Nephrocalcinosis, or kidney stones, is reported by the industry as one of the most important risk factors for the production of farmed salmon. Kidney stones in salmon were first described in scientific literature in 1999 and are characterized by the precipitation of minerals in the kidneys. In humans, nephrocalcinosis is a multifactorial disease, and it is likely that this also applies to farmed fish. Nephrocalcinosis has been reported in a wide range of different fish species, both for freshwater and marine species, but primarily in aquaculture rather than in wild fish populations. Both in acedemia and in the industry, there is a common perception that nephrocalcinosis is linked to intensive production, but several different causative factors have been suggested. High CO2 levels in the water have long been the primary proposed cause for nephrocalcinosis, although the published studies show conflicting results. The main aim of this project was to provide a better understanding of the prevalence and severity of nephrocalcinosis in farmed salmon and to assess the extent to which the condition is linked to environmental factors. Field work was carried out at 11 hatcheries and their receiving facilities at sea in central and western Norway. Samples were taken from 12 fish groups and the data collection consisted of feed and water samples, in addition to kidney and gill tissue, and plasma from fish. In one of the 11 hatcheries, a group of fish was monitored for 6 months with monthly sampling of the same samples. Nephrocalcinosis was detected in all hatcheries, but the extent varied between facilities, from less than 5% to 100% of sampled fish. Samples were taken of the precipitates and the results showed that kidney stones in salmon mainly consisted of amorphous carbonate apatite (amCAP: calcium, phosphate and carbonate), which precipitates at pH > 6.8. We also found that the blood chemistry of fish with nephrocalcinosis was significantly different from fish without nephrocalcinosis, which points to disturbed physiology and homeostasis in fish with nephrocalcinosis. Histopathology was the most precise tool for assessing the severity of nephrocalcinosis, but requires the fish to be euthanized. We therefore explored radiology as a non-invasive tool for the detection of nephrocalcinosis. The results showed that amCAP was detectable by X-ray, although further optimization should be carried out before the method can be used with high enough sensitivity and specificity. It would be beneficial to use radiology to test the reversibility of nephrocalcinosis under controlled conditions. The method also shows potential as an initial investigation of nephrocalcinosis, with follow-up samples treated with histopathology, saving time, money and fish. A multivariate analysis indicated that the most important factor for the occurrence of nephrocalcinosis was the addition of seawater during production. In the long-term monitoring, we observed a temporal relationship between increased salinity and an increased incidence of nephrocalcinosis. The fish showed signs of prolonged osmoregulatory stress with elevated levels of divalent ions in plasma. It has previously been suggested that electrolyte homeostasis, osmoregulation and mineral metabolism are closely related in salmonids, and from our data the most likely explanation for the connection between seawater supplementation and nephrocalcinosis was osmoregulatory stress. The mechanism for calcium phosphate precipitation in the kidneys is still not known, but may be linked to urinary pH in Atlantic salmon combined with osmoregulatory stress.

Doktorgradsprosjektet har gitt bedriften økt kompetanse innen betydningen av vannkvalitet og fôrsammensetning for helse og velferd hos settefisk. Videre har prosjektet bidratt til at bedriften har fått på plass flere ulike metoder og analyseverktøy som kan benyttes i tjenester til blant annet havbruksnæringen. Doktorgradsprosjektet har åpnet opp for forskningssamarbeid med flere ulike akademiske partnere og gitt direkte tjenesteoppdrag fra næringen. Resultatene fra prosjektet har ført til at en rekke settefiskprodusenter har gjort tilpasninger og endringer i produksjon for å redusere innslaget av nefrokalsinose på sin fisk. Totalt sett har prosjektet bidratt med økt kunnskap om en produksjonslidelse som er utbredt blant oppdrettet fisk og således bidratt til økt helse og velferd hos oppdrettsfisk.

Det er et økende fokus på helse og velferd i akvakulturnæringen, både hos matfisk (i sjø) og hos settefisk (på land). En problemstilling i settefisknæringen er nefrokalsinose, som er en subletal tilstand som beskrives som en utfelling av kalsium- og magnesiumsalter i nyrene. Utfellingene på nyrene forstyrrer normal nyrefunksjon og medfører fysiologisk ubalanse blant annet hos laksen. Det er sannsynlig at den forstyrrede nyrefunksjonen svekker laksen og gjør den mer utsatt for stress og sykdom. Nefrokalsinose er ikke en ny problemstilling i oppdrettet laks, og sees relativt hyppig i settefiskanlegg. Den opptrer både i anlegg med resirkulering av vannet (RAS) og i tradisjonelle gjennomstrømningsanlegg. Bransjen melder om en økning av tilstanden i både RAS- og gjennomstrømningsanlegg, men også etter utsett i sjø. Tilstanden er også relativt vanlig hos andre anadrome laksefisk, som regnbueørret. Det er gjort svært lite undersøkelser av hva nyreutfellingene består av. Det er heller ikke kjent om utfellingene er forskjellige mellom enkelte individ eller anlegg. Vi vet i dag ikke hvilke mekanismer som ligger til grunn for utviklingen av nefrokalsinose og kunnskapsgrunnlaget er begrenset. Prosjektets hovedmål er derfor å øke kunnskapen om nefrokalsinose og hvilke faktorer som spiller inn i utvikling av tilstanden. Dette vil bli oppnådd ved å undersøke, i felt og i laboratorieeksperimenter, hvordan ulike miljøfaktorer, som vannkvalitet og fôrsammensetning, påvirker dannelse og utvikling av nyreforkalkninger hos laks.
