KVAL: Transcriptional addictions in cancer

Glioblastoma multiforme (GBM) is the most prevalent and aggressive malignant tumor of the central nervous system. The median overall survival of GBM patients is around one year, making it one of the deadliest cancers. The standard care is surgical resection followed by radiotherapy and chemotherapy with temozolomide (TMZ). TMZ targets the DNA replication machinery of cells, and often causes very strong side effects. Many cancer types, including gliomas, have aberrations in the way the regulate protein production, and this affects their development. We have therefore studied the possibility of targeting such regulatory mechanisms in glioma cells, by inhibiting enzymes that modulate protein synthesis. Our results suggest that glioma cells are exquisitely sensitive to the inhibition of one family of such enzymes: Glioma cells responded to two different small-molecule inhibitors at 10 to 100 times lower concentrations than the control cells, indicating a large therapeutic window. This project allowed us to identify known molecules with the ability to bind the targeted enzymes. This lets us develop new molecules with improved properties that may later be tested for efficacy in animal experiments.

Prosjektet har bidratt med ny kunnskap om hvordan man kan oppnå effekt mot kreftceller ved hemming av enzymer som er involvert i proteinproduksjon. Dette er en helt ny tilnærming til kreftbehandling, med potensiale for kraftig reduserte bivirkninger sammenliknet med etablerte behandlingsformer som stråleterapi og cellegift. I prosjektet har vi samarbeidet med internasjonale tjenestetilbydere, og lyktes med å overføre teknologi fra disse til vårt forskningsmiljø ved OUS. Vi har også utviklet kompetanse på legemiddelutvikling innen kreftbehandling, og søker å bygge videre på resultatene fra dette prosjektet for vellykket kommersialisering av et nytt legemiddel mot hjernekreft.
