The mechanisms of pathogenesis of infection with infectious pancreatic necrosis virus (IPNV) are not well understood. This project will therefore address the cellular effect of individual proteins of IPNV expression of cloned proteins of infectious pancre atic necrosis virus in transfected cell cultures. The known sequences for individual virus proteins - Vp2, Vp3 og Vp5 will be cloned into defined vectors and subsequently used to transfect well established cell lines supporting the replicaiton of virus. T he expression of proteins will be assessed using immunfluorescence methods or through expression of fusion proteins coding for the green fluorescent protein. The effect of cell growth and viability will be assessed using standard methods to characterise c ell integrity and markers of apoptosis mechanisms. Cell lines persistently infected with IPNV serotype Sp will be established and transfected to characterise the interaction between whole virus and individual protein expression. Final ly, hybrid virus will be generated through a reverse genetics system and the effect of gene deletion and site-directed mutations on virus replication on virus assembly and growth will be studied. Also any difference in recovery of high and low virulent pr ofiles of Vp2 will be subject to detailed study.
