Dissecting bacterial subpopulation physiology with activity-based probes

Awarded: NOK 0.49 mill.

The long-terme aim of the ERC project is to establish a new biomarker system for bacterial physiology that is based on labeling with fluorescent chemical probes. To demonstrate the feasibility of this approach, we will perform an unbiased functional characterization (transcriptomic and growth analysis) of activity-based probe-labeled bacterial subpopulations, elucidate the molecular/cellbiological basis for probe labeling, overcome technical pitfalls and analyse potential artifacts. We will also perform pilot studies that assess the suitability of a diverse, expanded toolset of chemical probes for differentiating microbial single-cell and subpopulation physiology. Together, these experiments will lay the groundwork for the systematic exploitation of diverse chemical probes as biomarkers for subpopulation phenotypes (or phenoprobes) in the upcoming ERC-2022-COG application.
