. Dette vil oppnås gjennom integrering av avfallsstoffer i produksjonsprosessen, og derved fremme lukkede sløyfematerialestrømmer og redusere avfallsgenerering ved produksjon av nye miljøvennlige produkter.

The "Norwegian Women and Cancer Study" - a representative population based prospective study

AXL er en reseptor tyrosinkinase som er høyt uttrykt i flere kreftformer, og aktivering av AXL er forbundet med celleoverlevelse, spredning, migrasjon og immunologisk respons. På grunn av at AXL er involvert i mange cellulære prosesser på tvers av forskjellige ondartede vevstyper er det et kritisk behov for å forstå og modulere Axl-signalering i tumormikromiljøet.
Målet med dette prosjektet er å forbedre forståelsen av den molekylære mekanismen for interaksjonen mellom AXL og GAS6, PtdSer, og det terapeutiske antistoffet tilvestamab ved å bruke strukturell biologi som forskningsmetode. Metodene som blir brukt er cryo-elektronmikroskopi og røntgenstråling krystallografi. 
Etter at den globale Covid pandemien hadde startet  ble AXL identifisert som en mulig bindingspartner av COVID-19 sitt spike-protein. Et samarbeid med professor Wendy Maury ved Universitiy of Iowa bekreftet at AXL medierer en økning i COVID-19-infeksjon. Dette prosjektet viste også at mekanismen ikke er gjennom en direkte interaksjon med spike-proteinet, men snarere en interaksjon med et lipid i virusmembranen. Disse resultatene er nå del av en publikasjon i tidsskriftet PLoS Pathogens.

The receptor tyrosine kinase Axl regulates vital physiological processes including cell survival, cell proliferation, migration, motility, epithelial to mesenchymal transition, and immunologic response. Axl activation is associated with invasiveness and resistance to therapy in many cancers and has recently been shown to play a key role in a range of fibrotic diseases.
Axl is activated by its ligand, growth arrest-specific protein 6 (Gas6). Optimal activation of Axl requires clustering of Gas6 through binding to the lipid phosphatidylserine (PtdSer), found on the membranes of dying cells. This interaction depends on the Vitamin K-dependent post-translational modification of several glutamate residues near the N-terminus of Gas6 to ?-carboxyglutamate. In this project, in order to better understand extracellular events in Axl signaling, the structure of the entire extracellular domain of Axl receptor in complex with Gas6 and phosphatidylserine will be determined.
BerGenBio is focussed on the pre-clinical and clinical development of Axl inhibitors, including a high affinity function-blocking antibody tilvestamab that targets the N-terminal domain of Axl. To provide insight into the mode of action of tilvestamab, the crystal structure of Axl extracellular domain in complex with tilvestamab will also be determined. 
Key R&D challenges with this project will be the production of sufficient quantities of the individual protein components and finding appropriate conditions for the growth of well-diffracting crystals for X-ray structure determination. Gas6 is expected to be especially challenging due to the difficulty in ensuring complete ?-carboxylation.
The anticipated results will provide us with a clearer understanding of the molecular details of Gas6 activation of Axl, and how this activation can most effectively be inhibited therapeutically.

Molecular interactions important for Axl activation

Lungekreft er fortsatt den vanligste årsaken til kreftrelaterte dødsfall med anslagsvis 1,6 millioner tilfeller globalt som fører til 1,4 millioner årlige dødsfall. BerGenBio har nylig rapportert lovende resultater fra studier der Axl-hemmeren bemcentinib kombineres med immunterapi (pembrolizumab) hos disse pasientene. For å forstå hvilke pasienter som kan ha nytte av denne behandlingen, må man ta i bruk nye analyseverktøy som inkluderer «høydimensjonal» tumor bildeanalyse, serumproteomikk og DNA/RNA-sekvensering for på den måten å hente ut mest mulig informasjon fra kliniske prøver. Dette prosjektet har som mål å etablere nye analysemetoder som skal anvendes i BerGenBio sine pågående kliniske studier med lunge- og andre kreftformer, og å integrere data fra både nye og eksisterende plattformer for å bygge en omfattende modell som kan forklare mekanismen bak medisinenes effektivitet og forutsi hvem som vil respondere på behandlingen. 

Det siste året har det blitt utviklet verktøy som gir oss mulighet til multidimensjonal bildeanalyse av tumor vev og verktøy for bearbeiding og analyse av disse bildene. I tillegg har det blitt tatt i bruk maskinløringsverktøy som gir muligheten til å forutsi hvilke pasienter som vil respondere på behandling med bemcentinib. Det er ventet at resultater fra dette arbeidet vil bli publisert det neste halve året.

Immunotherapy, the therapeutic targeting of a patient’s immune system to treat cancer, has rapidly become established as a key treatment for multiple cancer indications. However, only a minority of patients respond to the treatment. An important challenge in immunotherapy is now to identify, in advance, which patients are likely to respond. BerGenBio has recently shown that the combination of bemcentinib, a selective Axl receptor inhibitor, with the immunotherapeutic pembrolizumab benefits a subset of particularly hard-to-treat non-small cell lung cancer (NSCLC) patients. This project will use cutting-edge high-dimensional imaging to examine tumor material collected from patients prior to treatment with bemcentinib and pembrolizumab in order to identify tumor- and immune-associated features that predict whether the patient will benefit from taking the drugs.
Establishing an Axl- and immunotherapy-targeted panel to extract single-cell information from patient samples using this imaging technology and analyzing the resulting data with respect to the spatial relationships between cells are both key R&D challenges in this project (Aims 1-3). The panel will first be used to analyse a collection of samples from NSCLC patients but will be expanded to analyze samples from patients receiving bemcentinib treatment for other diseases (Aim 4).
Patient samples from BerGenBio clinical trials are routinely tested with pre-validated assays to measure circulating protein biomarker levels and gene/protein expression in tumor tissue. These “multi-omics” data will be integrated with the imaging data (Aim 5) to yield comprehensive signatures of response, build on our existing mechanistic knowledge of Axl inhibition in cancer, and identify novel predictive biomarkers that can be tested using routine clinical diagnostics. Overall, we aim to demonstrate that we can identify patients who will benefit from Axl-inhibiting drugs.

Integrated multiomic analyses of patients treated with Axl inhibitors to predict therapy response, clinical benefit and mechanism of action.

Fibrose, økt bindevevsmengde i et vev eller organ, er en viktig årsak til sykdom og død over hele verden. Det er derfor et betydelig behov for nye behandlingsformer som kan forhindre fibrose. Bioteknologifirmaet BerGenBio ASA utvikler nye hemmere av AXL/EMT signalveien, en mekanisme som ikke bare driver legemiddelresistens og spredning (metastase) av kreft, men som også spiller en viktig rolle for utvikling av fibrose og i ulike infeksjonssykdommer som COVID19. I dette prosjektet skal BerGenBio forsøke å vise at hemming av AXL er en sikker og effektiv måte å behandle fibrose i pasienter, og å forstå mekansimene for hvordan hemming av AXL hindrer og reduserer fibrose. For å nå disse målene samarbeider vi tett med akademiske grupper som er eksperter på AXL singnalering og hvilken rolle AXL spiller for sykdomsutvikling i nyre, lever og lungefibrose. Pre-kliniske data fra dette prosjektet har blitt presentert som ePoster presentasjon ved ERA-EDTA i mai 2022 og som poster ved EASL i juni.

BerGenBio ASA is a clinical-stage oncology company advancing first-in-class, selective AXL kinase inhibitors against aggressive diseases. The receptor tyrosine kinase AXL is becoming a profiled target for developing drugs to treat resistant and metastatic cancer. Over the last decade it has become evident that the AXL pathway also plays a decisive role in fibrosis. Fibrotic diseases of the lung, liver and kidney are a significant cause of death worldwide. BerGenBio’s selective AXL inhibitors are effective in preventing fibrosis in a variety of model systems. The objective of this project is to perform research and development to demonstrate that AXL-inhibition is a safe and effective treatment for fibrosis in human patients and to deepen our understanding of the mechanisms by which AXL-inhibition prevents and reduces fibrosis. To achieve these goals, BerGenBio has established collaborations with academic groups that are experts in AXL signaling (Prof. James B Lorens, Department of Biomedicine, UoB, Bergen, Norway) and on the role of AXL signaling in liver fibrosis (PhD Albert Morales, Institute of Biomedical Research, Barcelona, Spain).

AXL as a therapeutic target in fibrosis; biology and biomarkers

BerGenBio er et biofarmasøytisk selskap som utvikler nye (first-in-class) legemidler til behandling av flere aggressive kreftformer. Selskapets fremste legemiddelkandidat bemcentinib (BGB324) er rettet mot et protein på celleoverflaten som driver spredning av kreft, svekkelse av kroppens immunforsvar mot kreftceller og legemiddelresistens. Dette prosjektet tar sikte på å støtte flere "investigator initiated" trials med bemcentinib som har blitt designet og vil bli gjennomført av ledende leger over hele verden. Indikasjonene er valgt på grunnlag av laboratorieforsøk der bemcentinib har hatt god effekt og derfor har potensiale til å gi stor nytte for pasienter. Fem av disse studiene med utprøving på pasienter med melanom, lungekreft,   bukspyttkjertelkreft, glioblastom og mesoteliom har allerede startet og rekrutterer pasienter, mens rekruttering er ferdigstilt  studien for akutt myelogen leukemi (AML) og myelodysplastiskt syndrom (MDS).

BGB324 is a first-in-class experimental drug inhibiting the hallmarks of aggressive cancers including acquired drug resistance, immune evasion and metastasis; this represents the most significant unmet need in cancer treatment. 

BGB324 is a highly selective inhibitor of the cancer cell signalling protein called AXL. Increased AXL expression correlates significantly with poor prognosis in many cancers and therefore represents a very large addressable market. 

BGB324 is in phase II clinical trials in patients with leukaemia and lung cancer. Encouraging clinical data already published confirms BGB324 is well tolerated by patients and a proportion of patients have reported clinical benefit and long term disease control. 

This funding will support Investigator initiated clinical trials (IITs) in Norway and overseas. IITs are clinical trials led by patient facing doctors who act as the regulatory sponsor and are partially supported by BerGenBio in bespoke clinical development collaborations. IITs represent good value additional clinical research that benefits patients and expands the potential clinical utility of BGB324. 

The IITs in this project will include patients with different cancers with high unmet medical need, where there is a strong scientific rationale for treatment with BGB324.

The data generated from these IITs will enlarge our experience with BGB324 in cancer patients, complement our safety database as well as inform future clinical studies and BGB324s route to market.

Investigator-Initiated Trials for AXL driven cancers with high unmet clinical need

Fibrose, økt tilkobling i et vev eller organ, er en viktig årsak til sykdom og død over hele verden. Det er derfor et betydelig behov for nye terapier som kan forhindre fibrose. Bioteknologiselskapet BerGenBio ASA utvikler inhibitorer av AXL / EMT signalveien, en mekanisme som ikke bare driver medikamentresistens og metastase, men også spiller en viktig rolle i utviklingen av fibrose. Vi har vist at spesifikke hemmere av AXL reduserer lunge-, lever- og nyrefibrose. Resultatene fra disse studiene klargjøres for tiden for publisering i internasjonale tidsskrifter. I dette prosjektet har BerGenBio også utviklet et nytt humanisert anti-AXL antistoff som hemmer AXL aktivitet. Dette antistoffet blir for tiden testet klinisk i et fase 1 studie og vil bli testet i pasienter ved utgangen av året.

Fibrotic disease, the accumulation of extracellular matrix components in organs or tissues, is a leading cause of morbidity and mortality worldwide. Nearly half of all deaths in the developed world are associated with or characterized by some type of chronic fibrotic disease. The biotechnology company BerGenBio AS has recently discovered that the Axl/EMT pathway, a focus of therapeutic development to treat drug resistant and metastatic cancer, also plays a decisive role in fibrosis. Fibrosis remains an area of a great and unmet need, and the funding from this project will facilitate BerGenBios research efforts to validate that inhibition of the Axl/EMT pathway is effective at preventing and reversing fibrotic disease.

The Axl/EMT pathway in fibrotic disease

In the Western world, 1 of 3 people die of cancer and lung cancer is the most prominent type. The vast majority (85%) of lung cancer patients are diagnosed with advanced disease where the tumor is more aggressive, resistant to available anti-cancer drugs and less sensitive to radiation. Consequently, there is a significant medical need for new treatments that can improve the treatment of lung cancer patients. 

We have identified a cellular signaling factor involved in resistance to anti-cancer treatments in many lung cancers. This factor has been proven important in epithelial to mesenchymal transition (EMT), a program utilized by several cancers to gain resistance to therapy and evade recognition and killing by the immune system. In addition, this factor also mediates transformation of epithelial breast cancer cells into a more basal like aggressive phenotype. We have discovered that it is the nuclear localization of this signaling factor that is mediating its tumor driving capacity and that preventing nuclear localization prevents tumor initiation. The focus of this project is to define the role of this novel signaling factor in cancer therapeutic resistance and validate inhibitors for this target. Novel tool compounds for this target is under evaluation for their therapeutic effect in cancer xenograft models.

Epithelial-mesenchymal transition (EMT) is an important pathway during embryonic development that allows differentiated epithelial cells to regain plasticity and turn into cells with a mesenchymal phenotype. Inappropriate activation of EMT in tumors is em erging as a predominant pathway in the development of metastases and drug resistance, the two critical traits that underpin most mortality from cancer. 
We recently identified the protein kinase Akt3 as a key component in the maintenance of EMT and stem c ell-like activity in triple-negative breast cancer (Tiron et al., submitted). Recent studies have shown that Akt3 also plays a role in EMT-dependent alternative splicing in non-small cell lung cancer (NSCLC) (Sanida, 2014) suggesting that nuclear targetin g of activated Akt3 may be a key molecular mechanism of EMT-dependent functions in carcinomas other than TNBC, and in particular in NSCLC with a mesenchymal gene expression phenotype. 
NSCLC accounts for approximately 85% of all lung cancers, and is often  diagnosed late when cancer spreading has already occurred. Most lung cancers cannot be cured with currently available therapeutics, so this disease represents enormous unmet clinical need. In the course of this project we aim to demonstrate that in NSCLC , as in TNBC, Akt3 plays a critical role in cancer EMT and in the associated development of metastases and drug resistance. Objectives 1-3 focus on identifying how often Akt3 is expressed in NSCLC, where it localizes within the cell, and how this correlat es with different subtypes and stages of NSCLC. Objectives 4-7 will demonstrate that experimental disruption of Akt3 signaling results in changes in the growth, invasiveness and drug-resistance of Akt3-expressing NSCLC cell lines in vitro. Finally, in Obj ectives 8-9, we will demonstrate that these results can be extended to the more complex situation that pertains in vivo. Overall we aim to demonstrate that Akt3 represents a novel therapeutic target in NSCLC.

Akt-isoform specific treatment approaches for lung cancer

BerGenBio utvikler nye legemiddelkandidater mot en grunnleggende biologisk mekanisme (Axl/EMT) som driver spredning og legemiddelresistens i aggressiv kreft. Fremdriften i prosjektet har i hovedsak fulgt planen som ble lagt, og har ført til utvikling av flere nye legemiddelkandidater mot Axl/EMT signalveien. I den siste delen av prosjektet har vi studert 5 legemiddelkandidater som angriper forskjellige aspekter av Axl/EMT, og vi har startet storskala produksjon av en av disse legemiddelkandidatene for å kunne starte klinisk utprøving.

Aggressive, metastatic cancers that are resistant to current anti-cancer treatments are the major cause of cancer death. The biotechnology company BerGenBio AS is focused on developing a pipeline of innovative therapeutics that target the biological mecha nisms underlying drug resistance in cancer cells.

The focus of this project is to implement state-of the-art R&D strategies to identify and develop novel drug candidates with the potential to prevent and reverse cancer drug resistance in the metastatic s etting. The funding applied for will facilitate our research efforts to define new therapeutic modalities to treat malignancies with unmet need.   

The output of the project proposal will be R&D leading to first-in-class drug candidates for: 1) licensing  to larger pharmaceutical companies for clinical development and marketing; or 2) co-developed with a suitable partner through later stage clinical proof of concept prior to subsequent large pharmaceutical company licensing. This business scenario is both  attractive to the investment community and can provide new therapeutic options for cancer patients.
