From the SEA to the SOLution. An integrative assessment of marine pollution in a changing Arctic

Marine chemical pollution poses known threats to human health and to biodiversity, ecosystem and societal services. The Arctic is a major sink of global mercury (Hg) pollution. Arctic sediments, sea and glacial ice constitute vast Hg reservoirs. Recent studies raise concerns about their increasing role as local Hg sources. In a rapidly warming Arctic, the consequences of potentially increased environmental Hg on marine biota remain unknown. The objective of SEASOL is to identify the pathway(s) of Hg bioaccumulation in marine food webs of the High Arctic Kongsfjorden (Svalbard). This changing glacial fjord is recognised as a ‘natural laboratory’ of climate change in the Arctic. The fellow’s earlier research has shown the sole measurement of Hg levels as insufficient to connect biotic Hg dynamics to environmental change, whereas the analysis of Hg stable isotopes proved to be a promising tool for source discrimination and biotic transfer. SEASOL will build on the novel application of stable isotope analysis to identify the sources of Hg within the fjord system and the pathways to Hg accumulation within sentinel marine species. On the short-term, the study will provide fundamental knowledge to help understand how emerging sources of Hg from rapid environmental change will impact Arctic marine predators. On the long-term, SEASOL will contribute to the development of early-warning, adaptive management strategies, including modelling, for mitigating the cumulative impact of pollutants and climate change on Arctic marine species. This multidisciplinary approach and the central position of the host institution (Norwegian Polar Institute) in the field of Polar Research will equip the Fellow with a broad set of new skills and lay the foundation for building her specific niche as a future leader in marine pollution science.

Vi vil arrangere Havforskarmøte 2024 i Tromsø. Norsk havforsking står sterkt internasjonalt, men det er få nasjonale arenaar der havforskarar i Noreg kan samlast for å dele forsking og bygge nettverk. Havforskarmøtet er derfor ein viktig møteplass. Dei ca. 40 - 100 deltakarane kjem frå ulike sektorar og representerer marine fagmiljø frå heile landet. Det er eit ope arrangement, med påmelding via nettsidene til NHF. 
Havforskarmøtet blir arrangert årleg, og kvart år er fokus på eit spesifikt tema. Eksempel på tema frå dei seinaste åra har vore: «Hvordan få mer mat fra havet» (2017), "Hvordan kan forskere gjøre økologi mer politisk relevant?" (2019), "Klimaendring i havet" (2020, online), "Nye metodar for å møte morgondagens utfordringar" (2021), «Bruk eller vern – arealbruk til havs» (2022) og «Mat frå havet» (2023) - sjekk https://www.havforsk.no/nb/arkiv/ 
I 2024 blir temaet “Effektar av klimaendringar og klimatiltak på marin natur”, kor vi ønskjer ein diskusjon både om direkte effektar av klimaendringane på enkeltartar og marine økosystem, men og korleis dei tiltaka vi iverksett for å motverke klimaendringane kan påverke marin natur. Eksempel på slike tiltak være naturbaserte løysingar i sjø, bygging av havvindmølleparkar for grøn energiproduksjon, eller gruvedrift i fjord og hav for å sikre mineral nødvendig for ei grøn omstilling. Vi ønskjer å ha ei tverrfagleg vinkling til forskingsspørsmåla, og vil gjere møtet til ein møteplass mellom akademia, forvaltning og industriaktørar. Dei største effektane av klimaendringane ser vi i nordlege havområde, som i Barentshavet der forskarar har observert eit skifte frå eit arktisk til eit meir atlantisk klima og økosystem dei siste tiåra. Samtidig er det i dette området fleire initiativ både knytt til gruvedrift og havvind (til dømes GoliatVind-prosjektet). Vi fant det derfor naturlig å legge årets møte til Tromsø.

Optimization of Free Space Optical Communication Link based Drones for Search and Rescue Operations in the Arctic Region

The Arctic region is witnessing more frequent shipping accidents than ever which require accurate search and rescue operations. The existing communication systems use drones with radio frequency (RF) technology, which has limited bandwidth and cannot offer 4G/5G connectivity due to harsh weather constraints. It further limits the capacity of drones and affects the resolution of images and videos. The current research proposes to design free space optical communication (FSOC) link-based drones that can enhance connectivity between multiple drones and rescue ships as well as the capacity of drones to produce high resolution images and videos. The proposed FSOC-based drones can provide highspeed connectivity than the existing RF technology. The research will collect atmospheric turbulences data from the Arctic region and use it for designing the FSOC link. Furthermore, to enhance the capacity of FSOC-based drones, the research will design advanced multiplexing schemes (Mode Division Multiplexing and Orthogonal Code Division Multiple Access) and to ensure link stability, omnidirectional receiver will be designed. The research will further evaluate the performance of proposed FSOC link under the impact of various atmospheric turbulences. The expected outcomes of this research will lead to the optimisation of FSOC-based drones in expanding search areas and improving communication systems for SAR operations in the Arctic region.

Sjøisen i Arktis har minket drastisk de siste tiårene, og Polhavet forventes å være isfritt om sommeren mot slutten av århundret. Denne nedgangen har utsatt store havområder for vind, noe som har resultert i at det oftere dannes store bølger i Polhavet. Disse bølgene kan forplante seg over flere titalls til hundrevis av kilometer i sjøisen, knekke den og akselerere smeltingen. Disse fysiske prosessene – bølger som knekker sjøis og effekten dette har på sjøissmelting og vekst – mangler i modellene vi bruker for å forutsi klimautviklingen. Dette bidrar til store usikkerheter i klimamodeller. Effekten av bølger på sjøisen er heller ikke tatt med i prognoser for havets og sjøisens forhold, som brukes av skip som opererer i islagte farvann. I ForWArd ønsker vi å vurdere bølgenes innvirkning på sjøisens utvikling i det stadig røffere Polhavet. I dette arbeidet brukes en svært avansert modell som inkluderer sjøis, hav, bølger og deres interaksjoner. Vi skal først vurdere hvor godt den representerer forholdene i Polhavet sammenlignet med de få tilgjengelige observasjonene. Vi vil så bruke modellen til å undersøke hvordan bølgehøyden og arealet av ødelagt sjøis har utviklet seg de siste tre tiårene. Deretter vil vi kombinere modellen vår med forholdene i det fremtidige Polhavet gitt av klimafremskrivninger, for å undersøke hvordan betydningen av interaksjoner mellom bølger og is vil utvikle seg ettersom sjøis avtar i det 21. århundre. Vår modells estimering av "bølgepåvirkningen" vil tillate polarvitenskapsmiljøet å vurdere hvor usikre deres klimaspådommer uten bølger er. Til syvende og sist vil våre modellresultater og utviklinger legge grunnlaget for å legge til bølgeeffekter i hav- og sjøisvarsler i Arktis. Dette vil gjøre varslene mer nøyaktige og bidra til sikkerheten for menneskelig aktivitet i denne regionen.

Arctic sea ice decline under climate change has allowed waves to form and grow more easily in the Arctic Ocean. These waves propagate into the sea ice over tens to hundreds of kilometres, fragmenting it into small floes. This area of broken ice, known as the Marginal Ice Zone (MIZ), is a highly complex region where many interactions occur between the ocean, sea ice, and atmosphere. The action of waves strongly affects these interactions, with rapid feedbacks on the sea ice. However, climate models do not account for these wave-ice interaction processes, which likely contributes to the strong biases in their estimation of modelled sea ice extent and volume and their inability to capture short-term sea ice extent variability. Given the lack of observations available, numerical models remain the best way to estimate how wave-ice interaction processes can impact Arctic sea ice, but most wave-ice coupled models are still prototypes. In ForWArd, we will use one of the most advanced of these models to assess this impact in the context of climate change. To this aim, we will use the latest observations to calibrate the modelled wave-affected area and produce a 30-year-long hindcast of the MIZ extent evolution to analyse how it responded to larger wave height, delayed refreezing, and thinner ice. Then, we will assess the relative importance of the wave-ice interactions suspected to affect sea ice in the MIZ using well-documented events. Finally, we will estimate how waves will contribute to sea ice extent and volume variability in the coming decades, as large areas of open water will remain open until late in the autumn. The flexibility of our setup will allow us to test the sensitivity of our results to model resolution and assumptions, giving invaluable insights into the biases induced by misrepresenting wave-ice interactions. ForWArd will also pave the way for the integration of wave effects in sea ice forecasts, a requirement for safer operations in ice-infested waters.

Forecasting Wave impact as Arctic sea ice declines

Blue Bio Boost tar sikte på å bidra til bærekraftig økonomisk utvikling av makroalgeindustrien ved å forbedre effektiviteten av formering og utvalg av egnede genotyper (mål 1), ved utnytte genetisk variasjon samtidig som potensialet for genetisk forurensning av naturlige populasjoner minimeres (mål 2), og ved aktivt å involvere interessenter i arbeidet med en plan for fremtidig makroalgeavl i Europa (mål 3).

Blue Bio Boost vil undersøke to alternativer for å unngå påvirkning på naturlige bestander på grunn av kryssing av dyrkede og naturlige bestander; (1) en "mix-to-match"-tilnærming der vi dyrker blandinger av genotyper som egner seg for dyrking og som har allelfrekvenser som ligner på allelfrekvensene i de lokale naturlige populasjonene, og (2) teknikker som kan bidra til avl av forbedrede genotyper og produksjon av sterile alger for dyrking i fremtiden.  En bred og representativ gruppe av interessenter vil bli etablert og delta både i utformingen av en strategi for fremtidig europeisk avl av makroalger med fokus på økonomiske og økologiske mål, og som en formidlingskanal.

Macroalgal biomass has multiple uses and is recognized as a target for increased production in Europe. In order to boost the growing seaweed industry, it is necessary to better utilise the genetic variation to increase yield, quality and stability of the cultivated crop. The current macroalgae cultivation practice in Europe starts every year from new seeding material, obtained directly from local natural populations. There is no selection of the most suitable genotypes, and no storage of seeding material. Hence, no genetic progress in terms of yield and quality is obtained in the cultivated material over time. This is in sharp contrast to virtually all land-based crop production, animal husbandry and fish aquaculture, where domestication has led to selection of genetic material with cumulative improvement of desired traits. There is, however, concern about the potential spread and mixing of domesticated macroalgae with natural populations (introgression), and the ecological consequences this may have, including erosion of the present natural genetic diversity. Blue Bio Boost will investigate two options to avoid impact on natural populations; (1) a “mix-to-match" approach where we cultivate mixtures of superior material with allele frequencies similar to those of the local natural populations, thus attempting to circumvent the problem of introgression, and (2) techniques that could facilitate breeding of improved genotypes and production of sterile sporophytes in the future. A broad and representative Stakeholder Engagement Group (SEG) will be established and function both in co-design of a strategy for the future European breeding of macroalgae focusing on economic and ecological objectives, and as a channel for dissemination activities.

SBEP - Boosting seaweed farming through better utilisation of genetic resources

Siden etterspørselen etter sjømat fortsetter å vokse, er det avgjørende at vi identifiserer regionale og bærekraftige fôrkilder som ikke bare øker næringsverdien til fiskefôr, men som også øker veksten og helsen til oppdrettsfisk og samtidig reduserer den økologiske påvirkningen av akvakultur. En lovende løsning ligger i bruken av tang-baserte produkter, som lenge har vært brukt i fôr på grunn av deres binde-egenskaper.
Hovedinnovasjonene i prosjektet inkluderer bruk av forbehandling av tang og tare for å øke utbyttet av bioaktive forbindelser fra tang, samt utvikling av et effektivt kaskadende bioraffineri for å forbedre utvinningsprosessen. Resultatet vil være miljømessig, økonomisk og sosialt bærekraftige prosesser for havbruksnæringen.
Ved å bruke tangavledede forbindelser og innovative teknikker kan vi jobbe mot en mer bærekraftig og miljøvennlig oppdrettsnæring, samtidig som vi forbedrer helsen og velferden til oppdrettsfisken.

Pursuit of a sustainable aquaculture industry must identify regional and sustainable feed sources that enhance the nutritional value of aquafeed, increase the growth and health of fish while reducing the ecological impact of aquaculture. Seaweed-derived functional compounds have long been utilised in aquafeeds, mainly focusing on their binding properties. Feed producers are now looking to substitute conventional feed ingredients, such as fishmeal and soybean meal, associated with environmental concerns. These challenges necessitate alternatives from marine resources to support the low-trophic organisms. Therefore, seaweed cultivation and harvesting can have a significant positive impact by mitigating damage to local ecosystems and marine wildlife: a prerequisite for further growth is the introduction of ecosystem services in European aquaculture.
The originality of our approach is the combination of expertise and crossover of techniques within pre-treatment, biorefinery, screening in in-vitro, and validating in in-vivo studies combined with sustainability and techno-economic assessments. The project engages early on with the industrial partners (within consortium) and other stakeholders to enable co-creation and co-production in both seaweed and aquaculture feed sectors. Our innovation lies in the choice of methodology, which includes pre-treatment to increase yield of bioactive compounds from seaweed and develop an efficient cascading biorefinery to enhance the extraction process. The outcome of this project will therefore be beyond the state-of-the-art to provide environmentally, economically, and socially sustainable processes.

SBEP - Aquafeed Upgraded: Enhancing Immune Function of Fish with Seaweed-modified Functional Compounds

I 2017 fikk EU-kommisjonen gode råd fra en gruppe vitenskapelige rådgivere om hvordan høste mat og biomasse fra havet uten å skade fremtidige generasjoner. Rapporten fremhever bærekraftig høsting og dyrking fra lavere trofiske nivåer som en viktig del av løsningen. EU mener sirkulær mat- og fôrproduksjon er avgjørende for å kunne nå bærekraftsmål, ressurseffektivitet, klimatiltak, en biobasert økonomi og matsikkerhet.
Norsk havbruksnæring har som mål å ha en årlig vekst på 4,4 %, og anslår 5 millioner tonn laks pr år innen 2050. Dette fører til et betydelig fôrunderskudd. I dag utgjør fôret 75 % av norsk laks sitt klimaavtrykk, og over 90 % av ingrediensene importeres. Geopolitiske hensyn nødvendiggjør kortere og sikrere matforsyningskjeder. Den norske regjeringen understreker behovet for økt produksjon av fôr for å styrke matsikkerheten, øke bærekraften og utvikle en mer klimavennlig matproduksjon. Utvikling av nye metoder for masseproduksjon av organismer fra lavere trofisk nivå er viktig for å møte behovene i oppdrettsnæringen.
Krepsdyret Gammarus er en av disse lavtrofiske organismene. Dette lille krepsdyret har en unik evne til å konvertere råvarer av lav kvalitet til høyverdig protein og omega-3 med en riktig ernæringsmessig sammensetning og er godt tilpasset som fôrråstoff; fosfolipider, proteiner med gunstige aminosyreprofiler, karotenoider og andre antioksidanter, vitaminer og mineraler. Forskningen ved SINTEF Ocean har vist at Gammaridaes kan dyrkes på ulike sidestrømmer fra biobaserte næringer som restråstoff fra landbruk og havbruk. SINTEF-teamet har utviklet kunnskap om pilotproduksjon av Gammaridae biomasse. 
Basert på lovende forskningsresultater er det kommersielle målet å industrialisere produksjonen, og dermed skape en ny og levedyktig kilde til fôrråstoff i store mengder for at havbruksnæringen skal kunne vokse på en bærekraftig måte.

Oppdrett av kveite er en lang prosess med kompleks biologi. En av de største utfordringene er den lange levendefôr perioden. den kan være opp mot 50-60dager, og krever store mengder med byttedyret Artemia. Artemia er et lite krepsdyr som klekkes, anrikes og fôres ut til kveitelarvene. Å lage dette fôret er en stor og krevende prosess og det skal produseres mye Artemia for å mette alle kveitelarvene i en produksjon. Grunnen til at kveite ikke kan spise vanlig tørrfôr i begynnelsen av livssyklusen, er at den ikke har utviklet et mage og tarmsystem som kan fordøye dette. Den må få mat i form av levendefôr. I dette prosjektet vil vi, sammen med det portugisiske selskapet SPAROS, utvikle et tørrfôr som er spesialdesignet for kveitelarver slik at de vil være i stand til å ta opp næringen fra fôret. Hvis vi lykkes med dette kan vi kutte ut store deler av levende fôr produksjonen og spare enormt med ressurser og samtidig forbedre vår yngelkvalitet. Dette vil være em "game changer" i kveitenæringen.

Måler er å utvikle et microfôr som er skreddersydd til kveite larver. På grunn av lang produksjonstid i larvefasen (50-60dager) hvor det utelukkende brukes levendefôr, er det behov for å utvikle et fôr som kan overta denne rollen i oppdrett av kveite. I dag bruker vi i SWH 1,5tonn med Artemiacyster til dagens produksjon (for å produsere 350-450 000yngel) og i nær framtid må vi mange doble denne bruken. Så å utvikle et fôr som kan ta ned bruken av levendefôr vil være av ytterst betydning i forhold til kostnader, miljø avtrykk og biosikkerhet.
Sammen med SPAROS vil vi utvikle et skreddersydd microfôr til kveite. Sparos vil stå for fôrformulering og resept utviklingen basert på innspill fra SWH, litteraturen samt Sparos mangeårig erfaring med å utvikle spesial fôr til marine arter. SWH vil deretter teste fôret i storskala systemene våre. 

Dette prosjektet (dvs sluttproduktet) vil også, mest sannsynlig, være direkte overførbart til andre arter som f.eks torsk.

4876 - Tidlig weaning av flatfisk på microfôr: forbedret kvalitet, overlevelse på larver og yngel

Dagens matsystemer står overfor mange utfordringer, der klimaendringer er en av de største. Sjømat spiller en nøkkelrolle i overgangen til bærekraftige og næringsrike matsystemer. Sjømatforedlingsindustrien fører med seg betydelige sidestrømmer, som ofte kastes eller er underutnyttet, og som er rike på bioaktive forbindelser for høyverdige markeder som nutraceuticals og funksjonelle matvarer. Prosjektet har som mål å øke effektiviteten og ressursutnyttelsen av eksisterende sjømatsystemer (remove the - that was in this word) og fiskerier gjennom verdiskaping av sidestrømmer til næringsrike matvarer med høy verdi ved hjelp av grønn teknologi. Verdiskaping av biprodukter fra prosessering og en overgang til sirkulær økonomi bidrar til den fysiske og økonomiske motstandsdyktigheten til EUs sjømatforedlingsverdikjeder, samt til miljømessig og økologisk bærekraft i sjømatindustrien. FOODIMAR har som mål å utvikle innovative, kostnadseffektive og klimavennlige metoder for å produsere høykvalitets marint kollagen, gelatin og GAGs (glycosaminoglycan) for å imøtekomme behovene til industrien og forbrukerne. Dette vil oppnås gjennom utforskning av tre EU-piloter: Hvitfisk fra Nordsjøen, maneter fra Østersjøen og havabbor fra Middelhavet. I tillegg fokuserer FOODIMAR på utviklingen av verktøy og retningslinjer for å vise det økonomiske potensialet til disse ressursene i en klimasikker sektor.

The project aims to increase the efficiency and resource optimisation of existing seafood systems and fisheries through seafood side-streams valorisation into high-value-added nutritious food using green technologies. The valorisation of processing by-products and a shift towards a circular economy contribute to the physical and economic resilience of EU seafood processing value chains, as well as to the environmental and ecological sustainability of the seafood industry. This will be achieved through the exploration and characterization of bioactive compounds sourced through 3 EU pilots: North sea whitefish, Baltic sea jellyfish and Mediterranean seabream. Additionally, FOODIMAR focuses on the development of tools and guidelines to showcase the economic potential of these resources in the context of a climate-proof sector. This proposal involves transfer of the CMET balloon technology from US to Norwegian scientists through common fieldwork in Ny-Ålesund in 2010 and 2011. Multiday flights with CMET balloons will be valuable for the modeling community for improving transport  models used to predict pollution in the Arctic as described below.

The proposed activity is an extension of the collaboration between Paul Voss, Andreas Stohl, Lars R. Hole and others within the RCN-funded  project IPY POLARCAT (International Polar Year , www.polarcat.no). CMETs fully adapted to the Arctic environment will be a very useful tool in future projects involving studies and monitoring of tropospheric pollution transport to the Arctic.

Since Svalbard is located in one of the main pathways for  Arctic transport, knowledge of transport properties in this area is of particular interest.

Autoimmune sykdommer oppstår når kroppens immunsystem angriper sine egne, friske celler. Både gener og miljøfaktorer som ofte er ukjente er involvert i utviklingen av autoimmune sykdomer. De genene spesielt disponerer for utvikling av autoimmune sykdommer er såkalte MHC klasse II gener. Hvordan disse MHC klasse II genene disponerer for utvikling av autoimmun sykdommer er ikke klart. Dette prosjektet tar som utgangspunkt at interaksjoner mellom to typer celler i immunsystemet –  T -celler og B -celler kan forklare den genetiske effekten av MHC klasse II gener. Vi utforsker denne hypotesen i rammen av sykdommen cøliaki. Cøliaki er en vanlig autoimmun sykdom som skyldes intoleranse for glutenproteiner fra korn. Pasientene har antistoffer mot en komponent i kroppen selv: enzymet transglutaminase 2. Disse såkalte autoantistoffene dannes bare hos individer som uttrykker visse MHC klasse II- variantmolekyler, spesielt molekylet HLA-DQ2.5 og i mindre grad molekylene HLA-DQ2.2 og HLA-DQ8, og det skjer bare når gluten er en del kosten til disse individene. B-celler er cellene som produserer antistoffer. Disse cellene uttrykker MHC klasse II molekyler og de bruker MHC molekylene til å presentere fragmenter av proteiner for T-celler. I dette prosjektet bruker vi transgene mus som uttrykker HLA-DQ2.5 og sykdomsrelevante B-celle- og T-cellereseptorer fra cøliakipasienter for å studere hvordan spesifikke B-celler og T-celler interagerer. Spesielt studerer vi rollen som HLA-DQ2.5 utøver ved å bruke reagenser som blokkerer funksjonen til dette molekylet. Prosjektet vil gi grunnleggende kunnskap om hvorfor MHC klasse II gener er risikofaktorer ved autoimmune sykdommer.

This project aims to give a proof-of-principle of the mechanism underlying the major role of MHC class II genes in autoimmunity. Central in the project will be studies of cross-talks between pathogenic T cells and B cells. As such cross-talks in vivo take place primarily in organized tissues, whole organism models are required. Thus, the project will utilize experimental mouse models. Taking advantage of the identification of adaptive immune receptors of culprit T cells (T-cell receptors, TCRs) and B cells (B-cell receptors, BCRs) of celiac disease patients, a collection of unique TCR and BCR transgenic mouse strains have been established. In combination with a novel HLA-DQ2.5 knockin mouse strain, these humanized mouse strains serve as a powerful tool-box for exploring the role of HLA-DQ2.5 in the formation of antibodies to the autoantigen of celiac disease, transglutaminase 2 (TG2). In celiac patients anti-TG2 antibodies are formed in a gluten dependent (i.e. gluten eating) and HLA dependent (i.e. positivity for HLA-DQ2.5/DQ2.2/DQ8) manner. Mechanistically, this gluten and HLA dependence can be explained by gluten-specific T cells providing help to TG2-specific B cells via the involvement of TG2-gluten complexes. In the project this mechanistic model will be explored by testing of interaction between antigen-specific T cells and B cells of the humanized mice. In particular the role of HLA-DQ2.5 in facilitating production of anti-TG2 antibody will be probed by blocking with anti-HLA-DQ and anti-HLA-DQ2.5/peptide monoclonal antibodies. The project will give new insights with demonstration of key pathogenic principles in celiac disease, a poster child of MHC associated diseases. Altogether, the project should provide fundamental knowledge into processes causing MHC association with disease – a scientific area where our insights into the underlying molecular mechanisms are appallingly scarce.

Exploring T cell – B cell collaboration resulting in autoimmunity

Smittsomme sykdommer hos husdyr fører til store økonomiske tap globalt, og truer i tillegg matsikkerhet og folkehelse. Vaksinasjon er fortsatt det mest kostnadseffektive verktøyet for å forebygge, håndtere og til og med utrydde sykdommer. De fleste vaksiner gir beskyttelse ved å indusere antistoffer, som kan overføres til avkommet gjennom morkaken eller gjennom råmelk tidlig etter fødselen. Slike maternelt avledede antistoffer (MDA) spiller en kritisk rolle for å beskytte nyfødte husdyr mot smittsomme sykdommer i løpet av deres tidlige liv. Imidlertid kan MDA også dempe immunresponsene hos nyfødte etter vaksinasjon ved å nøytralisere vaksinen før dyrene induserer en immunrespons. Så når MDA avtar, forblir immuniserte dyr sårbare for smittsomme sykdommer. I dette prosjektet vil vi utvikle og teste DNA vaksiner designet for å forbedre immunresponsene hos nyfødte dyr med MDA.

Vi har fokusert på Porcine reproductive and respiratory syndrome virus (PRRSV) hos griser. PRRSV regnes som den sykdommen som medfører den størst økonomiske belastningen for den globale svinindustrien med estimerte tap i Europa som overstiger 1,5 milliarder euro per år. MDA forstyrrer vaksinasjon av grisunger mot PPRSV, noe som fører til høy sårbarhet på tidspunktet for avvenning eller oppforing der MDA er lav og indusert immunitet ennå ikke er på plass.

I prosjektet har vi laget vaksiner der PRRSV-antigener leveres spesifikt til dendrittisk celle (DC) som en metode for å forbedre immunresponsen. Vi har testet om en målrettet vaksine, etterfulgt av en svekket levende vaksine (MLV), forbedrer antistoffresponsen og øker vaksineeffektiviteten i grisunger med MDA. Forsøkene har vist at målstyrte vaksiner gav øke immunresponser i grisene, og vi jobber nå med å evaluere effekten på MDA. Utfyllende eksperimenter utføres for å bedre forstå de molekylære mekanismene bak den negative effekten av MDA på immunresponsene i grisunger.

Infectious diseases of livestock continue to cause a major financial impact globally, threatening food security and public health. Disease control measures comprise a combination of biosecurity with both preventative and therapeutic treatments. However, vaccination remains the most cost-effective tool by far to prevent, manage and even eradicate diseases. Maternally derived antibodies (MDA) play a critical role in protecting neonatal livestock against infectious diseases during their early life. However, MDA can dampen immune responses of neonates to vaccination through a number of distinct mechanisms, including the neutralisation of live attenuated vaccines and the inhibition of neonatal B cell responses. Thus, as MDA wanes immunised animals remain vulnerable to pathogen challenge. In this project, we will test a DNA vaccine strategy designed to enhance immune responses in neonatal animals with MDA. 

We will focus on porcine reproductive and respiratory syndrome virus (PRRSV) in pigs, for which the applicants have developed preliminary tools  to counteract different mechanisms of MDA interference. PRRSV is considered the most economically important disease affecting the global pig industry with estimated losses in Europe exceeding €1.5 billion p.a. MDA interfere with neonatal vaccination against PPRSV leading to high vulnerability at the time of weaning or feedlot/fattening where MDA are low and active immunity is not yet in place.

We will generate DNA vaccines where PRRSV antigens are fused to XCL1 or anti-MHC-II scFv, as dendritic cell (DC) targeting moieties that we have shown enhance immunogenicity. We will then test if a targeted DNA prime/modified live vaccine (MLV) boost vaccine strategy enhances cell mediated and antibody responses and increase vaccine efficacy in MDA+ piglets. Complementary in vitro experiments will be conducted using DC targeted antigens to understand how they counteract the MDA-negative effect on B-cell responses.

ICRAD Novel Strategies to enhance vaccine immunity in neonatal livestock.

Stem cell and regenerative medicine research in Norway is being carried out at multiple research and clinical institutions, and there is a strong need to provide a national forum in which researchers, clinicians, patient groups and health policy makers can meet to interact and network. This project is based on prior experience in organizing national conferences in this area.

Cøliaki er en vanlig sykdom som skyldes en intoleranse mot glutenproteiner i korn. Pasienter med cøliaki lager antistoffer mot en komponent i kroppen selv - enzymet transglutaminase 2 - såkalte autoantisoffer. Hvis en person lager slike autoantistoffer, er det meget sannsynlig at personen har sykdommen. Påvisning av anti-transglutaminase 2 autoantistoffer blir brukt til å stille cøliakidiagnosen.

Målet for dette prosjektet har vært i få innsikt i hvorfor og hvordan autoantistoffer mot transglutaminase 2 dannes. Antistoffer produseres av B-celler. B-celler trenger hjelp av T-celler for å produsere antistoffer. Vi har utviklet et eksperimentelt system med transgene mus, der sykdomsrelevante B- og T-celle antigenreseptorer fra cøliakipasienter har blitt introdusert i musenes genomer. Vi studerer hvordan B-celler og T-celler interagerer i disse musene. Studiene har gitt grunnleggende ny innsikt i hvorfor immunsystemet går til angrep på kroppen selv hos cøliakipasienter når de spiser gluten.

Normalt unngås autoimmune sykdommer ved at B-celler som gjenkjenner kroppsegne strukturer blir fjernet fra kroppens B-cellerepertoir. Alternativt kan autoreaktive B-celler eksistere, men de blir holdt inaktive. Med vår musemodell har vi vist at B-celler som gjenkjenner transglutaminase 2 eksisterer, og at de kan produsere autoantistoffer hvis de får hjelp fra T-celler.

Among the most disease-specific autoantibodies in humans are those reactive with transglutaminase 2 (TG2). IgA anti-TG2 antibodies are diagnostic for celiac disease, and strikingly, they are only formed when celiac patients are consuming cereal gluten proteins. 

In this project we will harness new humanized mouse models to investigate 
a) whether and to which extent there is B-cell tolerance to TG2 and 
b) how anti-TG2 autoantibodies can be formed in response to exposure to the exogenous antigen gluten.

We have established novel transgenic mouse strains that carry a B-cell receptor of a TG2-specific plasma cell and a T-cell receptor of a gluten-specific T cell - both based on immune cells isolated from lesions of celiac disease patients. Our preliminary results indicate that there is minimal or no B-cell tolerance to the TG2 autoantigen despite the fact that the transgenic knockin B-cell receptor has equal affinity for human and mouse TG2. This suggests that the main control of antibody production to TG2 operates at the level of provision of T-cell help. Conceivably, gluten-specific T cells can provide such help by involvement of hapten-carrier like TG2-gluten complexes. Preliminary data from adoptive transfer experiments with transgenic T cells and B cells indicate that gluten-specific T cells indeed can provide such help. Further experiments providing mechanistic insight into the T-cell/B-cell collaboration involved in TG2 autoantibody production will be undertaken. Work of the project should provide answers to fundamental questions related to celiac disease, and it should help to advance the understanding of autoimmune diseases in general.

Understanding how gluten drives autoimmunity in celiac disease

Utvikling av immunterapi mot kreft basert på aktivering av immunceller i tumor 

Det grunnleggende prinsippet ved immunterapi er å styrke pasientens naturlige immunforsvar mot kreft slik at immunforsvaret kan virke som en kur. I 2010 var det et stort gjennombrudd innen fagfeltet immunterapi mot kreft. Da ble det vist at en behandling basert på hemmere av bremser (sjekkpunkt) i immunsystemet, kunne forlenge levetiden til så mange som 10-20 % av pasientene med avansert kreft. Denne behandlingen benyttes nå mot flere typer kreft, og aktiverer en type av immunceller kalt T-celler. I dette prosjektet har vi hatt som mål å utvikle nye former for immunterapi som kan gi effektiv behandling av flere pasienter. Vi fokuserer forskningen vår på en immuncelle kalt makrofag. Denne celletypen finnes i alle solide kreftsvulster. Makrofager kan drepe kreftceller, men bare dersom makrofager mottar bestemte molekylære signaler fra miljøet rundt. Ved å dyrke makrofager in vitro, har vi definert hvilke signaler som kreves for at makrofagene skal bli aktivert til å drepe kreftceller. Vi fant ut at makrofager krever en kombinasjon av to molekylære signaler for å få lisens til å drepe. Vi har identifisert flere slike molekylære oppskrifter. For eksempel har vi funnet at store sukkermolekyler isolert fra medisinsk sopp, har stort potensial i makrofagbasert immunterapi. Ved hjelp av musemodeller for lungekreft og brystkreft, har vi utviklet strategier for å levere disse signalene i kreftsvulster slik at makrofagene der kan aktiveres til å drepe kreftceller og kurere kreft. Vi har oppnådd lovende resultater med bruk av virusbasert genterapi. I denne strategien benyttes virus til å levere molekyler inne i kreftsvulster. Molekylene programmerer makrofagene til å utrydde kreften. Vi har også publisert en detaljert beskrivelse av hvilke type immunceller som finnes i humane lungekreftsvulster. Studien representerer en milepæl for fremtidig forskning på området. Så mange som tretten ulike typer immunceller, inkludert makrofager, ble identifisert i kreftsvulstene som hadde blitt fjernet ved kirurgi. Til slutt har vi etablert en metode som kan identifisere immunresponsen som etableres i hver enkelt kreftpasient. Målet er å bruke metoden til å kunne gi en prognose og kunne si noe om hvorvidt pasienten kan ha effekt av immunterapi. Metoden er blant annet basert på bruk av kunstig intelligens som benyttes til å analysere flerfargede bilder av kreftsvulsten. På denne måten kan vi identifisere og kvantifisere alle typer immunceller som er tilstede i kreftsvulstene. Dette prosjektet legger grunnlaget for en ny generasjon av immunterapi mot kreft basert på optimalisert aktivering av makrofager i kreftsvulster.

Immunotherapy is considered one of the most promising novel strategies to cure cancer. Enhancement of T-cell mediated antitumor immunity by "immune checkpoint" blockade or by T-cell adoptive transfer seems to be particularly promising. However, most patients do not respond to current treatments. By using a mouse model for myeloma, we discovered that tumor-infiltrating macrophages may be very efficient at eradicating cancer upon activation by tumor-specific Th1-polarized T cells. We have recently reported that the Th1-derived cytokine interferon-gamma and the inflammatory cytokine interleukin-1 could synergize to render macrophages cytotoxic to cancerous cells. Thus, it appears that two molecular signals are required for optimal activation of macrophages. In this proposal, we aim at translating our recent findings to novel immunotherapeutic protocols based on 2-signal activation of tumor-infiltrating macrophages. We will use two mouse models for non-small cell lung cancer (NSCLC) including patient-derived tumor xenografts (PDX), which we have already established. Three strategies will be tested for rendering tumor-infiltrating macrophages cytotoxic to cancerous cells: i) in situ macrophage activation with optimized molecular combinations including cytokines and pectic polysaccharides isolated from medicinal plants; ii) intratumoral delivery of immunostimulatory cytokine genes by recombinant alphaviral particles; iii) enhancement of antitumor immunity by immune checkpoint blockade on macrophages. Furthermore, we plan to develop novel prognostic tools for NSCLC based on the analysis of tumor-infiltrating immune cells, and to evaluate PDX as preclinical models for NSCLC immunotherapy. Lung cancer is considered to be potentially quite immunogenic and thereby appropriate for immunotherapy. Therefore, this project may significantly impact the treatment of NSCLC which currently represents a major burden in Norway and worldwide.

Optimized activation of tumor-associated macrophages as a novel strategy for cancer immunotherapy

Til tross for den enorme effekten antibiotika og vaksiner har hatt på vår allmenhelse, så utgjør fortsatt infeksjonssykdommer den nest vanligste dødsårsaken på verdensbasis. Fraværet av effektive vaksiner mot sykdommer som tuberkulose, HIV og malaria, er eksempler på hvorfor videre forskning på vaksineutvikling er nødvendig. En lovende metode for å forbedre effekten av vaksiner er å målstyre antigener (dvs. komponenter tatt fra virus eller bakterier) til bestemte immunceller som er ansvarlig for å sette i gang immunsvaret (såkalte antigen presenterende celler (APC)). I dette prosjektet har vi studert immunsvaret når antigener målstyres mot en bestem underklasse av APC som heter Xcr1+ dendritiske celler (DC). 

Tidligere studier har vist at denne vaksinasjonsmetoden fører til et forbedret immunsvar med gode CD8+ T celle responser, også kalt dreper celler på grunn av deres evne til å drepe virus infiserte celler og kreftceller. I tillegg har vi sett at målstyring av antigen mot Xcr1+ DC også fører til bedre antistoffresponser. Gjennom dette prosjektet har vi jobbet for å beskrive de bakenforliggende molekylære mekanismene som gir et forbedret immunsvar når antigener målstyres mot Xcr1+ DC. Vi har vist at målstyring mot Xcr1+ DC med vaksiner som ikke blir aktivt tatt opp av cellene gir sterkere antistoff responser, og gir bedre beskyttelse mot influensainfeksjon. Derimot gir målstyrte vaksiner som blir aktivt tatt opp av DC bedre CD8+ T celle responser. Dette informasjonen kan brukes til å spesifikt lage vaksiner som fortrinnsvis gir antistoff eller T celle responser, og teknologien blir nå evaluert i kontekst av SARS-CoV-2.

For å målstyre antigener mot Xcr1+ DC så har vi koblet dem til signalmolekylet Xcl1, det eneste kjente signalmolekylet som binder Xcr1 reseptoren. Vi har sett at vaksineringssted påvirker immunresponsen ulikt om man bruker Xcl1 eller et annet signalmolekyl (CCL3) for å målstyre DC. Mens antigen koblet til Xcl1 gav svært like immunresponser ved intramuskulær og intradermal vaksinering, så gav vaksinering med antigen koblet til CCL3 betydelig bedre responser ved intramuskulær vaksinering enn ved intradermal. Videre har vi sett at Xcl1 gir en immunrespons dominert av CD4+ T celler av en Th1 type og antistoff av typen IgG2a, i tillegg til dreper T celler. Denne typen immunresponser er spesielt godt egnet for å eliminere virus- eller bakterieinfiserte celler, og kan potensielt sett også brukes innen immunoterapi for å drepe kreftceller. Antigen koblet til Xcl1 har også vist seg å mer effektivt gi Th1 celler og dreper T celler sammenlignet med målstyring mot overflate reseptorene DEC-205 og Clec9a, som også er uttrykt på samme type Xcr1+ DC. Resultatene viser dermed at valg av overflate reseptor på DC spiller en viktig rolle i bestemmelse av immun responsen etter vaksinering, hvilket utfordrer tidligere studier som har hevdet at immun responsen i hovedsak blir bestemt av DC populasjonen.

Despite the huge impact that antibiotics and vaccines have had on human health, infectious diseases are still the second leading cause of death worldwide (WHO, 2002). The continued lack of efficient vaccines against diseases such as tuberculosis, HIV and malaria, and new emerging diseases such as the recent ebola epidemic, remind us that continued research into vaccine development is of great importance. In this research proposal project manager (PM) wishes to study a novel vaccine strategy based on targeting antigens to the Xcr1 receptor, specifically expressed on cross-presenting dendritic cell (DC). PM's recent work has demonstrated that targeting Xcr1 using the chemokine Xcl1 is an efficient method for inducing cytotoxic T-cell responses, and for selective induction of Th1 associated antibodies of IgG2a/2b subclasses. By comparing antigen targeting to different receptors on the same population of DCs (Xcl1 vs Clec9A), in combination with more functional studies using Xcr1 knockout mice and bone marrow derived DCs, PM hopes to determine the mechanism behind an Xcl1 mediated immune response. One of the main questions to be addressed is whether the observed immune response is a result of targeting a specific DC population, or if ligation of the Xcl1 chemokine to the Xcr1 receptor influences the response. Using this information PM will further develop and optimize the Xcl1-mediated targeting approach for improved induction of Th1 associated IgG2a/2b antibody responses and/or CD8+ T cell responses. A better understanding of how the immune responses can be manipulated towards a CD8+ T cell or a Th1 associated antibody response could be of great value in future vaccine development against both pathogens and cancer.

Vaccine molecules that polarize the immune response towards CD8+ T-cell and IgG2a/2b antibody responses (X-VAC)

A fundamental question in neuroscience is how neuronal circuits are assembled during embryonic and fetal development. Critical features include the establishment of functionally appropriate patterns of synaptic connections and a proper balance between exc itatory and inhibitory connections. Sensorimotor reflex circuits provide excellent models for the study of these features since they involve highly specific connections from afferents onto premotor neurons and from premotor neurons onto motoneurons (Glove r 2000). The vestibulo-ocular reflex (VOR) arc is a particularly tractable sensorimotor reflex circuit in this regard because it comprises a well-described pattern of polysynaptic connections with a stereotyped mapping of sensory inputs to motor outputs.  Thus, the VOR circuit provides an excellent system for studying both the molecular mechanisms that specify synaptic connections during development and the acqusition of proper balance in excitation and inhibition, perturbations of each of which can lead t o inappropriate activity such as seen in various human neurological disorders (movement disorders, spasticity, epilepsy). 

In this project two laboratories, one in Oslo and one in Munich, will collaborate to establish genetically mediated pharmacological  and optical manipulations of activity in specific neurons and neuron groups in the VOR circuit. This will significantly advance our understanding of VOR function and development through 1) a more precise dissection of synaptic interactions within the ind vidual sensorimotor channels of the VOR, including characterization of patterns of synaptic convergence, and 2) temporally controlled perturbance of dynamic activity levels to assess their effects on synaptic patterning and scaling. This will provide insi ght into the mechanisms of synapse formation, patterning and balance, which are of high general relevance for the development of circuits in all parts of the brain.
