THE ROLE OF HIPPOCAMPAL CAJAL-RETZIUS CELLS IN THE MATURATION OF THE HIPPOCAMPAL REGION

Vårt ultimate mål er å forstå hvilke roller ulike celletyper har i dannelsen av kortikale nettverk i fysiologiske samt patofysiologiske forhold. Endringer i mekanismene som styrer differensiering og migrasjon av nevroner eller synaptogenese kan medføre alvorlige nettverksforstyrrelser og svekkede utviklingsforstyrrelser. Prosjektet fokuserer på å forstå Cajal-Retzius celler sine roller i utviklingen av hippocampus-regionen. Cajal-Retzius celler ble først oppdaget mot slutten av 1800-tallet av en av nevrovitenskapens grunnleggere – Ramon y Cajal – men egenskapene til cellene er fremdeles et mysterium. Cajal-Retzius cellene er avgjørende for riktig dannelse av cortex under prenatal utvikling, men de fleste av disse cellene dør like etter fødselen. Imidlertid overlever disse cellene i lang tid i en bestemt del av hjernen, hippocampus (et sentralt område for læring, hukommelse og navigasjon). For eksempel, hos en mus vil slike celler overleve i minst 3-5 måneder etter fødselen. Spørsmålet er: hvorfor overlever disse cellene kun i dette området? Vi spekulerer at cellene spiller en viktig rolle i dannelsen og modningen av de nevrale nettverkene som er grunnleggende for hukommelse og navigasjon. Slike funksjoner utvikles hos mus, så vel som hos mennesker, først etter fødselen, hvilket ofte krever flere måneder eller år. Med dette stipendet vil vi kunne benytte ulike komplementære tilnærminger – alt fra å spore nevrologiske forbindelser på tvers av hjerneområder, til in vivo eksperimenter av nevral aktivitet til analyse av genuttrykk – for å forstå hva som skjer dersom disse cellene fjernes på kunstig vis kort tid etter at dyret blir født. Hvordan endres nettverket? Hvordan endres genuttrykket? Er dyret fortsatt i stand til å lære enkle oppgaver? Ved å få svar på disse spørsmålene kan vi få en bedre forståelse av hvordan nevrale nettverk er etablerte både i fysiologiske samt patologiske forhold.

The hippocampal region plays a major role in the encoding, association, consolidation and retrieval of memories. It creates spatial maps that animals use to navigate the environment. The circuits for these features are refined around birth as well as during the first weeks of postnatal development, in rodents, and years, in humans. The mechanisms guiding this maturation are activity-regulated, and disruptions in this activity are detrimental for the proper functioning of the network. Defining the development of the hippocampal circuitry during this fundamental period of maturation is critical to understand the network function in both normal development and in neurodevelopmental disorders. Studies on neocortical development have showed that specific cell types and specific circuits are critical for this early period of network maturation, and that their perturbation leads to long-lasting deficits. However, an in-depth analysis of the development of the hippocampal circuit formation is still lacking. Indeed, a specific cell type characterizing the hippocampus during postnatal development, Cajal-Retzius cells, has received little attention. Our data show that ablation of these cells at early postnatal stages affects the maturation of the hippocampus, with defects in protein expression levels, synaptic physiology, spine morphology and place cells function. To properly comprehend the role of CR cells in the circuit of the hippocampal region , much about these cells has yet to be revealed. Our aim is to study the integration of Cajal-Retzius cells in the circuit of the hippocampal region and understand how they contribute to its maturation. We propose a collaborative, ambitious and interdisciplinary approach combining state-of-the-art anatomical, physiological and molecular techniques, aiming to reveal the importance of these cells in physiological and pathophysiological development.
