Forskere har lenge debattert om spesifikke endringer i måten genene våre reguleres på, kalt epigenetiske endringer eller "epimutasjoner," kan øke risikoen for kreft. Våre nylige banebrytende undersøkelser har gitt sterke bevis på at epimutasjoner faktisk er en risikofaktor for visse typer kreft.
I en studie av en stor gruppe kvinner fant vi at de som hadde epimutasjoner i BRCA1-genet, hadde mye større risiko for å utvikle trippel-negativ brystkreft og høygradig serøs eggstokkreft senere i livet. Disse epimutasjonene ser ut til å oppstå tidlig under fosterutviklingen, og fører til at det fødte barnet (og senere det voksne individet) har en mosaikk av celler i kroppen, der noen få celler bærer epimutasjonen.
Nå har vi som mål å utvide disse studiene til andre gener og andre former for kreft. For å gjøre dette vil vi analysere blodprøver fra en studie som har rekruttert over 160 000 kvinner i USA.
Hvis denne forskningen lykkes, kan den revolusjonere vår forståelse av hvordan kreft utvikler seg og føre til bedre forebyggende tiltak. Det kan også ha store implikasjoner for kreftscreeningprogrammer og genetisk veiledning for personer med høy risiko for visse typer kreft.
Alt i alt fremhever vår forskning den viktige rollen som epigenetiske endringer kan spille i kreftutvikling og understreker behovet for ytterligere undersøkelser på dette området.

Cancers result from accumulation of genetic and epigenetic alterations. The minority of cancers are due to inherited (germline) alterations that affect all cells in the body. On the other hand, the majority of cancers (>85%; sporadic cancers) are believed to occur due to localised alterations that accumulate after birth as a result of stochastic processes and/or environmental effects. Recently, in a major breakthrough work, we have provided definitive evidence that some sporadic cancers arise in adult life due to mosaic BRCA1 epimutations that occurred during the early embryonic development. BRCA1 epimutations have a surprisingly high population frequency (>5% of adult women) and hazard ratios for breast and ovarian cancer (2.4 and 1.9, respectively). Together with the findings of epimutations in other tumour suppressor genes (MGMT, MLH1, etc) this has led us to hypothesise that a substantial fraction of other sporadic cancers can be attributed to prenatal epimutations in other tumour suppressor genes.

In this project, we will use ultradeep targeted methylation sequencing for quantitative and qualitative analyses of epimutations. Evaluation of cancer risk related to epimutations will be done by studying samples from prospective biobanks (WHI) with samples from healthy women that remained cancer-free or developed different types of incident cancers years/decades after samples were taken. We will also characterise epimutations present in samples of newborn boys and girls, as well as their parents (MoBa biobank). Finally, using cryopreserved blood-derived stem-like cells from young healthy males and female breast cancer patients (in-house biobank), we will assess mutational and epimutational mosaicism at the single-cell level, generating clonal phylogeny for the methylated cells and thus providing insights on early-developmental processes that create epimutations.

Mosaic epimutations: population prevalence, mechanistic origins, and their role in cancer

Kunstig lys om natten (ALAN) setter naturen ut av balanse. Mange dyr er avhengige av lengden på dagslyset (fotoperioden) som signal for å synkronisere reproduksjon med gunstige forhold. Forstyrrelser i dette naturlige signalet, enten gjennom lysforurensning fra urban belysning eller akvakultur, kan ha alvorlige konsekvenser for dyrs livssyklus og overlevelse. Prosjektet mitt tar sikte på å avdekke de biologiske mekanismene bak sesongmessig reproduksjon for bedre å forutsi og redusere disse påvirkningene. Jeg vil undersøke et nøkkelhormon, thyroidstimulerende hormon (TSH), og dets funksjon i reguleringen av sesongmessig reproduksjon hos dyr ved å bruke medaka (japansk risfisk) som modell.
Denne lille fisken er perfekt for genetiske studier, da det allerede finnes mange verktøy og protokoller for denne arten for å studere i detalj de cellulære og molekylære mekanismene som er involvert i essensielle kroppsfunksjoner, som for eksempel reproduksjon. For å undersøke dette vil jeg bruke avanserte genetiske verktøy og optogenetikk, en metode som bruker lys til å kontrollere celleaktivitet, for å avdekke mekanismen som regulerer det sesongmessige tidspunktet for reproduksjon. Ved å dekode hvordan dyr tilpasser sin sesongmessige reproduksjon til forstyrrelser i lyssignaler, vil dette prosjektet belyse hvordan vi kan ivareta biologisk mangfold og utvikle bærekraftig akvakulturpraksis.

Seasonal reproduction in vertebrates ensures offspring survival by aligning reproductive timing with photoperiod as the primary cue. However, artificial light at night (ALAN) disrupts these cues, posing a threat to species survival. Similarly, continuous light used in aquaculture to inhibit fish reproduction may have unintended physiological effects. OptoSeason seeks to decode the mechanisms underlying seasonal reproduction as a critical step toward predicting and mitigating the impact of these environmental disturbances.
Research in mammals and birds suggests that thyroid-stimulating hormone (TSH) in the pituitary pars tuberalis (PT-TSH) regulates seasonal reproduction. However, the role of PT-TSH remains controversial due to the challenges of knocking out the Tshb gene without affecting essential thyroid functions. Thanks to the teleost-specific whole-genome duplication (WGD), we can now study a TSH gene specifically involved in seasonal reproduction using teleost fish models. Our previous findings indicate that tshbb plays a crucial role in this process.
To unravel these mechanisms, OptoSeason will develop a next-gen optogenetic tool in Japanese medaka, a seasonally breeding teleost suitable for genetic manipulation and knockout studies. Considering its potential, this tool also has broader applications across various biological research areas. We will also employ advanced techniques such as CRISPR/Cas9-mediated knockout/knock-in, receptor assays, laser microdissection, transcriptomics, calcium imaging, and patch clamping to explore the tshbb pathway and its link to reproduction.
Understanding this pathway will help identify markers for out-of-season spawning risks due to ALAN and provide insights into environmental and genetic influences on reproduction. This research will address challenges like early puberty in aquaculture, advance scientific knowledge, and develop tools to tackle key environmental and industrial issues.

OptoSeason: decoding seasonal reproduction in vertebrates using a next-gen optogenetic tool

Cøliaki er en sykdom som rammer 1-2% av befolkningen. Den eneste eksisterende behandlingen for sykdommen i dag er en livslang glutenfri kost. Det er ønskelig å utvikle alternative behandlingsmetoder. Nyere forskning har avdekket at enzymet transglutaminase 2 spiller en sentral rolle i utviklingen av sykdommen. Dette enzymet forandrer glutenproteiner slik at immunsystemet hos cøliakere kan reagerer på disse proteinene. Denne immunresponsen hos cøliakere involverer B-celler. B-celler produserer antistoffer, og cøliakere har B-celler og antistoffer som er rettet mot transglutaminase 2. I projektet ønsker vi å ta denne kunnskapen i retning mot utvikling av nye behandlingsprinsipper. En del av prosjektet dreier seg om hemming av enzymaktiviteten til transglutaminase 2, og å avdekke hvor i kroppen hvor transglutaminaseaktiviteten bør hemmes. En annen del av prosjektet dreier seg om å finne en metode som kan elimimere B-cellene som gjenkjenner transglutaminase 2.  Prosjektet vil studere celler og vev fra cøliakere samt mus som er genetisk modifisert til å uttrykke reseptorer som brukes av cøliakere til å gjenkjenne gluten og transglutaminase 2.

Celiac disease is a prevalent autoinflammatory condition caused by immune reaction to dietary gluten proteins. The only available treatment is lifelong avoidance of dietary gluten. However, symptoms and enteropathy often persist in patients who attempt to adhere to a gluten-free diet. Thus, there is a need to develop non-dietary therapies for celiac disease. 

The enzyme transglutaminase 2 (TG2) has critical roles in celiac disease pathogenesis. The enzymatic activity of TG2 creates by deamidation gluten peptides that have high affinity for disease-predisposing HLA-molecules, and these deamidated peptides are recognized by the disease-driving CD4+ T cells. Moreover, TG2 is the target of a highly specific autoantibody response. In this project we will explore two novel treatment avenues focused on TG2. In the first approach, we seek to develop novel immunotherapeutic molecules, Bispecific AutoAntigen T-cell Engagers (BiAATEs), to specifically eliminate the subset of autoreactive TG2-specific B cells while avoiding general immunosuppression. The results will be important not only for celiac disease but also for other autoimmune diseases having cytosolic autoantigens.

Second, we will explore therapeutic inhibition of TG2. ZED1227, an irreversible inhibitor of TG2, was recently shown in a phase 2 clinical trial to prevent gluten-induced intestinal damage in celiac disease patients. It is however unknown where in the body the drug is active. Our group has obtained evidence that the gut lumen likely is the relevant site where TG2 is active and encounters gluten peptides for deamidation. We will test the new hypothesis that TG2 in gut lumen is pathogenic in celiac disease and will explore inhibition of gut luminal TG2 as therapy.  

Both arms of the project will combine studies on patient samples with mechanistic experiments in state-of-the art humanized mouse models that we recently developed.

Exploring novel therapies for celiac disease centered around the autoantigen transglutaminase 2

Mennesker påvirker marine økosystemer på mange måter, som gjennom klimaendringer, forurensning og overfiske. Kumulative effekter refererer til de samlede effektene av slike stressfaktorer. Disse utgjør en av de største truslene mot økosystemer og biologisk mangfold. Likevel vet vi lite om hvordan stressfaktorer virker sammen i komplekse økosystemer. Dette gjør det utfordrende å forvalte våre kyst- og havområder bærekraftig.

CLIMAX-prosjektet har som mål å bedre forstå kumulative påvirkninger gjennom datasimuleringer. Det introduserer innovative modellerings- og dataanalysemetoder som kombinerer dagens datakraft med kunnskapen som økologer har bygget i flere tiår gjennom felt- og laboratoriestudier.

CLIMAX vil vise potensialet til disse nye metodene med en casestudie: menneskelige påvirkninger i Skagerrak og Kattegat. Her tar prosjektet for seg to sentrale miljøspørsmål. For det første vet vi allerede mye om de enkelte stressfaktorenes effekter på individuelle arter – men hvordan samhandler disse stressorene ved å skape økosystemendringer som redusert biologisk mangfold og produktivitet? For det andre, hvordan påvirker langvarig stress økosystemets evne til å motstå og komme seg etter ekstreme hendelser som marine hetebølger?

Marine økosystemer er bemerkelsesverdig stabile, men kumulative påvirkninger kan overvelde deres evne til å motstå og komme seg etter forstyrrelser. Derfor kan CLIMAX sine resultater forbedre bærekraftig bruk og vern av marine økosystemer i Norge og andre steder.

Cumulative impacts refer to the combined effects of multiple stressors on an ecosystem, e.g., climate change, pollution, and fishing. They are one of the biggest threats to ecosystems and biodiversity, but understanding them better remains a grand challenge in marine ecology. Modeling is a crucial component of tackling this challenge. However, current modeling approaches contradict empirical studies, suffer from data limitations, or ignore the statistical possibilities arising when ecosystem simulations are run many times. New modeling and analysis approaches are therefore needed. CLIMAX aims to 1) make a leap in the modeling of cumulative impacts and 2) exploit this methodological progress to understand how cumulative impacts emerge at the ecosystem scale.

CLIMAX’s scientific potential comes from a novel combination of leading-edge computational methods and a strong foundation in the empirical literature. This integration of methods is based on three ideas. First, ecosystem models (here, Ecospace) can simulate how stressor effects identified in empirical studies might ripple through the food web. Second, ecosystem models can generate big data about the simulated ecosystem in computational experiments (running the model many times with changing inputs). Third, such big data allow for unprecedented statistical analyses of how ecosystem-scale changes might emerge in the model system. CLIMAX will develop these methods, make them available in free software, and show their scientific potential by addressing two critical research questions: 1) How might stressor interactions at the ecosystem scale emerge from known effects on individual species? 2) How do chronic stressors affect ecological resilience to short-term, extreme disturbances like marine heatwaves?

By combining the best computational methods and harnessing the unprecedented computing power available today, CLIMAX has the potential to set an ambitious agenda for modeling cumulative impacts for decades to come.

Understanding cumulative human impacts in a marine ecosystem with computational experiments

Over hele Europa er det tydelig at dagens jordbrukspraksis ikke lenger er bærekraftig, og at jordbrukssystemene må endres for å opprettholde matproduksjonen og samtidig ivareta det biologiske mangfoldet, økosystemene og de mange fordelene de gir samfunnet.

En løsning på dette er agroøkologi. Agroøkologisk jordbruk har til felles å utnytte naturlige økologiske prosesser for å gjøre jordbruk mer bærekraftig. Dessverre, til tross for mye forskning på agroøkologisk jordbrukspraksis, er bruken av dette svært begrenset i Europa.

I SAFER-prosjektet vil et stort team av europeiske forskere jobbe på tvers av et nettverk med seks 'levende laboratorier', det vil si at gårder i drift på tvers av Europa vil iverksette agroøkologisk praksis. Ved samarbeid på de involverte gårdene, vil bønder, forskere og andre interessenter sammen bidra til økt kunnskap om agroøkologi. Det vil gi mer kunnskap om de landbruksmessige, miljømessige og sosialøkologiske fordelene, samt de mulige avveiningene mellom ulike fordeler.  

Teamet fra Universitetet i Bergen, ledet av prof. Peter Manning, vil være en viktig bidragsyter i prosjektet. De skal oppskalere data fra prosjektet på felt- og gårdsnivå til hele landskap, og simulere endringer i gårdspraksis ved å bruke en kombinasjon av samfunnsvitenskap, fjernmåling og matematisk modellering. Dette vil gi prognoser for hvordan konsekvenser av landsbruksstrategier på landskapsnivå påvirker hele systemet, fra bønder til lokale interessenter. 

Sluttproduktet vil være detaljert kunnskap om hvordan ulike økologiske praksiser påvirker bønder, bygdesamfunn og om hvilke strategier som øker bærekraftig produksjon og flerfunksjonalitet i europeiske jordbrukslandskap. Ved aktiv involvering av bønder og andre interessenter gjennom hele forskningsprosessen, tar vi også sikte på å øke relevansen og bruken av de identifiserte løsningene.

Agroecology offers an opportunity to reconcile agricultural production with human well-being across Europe. However, despite significant evidence that agroecological farming reduces the environmental footprint of agriculture, agroecology is struggling to expand across Europe. This limited adoption emerges from a limited understanding of the cross-disciplinary impacts (agronomic, ecological, economic and social dimensions) of upscaling agroecological farming and recent social unrest following the deployment of agroecological policies indicates that there is an urgent need to address this issue. In addition, a major challenge for agroecology is that no single agroecological practice is suitable for all contexts. Instead, practices need to be targeted and adapted to local socio-ecological contexts, with a strong involvement of stakeholders. Addressing such challenge implies a paradigm shift towards co-design in research and innovation in agroecology.

SAFER will address these challenges by mobilizing a European network of 6 Living Labs distributed across contrasting regions and social-ecological systems, in a multidisciplinary approach. We will: (i) co-design best suited agroecological practices adapted to local social-ecological conditions via a participatory approach that actively involves stakeholders; (ii) quantify the multiple agronomical, environmental and socio-economic benefits and risks of scaling up these agroecological practices from the field to the landscape scale using a similar landscape-scale design across Living Labs; (iii) forecast the whole-system consequences of landscape-scale strategies for farmers and local stakeholders; (iv) identify how local policies, stakeholder demands and the Living Lab methodology can enhance or limit the expansion of agroecological practices

Work at UiB will concentrate on modelling work in (ii) and (iii) but will contribute to all aspects.

AGROECOL 1 SAFER Strengthening Agricultural landscape multiFunctionality through expansion of agroecological farming in EuRope (SAFER)

Dette prosjektet har som mål å undersøke nervedannelse i primær brystkreft og hjernemetastaser fra brystkreft. Vår hypotese er at en undergruppe av aggressive brystkreftsvulster involverer nervestrukturer for å fremme tumorprogresjon, inkludert metastaser til hjernevev, som er den mest nevrogene nisjen i kroppen.

Hos kvinner er brystkreft den hyppigste kreftsykdommen og den ledende årsaken til kreftrelaterte dødsfall verden over. Trippel-negativ eller basaloid brystkreft er de mest utfordrende subgruppene, på grunn av mangel på effektive behandlingsmål. Studier av nervefiber-tetthet i brystkreft har vist at høyt innerverte svulster er mer aggressive, og tilstedeværelse av nerve-elementer er assosiert med økt metastasering og forverret pasientprognose. Våre foreløpige data, fra vevsanalyse av to brystkreftserier, bekrefter disse funnene. Vi anser derfor nerveinvolvering som en ny og lovende biomarkør i vår forståelse av aggressiv brystkreft.

Nerver er små fiber-strukturer i svulster, som nåler i en høystakk, og vanskelig å oppdage ved vanlig mikroskopisk undersøkelse. Derfor eksisterer det ikke automatiserte prosedyrer for deteksjon av slike svulst-nerver, og nervene sin klinisk-patologiske betydning er lite kjent. I dette prosjektet vil vi utvikle mer sensitive metoder for å påvise små nerveelementer i vev fra svulster, ved hjelp av avansert bildeteknologi. Interaksjon mellom kreftceller og nerver vil bli analysert i funksjonelle studier.

Vår hypotese er at aggressive brystkreftceller utrykker hittil ukjente markører som kontrollerer tiltrekning av nerver til primærsvulsten, og at denne tiltrekningen forårsaker endringer i en undergruppe av brystkreftceller som fremmer spredning til hjernen.

Recent data indicate that nerves might play an active role in the initiation and progress of cancer. In the breast, research by us and others has demonstrated that enhanced innervation is associated with more aggressive tumors, such as the triple negative subtype (TNBC) supporting the formation of a neurogenic niche in primary tumors. However, the detailed clinical relevance of this phenomenon, and the mechanisms by which nerves are attracted to breast tumors and how breast cancer-nerve interaction promotes aggressive tumor features, remain unexplored. Moreover, we lack good experimental models for breast cancer innervation in a human setting. 

TNBCs with a basal-like phenotype are also prone to metastasize to the brain, the most neurogenic niche in the body, but the preference for the brain as a metastatic site is not understood, neither are the phenotypic alterations that breast cancer cells undergo in brain tissue. Here, we launch a new concept - epithelial-to-neural transition (ENT) – linking cancer cell plasticity and nerve involvement in primary and brain metastatic breast cancer. We suggest that aggressive breast cancer cells express yet unknown markers that control attraction of nerves at the primary site, and that some breast cancer cells even transform by acquiring neural-like properties and spread to the brain. Epithelial-to-neural transition will be investigated both clinically and experimentally in primary and brain metastatic breast cancer by combining large clinical cohorts, spatial omics technology, and stem cell methods to establish cutting-edge models. 

This project has the potential to change the way we view neural elements in breast cancer and provide the field with new clinical relevance, models and technologies to study them.

When breast cancer hits a nerve
- Neural involvement as a hallmark of tumor progression

Økonomisk politikk i vestlige land inkluderer i økende grad tiltak for å redusere avhengighet av Kina og mer generelt et fokus på å styrke såkalt "økonomisk sikkerhet".  Denne endringen drives av bekymringer for at økonomisk gjensidig avhengighet kan brukes som maktmiddel. Som et resultat implementerer vestlige regjeringer politikk som eksportkontroller og investeringsscreening, eksemplifisert ved USAs Inflation Reduction Act og EUs europeiske økonomiske sikkerhetsstrategi. Slike tiltak for såkalt "de-risking" plasserer i stor grad ansvaret for risikovurdering på private selskaper, som må navigere motstridende press fra statlige reguleringer og aksjonærs forventninger. Vi vet lite om hvordan selskaper forholder seg til og vurderer statlige signaler om behovet for "de-risking" og hva som påvirker deres strategier og atferd. DERISK vil tilby en systematisk analyse av hvordan selskaper i USA og Europa navigerer innenfor et nytt regulatorisk rammeverk fokusert på "de-risking." Analysen vil fokusere på hvordan selskaper vurderer denne type risiko, hvordan de håndterer og søker å påvirke statlig politikk, samt hvordan forholdet mellom stat og marked former selskapers atferd.

The global economic landscape is undergoing a paradigm shift, with Western ideas transitioning from a belief in the virtues of economic integration to concerns over economic interdependence as a tool for power and coercion. This shift marks a move away from the neoliberal global order towards a new era of "unpeace," where economic dependencies are seen as vulnerabilities. Western governments are responding by implementing measures like export controls, investment screenings, and national industrial policies, as evident in the US Inflation Reduction Act and the EU's European Economic Security Strategy. This change has led to the rise of the "Economic Security State," characterized by a political impetus to de-risk imposed by states, but where the actual burden of de-risking primarily falls on private firms, impacting their supply chains and market strategies. DERISK studies how firms respond to these governmental directives and their influence on state-market relations. The project will analyze variations in de-risking regimes and corporate responses, examining the interplay between government policies and corporate strategies. It will do so both through a broad  mapping (WP1), as well as in-depth case studies in communications, energy, and insurance (WP 2 & 3). WP 1 of DERISK involves systematic mapping of de-risking regimes in the US and Europe, focusing on variations in government and corporate articulations of de-risking. WP 2 analyzes corporate responses to these regimes, using interest alignment as a key determinant of their strategies, ranging from compliance to resistance. WP 3 explores how firms shape these regimes, and how different forms of state-market relations result in different mechanisms for influence. The project uses mixed-methods, including content analysis and interviews, to understand the dynamics of de-risking in state-market relations.

DERISK: How firms navigate de-risking regimes

Kunstig intelligens (KI) forandrer økonomien i rekordfart, men vi vet fortsatt ikke helt hvilke konsekvenser det vil få. Vil KI gi oss mer innovasjon, høyere produktivitet og økonomisk vekst? Eller vil det føre til færre jobber, økende ulikhet og mer makt til store selskaper? Politikken vil spille en avgjørende rolle i å styre utviklingen. Myndigheter står overfor et viktig spørsmål: hvordan kan vi sikre at KI gir flest mulig fordeler uten å skape store samfunnsproblemer? Skal politikken først og fremst fremme økonomisk vekst, eller bør den fokusere på å redusere ulikhet og risiko? Å finne riktig balanse er avgjørende, men vi har fortsatt ikke klare svar på hvordan dette kan gjøres best mulig. Dette prosjektet søker å gi svar ved å bruke en ny tilnærming. I stedet for å basere politikk på teori alene, bør vi lære av tidligere erfaringer. Ved å analysere hvordan tidligere KI-relaterte tiltak har påvirket bedrifter og samfunnet, kan vi utforme smartere og mer treffsikre løsninger. Målet er å utvikle politikk som gir størst mulig nytte for samfunnet – både nå og i fremtiden.

There is currently a vivid societal debate and nascent research about the economic effects of artificial intelligence (AI). Some expect to see positive impacts in terms of innovation, productivity and economic growth. Others point to possible economic harms such as technological unemployment, increasing market concentration and exacerbation of income inequalities. 

Public policies will have a crucial role to shape the direction and extent of these effects. However, it is still quite unclear what policies could do to foster the future economic benefits of AI, what they could do to avoid its possible harms, and how they could balance the positive and negative impacts. 

The present project will investigate the trade-off between growth and inequality effects of AI, and analyze how public policies for AI should be designed in order to take this trade-off into account. 

The key novel aspect of the project will be the application of the optimal policy learning (OPL) approach to the study of policies for AI. The main idea of this approach is that optimal policies should be designed by exploiting information about the effects of previous policies on the treated agents, in such a way that new policies will seek to maximize ex-ante the empirical welfare of the target population. In other words, the design of new policies is optimal because it is based upon, and it learns from, the effects of previous policies and the heterogenous characteristics of agents. 

To apply this approach to the study AI policies, the project will develop the following pillars: (1) a conceptual framework of firm heterogeneity and AI, which points out the main factors that affect firms’ adoption of AI and its effects; (2) empirical analysis of how AI and related policies affect different types of firms; (3) analysis of how different policies should be designed in order to maximize empirical welfare, and how these policies can address the trade-off between efficiency and equity effects of AI.

Artificial Intelligence: Growth, Inequalities and Public Policies

Antibiotikaresistens er i ferd med å bli en alvorlig global helsetrussel. Etter hvert som bakterier utvikler motstand mot dagens medisiner, blir infeksjoner som tidligere var enkle å behandle, igjen livstruende. 
Verdens helseorganisasjon (WHO) har laget en liste over farlige bakterier hvor behovet for nye antibiotika er akutt. Dessverre er de fleste antibiotika som utvikles i dag bare små endringer av gamle medisiner – og bakteriene lærer raskt å stå imot disse også.

For å løse dette problemet må vi finne helt nye måter å angripe bakteriene på. Det betyr at vi må oppdage nye angrepsmål i bakteriene og utvikle antibiotika som kan nå frem til og slå ut disse.

Dette prosjektet har som mål å gjøre nettopp det. Et internasjonalt team av forskere samarbeider for å:

Undersøke to spesifikke bakterieenzymer for å se om de kan være gode angrepsmål for fremtidige antibiotika.
Bidra til å øke vår forståelse av hvordan antibiotiske legemidler kommer seg inn i bakteier, noe som er en av de største utfordringene i utviklingen av nye antibiotika.
Vi fokuserer på tre typer skadelige bakterier: Pseudomonas aeruginosa, Escherichia coli og Klebsiella pneumoniae. Disse er alle såkalte «Gram-negative» bakterier, som er spesielt vanskelige å behandle på grunn av deres oppbygning.

I prosjektet vil vi studere to viktige enzymer som disse bakteriene bruker til å lage fettsyrer – molekyler de trenger for å overleve. Ved bruk av ulike teknikker skal vi designe og teste nye inhibitorer som kan blokkere disse enzymene. Samtidig skal vi undersøke hvordan disse inhibitorene kommer seg inn i bakteriene, slik at vi er sikre på at de faktisk når frem til målet sitt.

Ved å kombinere kjemi, biologi og legemiddeldesign vil dette prosjektet ikke bare hjelpe oss med å komme videre på veien til å finne nye antibiotika – det vil også gi verdifull innsikt i hvordan vi kan utvikle bedre antibiotika i fremtiden.

There is an urgent need for new antibiotics to prevent the looming crisis of antimicrobial resistance and WHO has published a list of bacteria for which antibiotics are in high demand and urged that research and drug discovery efforts are directed toward these. Antibiotics are a prerequisite for modern medicine, however, most of the compounds in clinical development are not fit for purpose at they are derivatives of already known antibiotics and therefore have cross-resistance with existing agents.
To tackle this crisis, we need fundamental knowledge on new targets for antibiotics, and we need compounds that can get into bacteria and work on these targets. This project will address this 1) by delivering potent inhibitors on two proposed bacterial protein targets to establish if they are promising targets for future antibiotics and 2) by establishing relationships between compound properties and uptake in bacteria. To reach these highly ambitious goals, we have assembled an interdisciplinary and international team of experts in antibiotic drug discovery, protein dynamics, structure-based drug design, covalent enzyme inhibition, organic synthesis and medicinal chemistry. We will explore two essential enzymes involved in fatty acid synthesis in Pseudomonas aeruginosa, Escherichia coli, and Klebsiella pneumoniae, Gram-negative bacteria for which WHO has stated that new antibiotics are critically needed. We will use a structure-based approach to develop potent inhibitors for chemical target validation. This process will be guided by experimental data about uptake into bacteria to ensure that our compounds reach their targets, a critical step in chemical target validation. Through this we will provide crucial information to advance our understanding of the compound properties that governs uptake in Gram-negative bacteria. This will deliver far-reaching insights, not only for the studied target but antibiotic drug discovery in general.

WebData vil tilrettelegge for forskning på data fra det norske internett, tilsvarende tjenester som Nettbiblioteket og DH-lab fra Nasjonalbiblioteket for fysisk materiale. WebData vil altså gi forskere adgang til data som ikke er tilgjengelige i en infrastruktur i dag, men som det er et stort behov for.

Nasjonalbiblioteket har hentet og lagret data fra det norske internett siden slutten av 1990-tallet og sitter på enorme mengder med data. Dette materialet har man i liten grad kunnet tilgjengeliggjøre, blant annet fordi lovverket setter strenge krav til håndtering av personopplysninger i slike arkiver. Samtidig har den offentlige samtalen for en stor del flyttet seg til nettet, slik at det er blitt viktigere å kunne forske på data fra denne delen av offentligheten. I tillegg spiller data fra nettet en særlig rolle i arbeidet med kunstig intelligens og store språkmodeller. Skal norsk og samisk overleve i den digitale tidsalderen, trenger vi så mye data som mulig av høy kvalitet for å støtte opp under språkteknologi.

WebData skal derfor bygge en plattform for forskning på materiale fra nettet i tråd med gjeldende lovverk. Det vil gis åpen tilgang til materiale publisert av det offentlige og adgang for forskningsformål til materiale utgitt med og uten ansvarlig redaktør. Vi vil ta i bruk kunnskap om og verktøy for identifisering og vasking av personopplysninger for å klassifisere materiale og bygge et system for sikker tilgang.

Plattformen skal utvikles i tett samarbeid med forskersamfunnet. Vi vil gjennomføre en behovsstudie tidlig i prosjektet for å kartlegge hvilken funksjonalitet forskerne trenger (f.eks. visualisering av ordbruk over tid, uttrekk og analyse av hendelser) som igjen vil legge føringer for tilrettelegging av data. Prosjektet har som mål å styrke samiske språks representasjon. Vi vil gjennomføre en representasjonsstudie for å undersøke hvor godt samisk er dekket i nettarkivet og gjøre tiltak for å forbedre høstingen av samiskspråklig innhold.

The National Library of Norway, a major cultural heritage institution in Norway, joins forces with some of the most prominent research communities for language technology in Norway to create WebData, a national research infrastructure for web data. WebData will offer researchers access to the Norwegian Web Archive, hosted on-premises at the National Library of Norway. The infrastructure will first and foremost consist of a data platform featuring a general purpose search interface for web data from the Norwegian web (the .no top-level domain) from the last 25 years, allowing researchers to search and explore web pages, documents, transcribed audio/video and images.

The platform will implement a model of layered access, using automatic categorization and identification of personal information to open parts of the collection that would otherwise be closed due to regulatory policies, which is a major R&D challenge and one of the main reasons why the material is not available today. The web archive will further be scaled up according to the needs of researchers and underrepresented communities, e.g. allowing for quantitative analysis of web data and by increasing the coverage of Sámi web content. The infrastructure will contribute to research on Norwegian and Sámi language and culture and produce language resources, e.g. corpora for large language models, that help prevent domain-loss of these languages.

The project is highly interdisciplinary and aims primarily at researchers in the social sciences and humanities, but will also be relevant for e.g. computer science, medicine and law.

The Sometimes Projector

Within Energi 21 (a unifying R&D strategy for the energy sector) five technology target areas are identified in order to respond to the key challenges and to the national objectives. In the strategy document for Energi 21, one of the target areas for more  intensive basic research is an energy system for the need of the future. The main priorities within this target area are related to cabling from offshore wind mill farms either to mainland Norway or to other countries by subsea cables. 
 
Reliable subsea  powerAC and DC cables are of crucial importance if an offshore transmission network shall be established. Dynamic cables installed on deep waters are not a proven technology; this will be important components for offshore wind farms in the future. This c alls for technology improvements, where the knowledge about limitations for the long term performance of high voltage power cables are important design criteria.
The research within this Renergi KMB project will have a generic approach in order to provide  knowledge for technical improvements and consequently long and reliable service performance of high voltage subsea AC and DC cable systems. 

Five main research tasks are defined within the project:

T1: Impact of dynamic mechanical loads and hydrostatic  pressure  on AC polymeric cable 	systems
T2: Long term properties of subsea DC cable systems 
T3: Development of sensitive Qualification Test Methods for very long AC and DC subsea polymeric cables 
T4: Development of test facilities for combined dynamic  / electric testing 
T5: Develop methods for condition assessment of long subsea AC and DC polymeric cables 

There will be one Ph D student and one Post Doc at NTNU working with selected activities within Task T1 or T2.
