Proposal for the purchase of an ESI mass spectrometer for the characterization of peptide, nucleic acid and carbohydrate modificatications

The goal of the project is to purchase a new high end elelctrospray MS instrument which would allow the high end analysis and characterization of modified peptides, modified oligosaccharides and modified nucleic acids. W. Egge-Jacobsen and M. Koomey are w orking on unique post-translational modifications of the Neisseria gonorrhoeae Type IV pilus colonization factor. Their goals are to identify their importance to the microbe, microbe-host interactions and the pathogenesis of gonococcal disease. In additio n, they have recently discovered that the O-linked glycosylation pathway targets a number of other proteins in addition to the pilin subunit protein. This is the first example of general O-linked glycosylation system in prokaryotics. Therefore in order to keep the position in the leading field of bacterial glycoproteomics, it would be necessary to have appropriate instrumentation for in house analyses.In addition, other groups at IMBV working on the modification of carbohydrates (K. Prydz) and nucleic aci ds (P. Falnes) like to use the MS expertise and equipment for their research fields. The Falnes group focuses on various demethylases involved in removing lesions and modifications from DNA and RNA, and the Prydz group studies modification of glycoprotein s and proteoglycans by sugars is general and particularly their sulfation by sulfotransferases. The differences between the new proposed instrument and the excisting MS at IMBV or even at UiO are the ability to performe - A. ETD fragmentation to character ize post-translationel modifications (especially glycopeptides) - B. mass determination with excellent accuracy of 1-3 ppm instead of 100 ppm on a 3D ion trap - C. analyses with 15 x times higher ion capacity and 3 x times higher scan rates. - D. intact protein MS/MS fragmentation allowing direct localization of protein modifications
