USA - Kill the messenger strategies in human pathogenic bacteria using RNase P, a molecular tool with an innovative therapeutic potential

"Kill the messenger"-strategies is a new innovative tool in functional genomic research of both bacteria and mammalian cells. This approach also holds promise for the development of a new class of therapeutics using RNA-molecules to silence unwanted gene expression in vivo. In eukaryotic cells discovery of RNA interference, awarded the Nobel Prize in Medicine 2006, has provided a powerful new experimental tool to study the function of genes in eukaryotic cells and raises expectations about future applicat ions of RNA interference in medicine. Specific down-regulation of gene expression through RNase P-mediated mRNA cleavage may provide the equivalent tool for prokaryotic cells. We will explore and use the capabilities of the endogenous, essential RNase P t o target, cleave and inactivate specific mRNAs in our research efforts to understand the evolution, spread and persistence of pathogenic lineages of antimicrobial resistant bacteria (enterococci). BILAT funding will allow us to perform in vitro studies of enterococcal RNase P in Dr. Altmans lab at Yale University and thus give us information about activity and substrate specificity of enterococcal RNase P necessary to develop RNase P as a gene knockdown tool in enterococci.
