Garden shrubs are popular plants in Norwegian gardens because they have beautiful flowers or fruits, and because they are shields from roads and insight from neighbors. The shrubs also bring a small piece of the forest into the residential areas - something both humans and birds appreciate. However, many of these popular garden shrubs do not belong in Norway and can have serious consequences for biodiversity if they spread outside the garden fence. Plants that constitute very high risk or high risk in Norwegian nature are listed on the Norwegian Biodiversity Information Centre's "Foreign species list". For example, they can affect the natural ecosystem and out-compete native species, or act as weeds in agriculture and in private gardens. The selection of Norwegian-produced garden plants can be greatly reduced if the Foreign Species list is used. This will also provide guidance for breeding and plant selection in the future. Selection work in the Nordic countries has been aimed at finding plants that can easily be propagated in nurseries and at the same time withstand the Nordic climate. Features that are focused on are hardiness, coverability, growth power and reproductive ability. It is a paradox that the same characteristics are now considered as risk characteristics, and entails entry on the Foreign Species List. In this project, Eliteplanter Norge SA wants to use the gene editing technology CRISPR to develop sterile garden plants that pose less risk to Norwegian nature. Using CRISPR technology to make garden plants sterile will both be a new production method and result in a sustainable product with completely new properties that will be of great economic interest to Norwegian garden shrubs producers. We have now established methods for growing these garden shrubs on plates in the greenhouse, and done transcriptome sequencing to identify active genes. Based on these transcriptome analyzes, we have identified a number of candidate genes that we will try to eliminate to make the plants sterile, and we have now made modifications to the CRISPR scissors that hopefully will knock-out these. One of the biggest bottlenecks in gene editing of plants is the transfer of the CRISPR tool to the plant cells and then microforming them so that they can develop into adult plants. These methods often need to be established and optimized for each individual species. We have now succeeded in establishing such methods for several of the species, and we have plants that have been modified with bacteria containing the CRISPR tool. Over the next few weeks, we will analyze the first plants to see if they have mutations in the desired gene.
