Pigment and beta-hemolysin activity of the genus Propionibacterium

Propionibacteria are used for production of Swiss-type cheeses and are in addition used for production of nutraceuticals and as probiotics. Propionibacterium thoenii and Propionibacterium jensenii contain an unknown red pigment and in addition beta-hemoly tic activity (Food safety and Health). These species belong to the group "Dairy propionibacteria", and some non-pigmented species are used for Swiss Cheese (Jarlsberg) production. Non-pigmented mutants of P. thoenii have in addition lost their beta-hemoly tic activity. Pigment(s) will be determined by traditional extraction methods of the mother culture and orange and yellow mutants, followed by different chromatographic methods. Since carotenoids often exist in complex with proteins, one will use extracti on methods with Triton and/or lysozyme for releas of the complex. Molecular studies of the genes for pigment formation and beta-hemolysin will be performed by molecular biological techniques, for instance PCR-techniques or by using transposition vector which have been used for pigment-beta-hemolysin studies on Streptoco ccus agalactiae. In addition,, biochemical methods will be used. One will determine if the loss by mutation is due to a common operon system on the chromosome or is due to transposon mutation. Probes will be made to search for dormant genes in the non-pig mented species
