Proteome ecotoxicology - a new strategy for developing biomarker tests for environmental monitoring, Postdr.stip.og utenlstip. til Danmark

Prosjektsammendrag: Inducible genes make attractive candidates for biomarkers, i.e. molecular and cellular responses to chemical exposure indicating adverse effects to organisms. The proteome represents the functional genome of a cell, i.e. that part of the genome expressed as proteins under a certain set of conditions. The proteome strategy is based on 2-dimensional gel electrophoresis and identification of protein spots by microsequencing using mass spectrometry and Edman degradation. By utilizing this strategy in combinat ion with invivo and in vitro studies where fish and invertebrates (and cell cultures) are exposed to different classes of environmental pollutants, it is possible to screen and identify a large number of pollutant-inducible proteins that will make good ca ndidates as biomarkers for marine environmental monitoring. The aim of this project is to identify 10-15 such proteins from fish and invertebrates, to develop specific antibodies for these proteins, and to investigate their usefulness in more extensive ecotoxicological test.
