Single Cell Profiling of Phospho-Protein Networks in Health and Disease - From Signalling Profiles to Future Biomarkers

This proposal describes plans for a research project to be undertaken by a strong concortium of PI's (Gjertsen, Lorens, UoB; Torgersen, Aandahl, Tasken, UoO) and aiming to characterize key events in intracellular signalling networks that regulate cellular properties and behaviour in health and disease at a single-cell level. Analyses at the single-cell level will allow examination of signalling in mixed cell population where different cell types interact with each other or respond (differently) in paralle l to a stimulus. Systems biology principles will be applied to address the function of unique reponse-dependent signalling epitopes hereunder, phospho-protein networks, associated with disease in order to understand pathogenesis and disease progression in cancer and HIV. The consortium will work to also develop and incorporate analysis of other signalling epitopes to monitor responses and develop bioprofiles of disease. Intracellular signalling networks control cellular processes through a complex series of protein-protein interactions, post-translational modifications, protein translocations and protein cleavage events. Phospho-specific flow cytometry, proteomics and bioinformatics will be applied for data acquisition and analysis. We expect to unravel k ey molecular events in the pathogenesis, new drug targets and develop new therapeutic strategies. The applicants employ advanced cell biological and biochemical methods, and have significant expertise in the fields of functional genomics, cellular signal ling and clinical immunology and haematology. The group employs methods that have been established as platform technologies in the national FUGE programme to identify and study the function of new genes implicated in specific pathways and networks. Our gr oup has ongoing collaboration with excellent national and international collaborators that will serve the project we are developing in terms of technology transfer, transfer of material and exchange of researchers.
