Method for identification of 5-hydroxymethylcytosine and 5-methylcytosine at single base resolution

Cytosine DNA methylation at the N5 position (5meC) is a well described modification affecting gene expression in mammalian cells. Aberrations in 5meC patterning can lead to tumorigenesis, developmental defects and many cellular anomalies. 5-hydroxymethylc ytosine (5hmC) DNA modification is a more recently described gene-regulatory modification. Aberrant, 5hmC levels and genomic patterning has been strongly correlated with cancer formation, aggressiveness and invasiveness. Methods for the identification of 5hmC patterning are necessary to determine the 5hmC patterns relevant in specific tumors and to furthermore provide predictive information about the prognosis for specific tumors. We have developed a method (HyLo method) that will allow for the differenti ation of 5hmC, 5meC and cytosine at base specific resolution. The optimization of this method will allow for the production of saleable products and the generation of intellectual property in both research markets and clinical markets. The three expected intellectual properties that will arise at completion of this project are: i) a methodology to differentiate 5hmC, 5meC and cytosine at base specific resolution at single genes, ii) in whole genomes and iii) the identification of tumor specific 5hmC patte rns that can be used to provide prognostic and diagnostic information about tumors.
