Assessment of Endocrine effects of POPs and heavy metals on an Arctic char population in the Arctic Lake Diesetvatnet RiS-ID: 11617

This AFG project (RIS-ID 11617) involves sampling activity for my master thesis dealing with endocrine disruptive effects on Arctic char. Among the pollutants that will be investigated are a selection of persistent organic pollutants (POPs) and some heavy metals such as mercury (Hg), arsenic (As), lead (Pb) and cadmium (Cd). Siloxanes will also be analyzed if additional funding is available. Field work will include catching (by fishing rod) 10 - 20 species of Arctic char from Diesetvatnet at Mitrahalvøya. To lower the risk of not reach the needed number of fish, we will collaborate with staff at KingsBay and Norwegian Polar Institute as well. Blood samples needs to be taken immediately after catching and will therefore be the most critical part. Our goal is to get blood samples from minimum 10 fish. Some sample material will be frozen and brought back to NTNU for further analyses, while some analyses will be done as fast as possible at UNIS in Longyearbyen. Total amount of mercury, arsenic, cadmium, lead and other trace elements will be determined by ICP-MS, and organic pollutants will be analyzed by GC-MS. Levels of Estradiol and Testosterone free concentration as well as VTG concentration will be determined by ELISA approach using an UV-Vis Spectroscopy, whereas Serotonin, Cortisol and Dopamine levels in blood will be analyzed by LC-MS. Additionally, gonadosomatic index of the Arctic char will be calculated. Standard abiotic parameters including pH, temperature, conductivity, redox potential, turbidity, and trace elements in the water samples from lake Diesetvatnet will be analyzed as well as environmental conditions. All instrumentation is available at Department of Chemistry at NTNU / UNIS and through collaboration with Akvaplan-NIVA. Trained personnel from Akvaplan-NIVA will join the fieldwork for helping with correct sampling protocol for fish blood.
