Diet, vitamin A and K, genetics and orofacial clefts

Several factors have been shown to promote orofacial clefting eg genetic factors, teratogenes, maternal alcohol and cigarette use, herbicides and nutrient intake like folate and other B-vitamins. Our primary aim is to study the association between materna l dietary intakes of the first three months of pregnancy and the risk of orofacial clefts in the offspring. This proposal builds on a unique data set including 574 Norwegian case-parents and 763 randomly selected control-parents that have been collected t hrough a population-based case-control study of Norwegian babies born in the period 1996-2001. We already have maternal diet, pregnancy outcomes as well as DNA from infant, mother and father. The availability of these data makes it possible perform analys es of the interaction between nutrients, foods, genetic polymorphisms and clefts. We will also measure the amount of vitamin K1/2 in some Norwegian foods to get a more accurate estimation of the dietary intake among the pregnant women. Studies using from 1 to 40 families and candidate genes suggest loci for clefts on chromosomes 1, 4, 6, 17 and 19. We will assay relevant genes and analyse those genes in combination with the dietary factors. The study is approved by the Norwegian Regional Ethics Committee of Region West and by the Data Inspectorate. Data for age, height, pre-pregnancy weight, smoking habits and detailed information about use of dietary supplements among the mothers and, data concerning the babies with cleft lip and or palate, are all avail able from in the database. Sputum samples were collected from parents as well as the infants. Genomic DNA will be extracted and analyzed for polymorphisms. The study is by far the largest and most complete study of diet, metabolism and risk of oral cleft worldwide. Our results may have important implications both for research on birth defects etiology and for nutritional recommendations for pregnant women.
