The maintenance of stem cells and the regulation of their differentiation are still poorly understood. Interstitial stem cells are easily accessible in the simple animal Hydra and give rise to different neuronal cell fates, gametes and gland cells. In con trast, no interstitial stem cells have been identified from the basal cnidarian Nematostella. We have identified several conserved transcription factors that mark stem cells in Hydra and the different phases of neuronal differentiation. The Nematostella h omologs show very similar expression patterns, suggesting that these genes may play a conserved role in stem cell differentiation. We intend to test the function of these genes during embryogenesis of Nematostella by Morpholino-based gene knock-downs.
