Effektene av fysisk og seksuell vold på ofre og deres familier kan være ødeleggende og vedvarende, med potensielt langvarige konsekvenser for fysisk og mental helse, utdanning, sysselsetting og bruk av velferdstjenester. Mens strafferettssystemet anses som en viktig del av offerets gjenopprettingsprosess, er det begrenset empirisk forskning på hvorvidt strafferettslige prosesser hjelper og/eller hindrer bedring langs disse ulike dimensjonene. I dette prosjektet vil vi adressere dette kunnskapshullet ved å bruke en kombinasjon av kvantitative og kvalitative forskningsmetoder. I de kvantitative analysene vil vi bruke administrative registerdata som dekker hele befolkningen sammen med kvasi-eksperimentelle metoder for å kunne trekke kausale slutninger. Vi vil også benytte maskinlæringsteknikker for prediksjon. De kvalitative analysene vil supplere de kvantitative analysene ved å gi kontekst til et komplekst felt av saker og ofre og ved å identifisere potensielle mekanismer mellom strafferettslige prosesser og utfall for ofrene. Samlet vil disse metodene gi en mer robust, representativ og nyansert forståelse av hvordan kontakt med strafferettssystemet påvirker offeret på tvers av flere viktige livsområder både på kort og lang sikt. Prosjektet ledes av Folkehelseinstituttet (FHI) med Frischsenteret og Universitetet i Oslo som partnere. Prosjektet har et rådgivende utvalg med medlemmer fra Nasjonalt kunnskapssenter om vold og traumatisk stress (NKVTS), ETH Zurich, Universitetet i Oslo, Frischsenteret, Universitetet i Bergen, og OsloMet. Det er også tilknyttet en brukergruppe bestående av personer og organisasjoner som er i kontakt med ofre for vold og overgrep.

Victims of sexual and physical violence is one of the most vulnerable groups in our society. The criminal justice system (CJS) serves a critical role in providing assistance to victims of these crimes, enabling them to seek justice, protection, compensation, and closure. Yet, knowledge about whether the CJS helps or harms these victims is extremely limited. This project aims to remedy this by providing a comprehensive understanding of the role of the CJS in supporting and providing restoration for victims of physical and sexual violence in Norway. We will utilize a combination of quantitative and qualitative research methods across disciplines to study outcomes across multiple important life domains, both in the short and long run. For the quantitative analyses we will use exceptionally rich longitudinal administrative register data with full population coverage combined with state-of-the-art quasi-experimental methods for credible causal inference and predictive machine learning techniques. The qualitative analysis will be based on narrative interviews with victims, providing context and insights into how interactions with the CJS shape life stories. The results will yield a robust and nuanced understanding of how the different stages of the CJS impacts victims. This is particularly urgent given how the current emphasis on legal responses to assaults has caused a sense of obligation and civic duty for especially women to report incidents, while the chances of legal closure remain small. In addition to advancing the methods for quasi-experimental designs and machine learning within quantitative criminology, this project will provide valuable insights for policymakers, stakeholders, and practitioners working in the field of victim support and rehabilitation and may inform current legal reform efforts aimed at addressing these issues.

Helpful or harmful: The effects of criminal justice system contact on victims of sexual and physical violence

Hovedmålet med kreftbehandling er å eliminere kreftceller og dette er tradisjonelt oppnådd ved kirurgi, bruk av giftige medikamenter (cellegift) og stråling. En ny og betydningsfull tilnærming til kreftbehandling kalt immunterapi, har derimot fått mye oppmerksomhet det siste tiåret; Her forsøker leger og forskere å utnytte immunsystemet vårt for å målrettet  ødelegge kreftceller. En nøkkelaktør i denne banebrytende tilnærmingen er en gruppe «drapsmenn» i immunsystemet kjent som cytotoxiske T-lymfocytter (CTL-er). Disse cellene har en bemerkelsesverdig evne til å oppdage kreftceller ved å gjenkjenne unormale molekyler på overflaten av kreftcellene - molekyler som friske celler ikke har. Når CTL-er gjenkjenner kreftcellene ødelegger de dem ved å frigjøre giftige molekyler som initierer kreftcellenes selvdestruksjon. 
 
Selv om betydelige innsats har blitt dedikert til å forstå hvordan CTL-er aktiveres og hvordan de gjenkjenner kreftceller, vet vi mindre om de påfølgende hendelsene inne i kreftcellene som fører til deres død. Fra før vet vi at CTL-er kan initiere to forskjellige typer celledød i kreft: den første, kalt apoptose, er en stille og ordnet prosess der kreftcellene krymper og brytes opp uten å varsle immunsystemet; den andre, kalt pyroptose, er en mer dramatisk prosess som involverer dannelse av åpninger i cellemembranen, noe som fører til at cellene svulmer opp og sprekker. Under denne cellebristen blir innholdet inne i cellene frigitt i omgivelsene, og dette kan fungere som 'faretegn' som kan stimulere en sterkere immunrespons mot kreftcellene. Betydningen av denne cellebristen for å forsterke anti-tumorresponsene vil bli undersøkt i dette prosjektet. 
 
Videre er vi fascinert av muligheten for å avdekke ytterligere, ennå ukjente, former for celledød indusert av CTL-er. Utforsking av disse dødsrelaterte prosessene i kreftcellene kan potensielt øke effektiviteten av immunterapi og bidra betydelig i den pågående kampen mot kreft.

The primary goal of most cancer therapies is to induce cancer cell death. Immunotherapy relies on the cytotoxic activity of lymphocytes such as natural killer cells (NKs) and cytotoxic T lymphocytes (CTLs) to achieve this. While a significant amount of research in immunotherapy focuses on the events within cytotoxic lymphocytes or at the interface between cytotoxic lymphocytes and cancer cells, less is known about the molecular mechanisms that orchestrate cancer cell death, particularly in the case of CTL-mediated death.
CTLs are known to induce two types of cell death in cancer: non-inflammatory apoptosis and proinflammatory pyroptosis. Pyroptotic cancer cells release proinflammatory signals that attract immune cells to the tumor site, thereby enhancing the anti-tumor response. Cell lysis is a common outcome of pyroptotic cell death. It is unclear whether lysis can be decoupled from pyroptosis in cancer cells and whether the proinflammatory properties of pyroptotic cancer cell death depend on lysis. Moreover, it remains unknown whether CTLs can induce other forms of cell death in cancer, apart from apoptosis and pyroptosis.
Studying the mechanisms of CTL-mediated cancer cell death is challenging because CTLs selectively target cells expressing specific antigens. However, given the critical role of CTLs in immunotherapy, it is essential to deepen our understanding of CTL-induced cancer cell death.
This project aims to develop novel methodologies to investigate the death mechanisms that are initiated by CTLs. In addition, we will identify new pathways and mediators involved in CTL-induced cancer cell death. Furthermore, we will assess the contribution of proinflammatory cell death and lysis in promoting anti-tumor responses in vivo. Overall, this research will improve our knowledge of the molecular mechanisms of CTL-mediated cancer cell death and potentially reveal novel targets to enhance the efficacy of cancer immunotherapy.

T-cell-mediated proinflammatory cell death and lysis in cancer

Renewable energy on the high seas – science-informed and indigenous knowledge-based recommendations for regulatory and governance challenges

The high seas are those marine areas under no State's jurisdiction. So far, all marine renewable energy (RE) activity is being conducted under State's jurisdiction. However, recent advances in technology are raising the prospect of carrying out RE activity on the high seas. Currently, only seabed mining activities in the Area have been regulated. The Area is the seabed and ocean floor beyond the limits of State's jurisdiction. From the legal viewpoint, the main gap is a lack of an international regulatory framework, informed by participatory mechanisms and supported by scientific evidence, for RE activities on the high seas, especially regarding their environmental and social impacts. 
The primary project objective (O) is to provide the scientific community and policymakers with science-informed and indigenous knowledge-based recommendations in support of the development of  RE regulation on the high seas. 
Two specific Os are set to complement the primary O, namely:
O1 To advance the use of systematic consultation processes that facilitates research at the crossroads of marine sciences and law. 
O2 To advance critical knowledge and understanding of environmental pressing issues comparable to the regulation of RE on the high seas, such as deep-sea mining in the Area.
A multistep innovative methodology will be adopted, by integrating marine scientific knowledge, consultative processes, and advanced critical research in the subject matter. A case study (interdisciplinary and participatory) involving marine scientists, indigenous and coastal communities and policy makers will be conducted onboard the Research Vessel KronPrins Haakon at the beginning of the project. The case study outcomes will be analysed through critical legal analysis to build a knowledge base for policy recommendations. 
The project will advance the PI’s career in an international research environment, allowing her to generate independent, high-quality, and reproducible results.




The Arctic region is witnessing more frequent shipping accidents than ever which require accurate search and rescue operations. The existing communication systems use drones with radio frequency (RF) technology, which has limited bandwidth and cannot offer 4G/5G connectivity due to harsh weather constraints. It further limits the capacity of drones and affects the resolution of images and videos. The current research proposes to design free space optical communication (FSOC) link-based drones that can enhance connectivity between multiple drones and rescue ships as well as the capacity of drones to produce high resolution images and videos. The proposed FSOC-based drones can provide highspeed connectivity than the existing RF technology. The research will collect atmospheric turbulences data from the Arctic region and use it for designing the FSOC link. Furthermore, to enhance the capacity of FSOC-based drones, the research will design advanced multiplexing schemes (Mode Division Multiplexing and Orthogonal Code Division Multiple Access) and to ensure link stability, omnidirectional receiver will be designed. The research will further evaluate the performance of proposed FSOC link under the impact of various atmospheric turbulences. The expected outcomes of this research will lead to the optimisation of FSOC-based drones in expanding search areas and improving communication systems for SAR operations in the Arctic region.

Optimization of Free Space Optical Communication Link based Drones for Search and Rescue Operations in the Arctic Region

Dette prosjektet, kalt Hukommelse for musikk, fokuserer på det økende antallet mennesker over hele verden som lever med demens, spesielt Alzheimer's sykdom (AD). AD starter typisk med hukommelsesproblemer og påvirker etter hvert daglige aktiviteter. Aktive musikkintervensjoner, spesielt sang, har vist positive effekter på humør, atferd og livskvalitet for personer med demens, men deres innvirkning på kognisjon er ikke godt forstått. Prosjektet har som mål å fylle dette kunnskapsgapet ved å studere effektene av å lære nye sanger på kognitiv, atferdsmessig og hjernemessig funksjon.

Studien vil omfatte hjemmeboende voksne over 65 år med AD fra Argentina, Østerrike og Norge. Deltakerne vil gjennomgå 5 måneder med intensiv musikalsk opplæring (to ganger i uken) og 5 måneder med minimal opplæring (en gang i måneden) i tilfeldig rekkefølge, med en to måneders pause imellom. Intervensjonene inkluderer å lære nye sanger med en personlig musikklærer. Generell kognisjon vil bli målt ved bruk av Alzheimer's Disease Assessment Scale - Cognitive (ADAS-cog), og hukommelse for musikk vil bli vurdert gjennom ulike metoder, inkludert atferdsoppgaver og hjerneaktivitet (EEG). Humør vil også bli evaluert i hver økt.

Målet er å inkludere 113 deltakere for å sikre pålitelig påvisning av meningsfulle effekter. Studien vil utforske hvordan humør og hukommelse for musikk bidrar til endringer i kognitive evner, og om disse effektene varierer basert på faktorer som kjønn, alder, AD-stadium, eller tidligere musikalsk opplæring og generell utdanning. Prosjektet vektlegger samarbeid mellom forskere, tjenesteytere og brukere for å sikre studiens relevans og anvendelighet.

The number of people living with dementia is increasing and is high in low, middle, and high-income countries. Alzheimer’s disease (AD), the most common type of dementia, manifests itself initially with cognitive impairment in memory domains, and later affects all activities of daily living. Active music interventions, particularly singing, can help to improve mood, behaviour problems, and quality of life. Too little is known about their effects on cognition, although some promising studies exist. The M4M project aims to fill this gap by measuring effects of learning new songs on clinical, behavioural and brain functioning. Specifically, M4M aims to examine changes in general cognitive functioning and memory for music in non-musician adults with AD undergoing intensive individual musical training based on singing novel songs, compared to minimal training.
        Home-dwelling adults with AD, 65 years or older, in 3 countries (Argentina, Austria, Norway), will receive 5 months of intensive intervention (2x/week) and 5 months of minimal intervention (1x/month), in random order, with a 2-month break in between. Interventions will entail learning new songs with an individual music teacher. At the end of each intervention period, general cognition will be measured with the Alzheimer's Disease Assessment Scale – Cognitive (ADAS-cog) by a person who is unaware of the intervention received. Memory for music will be tested with observations, behavioural tasks, and brain responses (EEG). Mood will be assessed in each session. We aim to include 113 participants; this will help us to reliably detect clinically meaningful effects. We will also examine how mood and memory for music lead to changes in cognitive abilities, and whether effects depend on sex, age, AD stage, or previous musical training or general education. M4M will be conducted in close collaboration between academic researchers, service providers, and service users to ensure relevance and applicability.

Memory for Music: Effects of individual intensive musical training based on singing in non-musicians with Alzheimer’s disease

Evaluation of thermohydraulic characteristic of printed circuit heat exchangers in pseudocritical region for supercritical CO2 cycle

Currently, 28.2% of the total EU-28 greenhouse gas emissions come from the power sector, a large contributor to greenhouse gas emissions. Consequently, the emphasis of the research in power generation has swung towards assessing highly efficient and greener power cycles. In this reference, the novel supercritical carbon dioxide Brayton cycle (sCO2-BC) is an ideal choice that outstrips other formally well-known power cycles (Brayton & Rankine cycles). In sCO2-BC, the role of the pre-cooler is critical. It serves as a sink to the power cycle and regulates the conditions at the compressor's inlet. The compressor's inlet temperature is intended to be maintained close to the critical temperature of carbon dioxide (CO2) to achieve greater cycle efficiencies. However, exceptionally higher values of the specific heat capacity of CO2 near its critical point (up to 40 times higher than water) require exceedingly high water flow rates on the cold side to achieve the desired exit temperatures of CO2. Consequently, the pre-cooler's pumping power requirements become high enough to deteriorate the cycle's performance. This problem can only be mitigated by exploring new channel geometries with enhanced thermohydraulic characteristics. Therefore, the proposed study plan to characterize the complex thermohydraulic characteristics in the pseudocritical region of CO2 using a multifaceted technique that includes, experimental, numerical, and machine learning techniques. The proposed work will provide a step forward to the success of sCO2-BC technologies that, in turn, will facilitate its integration with the green energy resources (generation-IV nuclear reactors and solar concentrated plants), helping to meet the EU's 2030 climate and energy framework goals of achieving at least 32% share for renewable energy.

Norge har under Parisavtalen påtatt seg en forpliktelse til å redusere klimautslippene sine med minst 55 prosent innen 2030 sammenlignet med nivået i 1990. Hittil har vi kun oppnådd 5 prosent av dette målet; vi har følgelig kun syv år på å redusere de resterende 50 prosent for å imøtekomme målene. For 2050 har vi satt oss enda større mål: å redusere klimautslippene med hele 90 prosent. Bakgrunnen for målene er at vi prøver å unngå de verste scenarioene FNs klimautvalg skisserer opp i sine rapporter. I tillegg har energiforbruket økt, og prognosene fra Statnett for ulike scenarier viser at energiforbruket skal fortsette å øke. Vi trenger følgelig mer energi. Dette har bidrat til at diskusjoner rundt kjernekraft i Norge er høyaktuelle og pågående. Norsk Kjernekraft AS har blitt opprettet med formål om å bygge Norges første kjernekraftverk. I tillegg ble det i fjor også bevilget 200 millioner kroner fra Regjeringen til et nytt senter som skal drive med nukleær forskning. Målet er å styrke den norske kunnskapsberedskapen på det nukleære området. Dette betyr at vi beveger oss i en retning hvor kjernekraft en dag kan bli aktuelt i Norge, og noen mener at dette er den eneste veien å gå hvis vi skal nå målene våre for 2050. Men i dag sitter vi med flere spørsmål enn svar, blant annet: Hvilken teknologi og leverandør burde vi velge? Er regelverket for snevert og strengt? Hvordan kan dette gjøres og samtidig ivareta sikkerhet? Hvordan tjener et kjernekraftverk penger?

Dette prosjektet skal svare på samtlige av disse spørsmålene i en detaljert og omfattende analyse. Her bruker vi grunnlagsdata til å vurdere dagens forskning opp mot hvordan industrien i andre land opererer, og ut ifra dette forsøke estimere hva risikoen knyttet til å bygge kjernekraft som industri i Norge er. Prosjektet sikter på å gi et så helhetlig bilde som mulig slik at vi kan gjøre oss opp en mening basert på fakta; om kjernekraft passer inn i miksen av det som utgjør norsk energi.

Denne prosjektbeskrivelsen skisserer forskningen Vala Maria Valsdottir (omtales herunder som stipendiaten) skal utføre innenfor sin doktorgradsavhandling. Prosjektet skal være en del av Senter for nukleær forskning (SNF) som fikk midler vinteren 2023.
Ph.d.-prosjektet er initiert av Inmeta Consulting AS, som igjennom denne ph.d.-stillingen vil bli en del av SNF. Inmeta planlegger en prosjektperiode på fire år, hvor stipendiaten arbeider 75 % med ph.d.-prosjektet og 25 % med vanlig virksomhet hos Inmeta. 
Prosjektet skal undersøke muligheten for kjernekraft som industri i Norge. Dette er både en samfunnsaktuell problemstilling samt en som medlemmene i SNF er interessert i å utforske. Stipendiaten kommer derfor til å samarbeide med alle aktørene som er involvert i senteret og benytte forskningsdata fra dem. Dette gjelder da spesielt Institutt for energiteknikk (IFE) som har mest grunnlagsdata som er relevant for dette prosjektet.
Kandidaten ferdigstilte en masteroppgave høsten 2020 i kjernefysikk på Universitetet i Oslo (UiO) og har derfor gode og relevante grunnkunnskaper for å ta på seg et slikt prosjekt. I tillegg har kandidaten tre års erfaring innenfor energibransjen, og er derfor oppdatert på industrien og forstår deres problemstillinger godt.

Er kjernekraft aktuelt for Norge?

Potetskrell, et biprodukt fra industriproduksjon av potet (pommes frites, flak, chips mm.) er en råstoff ressurs som kan bli bedre utnyttet til nye formål. Skrellet er nemlig en rik kilde på antioksidanter, stoffer som planter naturlig har i seg for å beskytte mot skadelige UV-stråler og reaktive forbindelser som oksygen radikaler. Antioksidanter behøver vi også å tilsette i matvarer for å beskytte og ta vare på umettet fett så det ikke blir harskt og vondt i kontakt med luft. I dag brukes flere ulike syntetiske stoffer i matvarer for å hindre oksidasjon av fett og oljer. Men det er reist bekymring rundt sikkerheten for bruken av dem. Matvareindustrien, myndigheter og forbruker ønsker derfor å fjerne flere syntetiske antioksidanter og erstatte dem med naturlig alternativer. Det er imidlertid svært få slike produkter som er godkjent og tilgjengelig på markedet. Prosjektet Peel Preserve, som har et klart bærekraft mål, skal vise om et uttrekk av antioksidanter laget på industrielt potetskrell råstoff kan fungere som konserveringsmiddel og erstatte noen av de stoffene som brukes i dag. Tre næringsmiddelbedrifter skal teste potetskrell-ekstraktet i noen av sine produkter og utføre både interne og eksterne analyser på effekten. Men en viktig forutsetning for at potetantioksidant kan tas i bruk som nytt tilsetningsstoff i mat er at det blir godkjent av den Europeiske matsikkerhets myndigheten (EFSA) og gitt et E-nummer. Prosjektet skal igangsette denne lengre flertrinns godkjennings prosessen. Parallelt skal det utvikles en forretningsmodell og nødvendig beskyttelse rundt teknologien. Dersom industrien etter testingen av potetantioksidanten, vurderer denne som er god erstatning til konkurransedyktig pris, og en EFSA-godkjenning kan oppnås på sikt, åpner det for å kommersialisere et naturlig plante-basert produkt til beste for forbruker og for miljøet. Bak prosjektet Peel Preserve står Høgskolen i Innlandet i samarbeid med Nofima og Klosser Innovasjon.

Preserving foods with potato peels: Industry-relevant validation and first regulatory assessment of a new natural antioxidant

Applied Biocatalysis is the development through science and engineering of useful biological catalysts and their commercial applications. The BNCM 2024 conference has an intention to gather approx. 100 international and national scientists from academia and industry in addition to PhD students from the international community. BNCM 2024 will be the 7th in this series of meetings with attendees from both academia and industry presenting and discussing the newest research in enzyme catalysed reactions in other media than water. The conference series was started in 1986 by professors Hans Tramper and Urs von Stockar, University of Wageningen, the Netherlands, and has later been chaired in Russia, Italy and Germany before Elisabeth Jacobsen at NTNU was give the honor of chairing the conference.  Collaboration in biocatalysis research has been supported by EU Frameworks as three subsequent COST actions. The first action, COST Action D25 "Applied Biocatalysis. Stereoselective and environmentally friendly reactions catalysed by enzymes " (2001-2007) was initiated and chaired by professor Thorleif Anthonsen, NTNU, the founder of the Biocatalysis Group at NTNU, which the project leader of this conference now chair. The strategy of applied biocatalysis fits well under the definitions of white biotechnology and Sustainable chemistry (Suschem) and fits also well with the EU Green Deal Strategy.

Bilberry Ecology and the Regeneration of forest Ecosystem Functioning

Human impacts such as clearcut harvesting have left forests bereft of the organisms that underpin their functioning. The decline of key plant species has reduced the capacity of forests to store carbon (C) and provide ecosystem services to the people of Europe. The regeneration of forest ecosystem functioning requires a deeper understanding of how the traits of its species can be linked to ecosystem processes. The BEREFT project aims to elucidate how the plant traits of bilberry, Vaccinium myrtillus, modulate the dynamics of C and nitrogen (N) within the boreal forest. The project combines advances in chemical ecology with an innovative experimental design situated in Norway pairing experimental plots in formerly clearcut forest plantations with those from near-natural forests. I will collect a comprehensive set of bilberry's chemical traits and test both in-situ and in-vitro experiments to determine the underlying mechanism linking bilberry traits to C and N dynamics. The project goes beyond the state-of-the-art in applying novel methods from chemical ecology to measure bilberry's traits and uses a creative design to link those traits to ecosystem functioning. Through the scope of traits measured and the innovation in experimental design pairing clearcut forests with near-natural forests, this project will significantly advance our knowledge of the ecology of Europe's forest ecosystems. I will receive training in chemical ecology to advance my career as an expert in data generation and exchange with the host my knowledge on long-term ecological experimentation and ecosystem functioning. The project will disseminate deliverables in trait data packages, an open-source chemical ecology pipeline, and a robust measure of the effect size of bilberry's impact on C and N in the boreal forest. Knowledge of bilberry's role in the forest C cycle can be exploited by forestry managers in efforts to maximize C storage to meet the EU's goal of carbon neutrality.

EXtremeClimTwin aims to strengthen and improve the research and innovation capacity of the Faculty of Science, University of Novi Sad (UNSPMF; Novi Sad, Serbia) in the areas of climate change and hydro-climate extremes through twinning with internationally-leading counterparts from Member States of the EU: the SME and research company Climate Risk Analysis (CRA, Germany) and Loughborough University (LU, United Kingdom) and Associated country the Center for International Climate research (CICERO, Norway). It is expected that a warmer climate will result in a higher incidence of heat waves, less summer precipitation and at the same time higher rainfall intensities throughout much of South East Europe. Increasing heat extremes are a key hazard in this part of Europe. Together with reduced summer precipitation, they can increase drought risk, health risks and energy demand in summer. However, there is a knowledge gap in this part of Europe to properly understand how climate change will continue to affect the region. Cold waves, heatwaves and droughts in the summer, put a strain on the health system and the economy. Therefore, UNSPMF recognized the need to build capacities in detection and attribution of extreme hydro-climate events through collaborations and training with institutions that examine these events in other parts of Europe. The close collaboration between researches from UNSPMF and other experts in the field of climate change and hydro-climate extremes in the EU will form a synergistic network that will permit the transfer of knowledge and training of the existing personnel and early stage researchers of UNSPMF. As a result, the EXtremeClimTwin will have both direct and indirect scientific, social and economic impacts. In addition, the implementation of the project will facilitate future collaborations with experts in the field at EU level, increase the UNSPMF research capabilities, as well as enhance the research and academic profile of all participants.

Twinning for the advancement of data-driven multidisciplinary research into hydro-climatic extremes to support risk assessment and decision making

When an organism experiences physiological stresses, such as nutrient starvation, it activates several mechanisms to promote its survival. One of these mechanisms is autophagy, which is a catabolic pathway conserved from yeast to man that functions as a recycling system during normal physiology, and which helps provide nutrients to sustain essential cellular processes during starvation. Although autophagy is an intensively studied process that plays a major role both in normal development and in a wide variety of diseases, several important aspects of autophagy remain to be elucidated. For instance, whereas the vast majority of studies have focused on how cells regulate the activation of autophagy, it remains largely unknown how cells and organisms shut off autophagy in response to returning nutrient availability. The limited knowledge of autophagy termination is partly due to a lack of available tools and appropriate methods. Addressing these challenges will not only provide important insight in this fundamental aspect of the autophagic process, but may potentially also identify new targets for development of drugs to manipulate this phase of autophagy. In FINALphagy I will develop new genetic and computational tools to quantify and manipulate the temporal dynamics of autophagy in an entire organism. I will unravel key mechanism of autophagy termination in individual cells within a tissue and dissect how this connects to systemic signaling and behavioral changes. This will generate a comprehensive toolbox for time- and space-resolved manipulation and quantification of autophagy levels, produce a whole-organism map of autophagy responses at the tissue-level, and conceptualize our understanding of the dynamics of autophagy regulation in an entire organism during changing nutrient levels.

Celler er overlevere. I en levende organisme må celler klare å respondere på endringer i tilgang på næring. Når en organisme sulter vil enkelte organer respondere ved å aktivere en cellulær resirkuleringsprosess som kalles autofagi. Autofagi er en prosess hvor celler «spiser» sine egne ødelagte, ubrukte eller overfødige bestanddeler. Aktivering av autofagi frigjør energi og byggesteiner for å vedlikeholde uunnværlige funksjoner i cellen og beskytter organismen mot sykdommer som kreft og nevrodegenerering. Det er viktig at disse cellene også skrur av autofagi når næring igjen er tilgjengelig, fordi uhemmet autofagi er skadelig for cellens overlevelse. Mye er kjent om hvordan autofagi skrus på, men overraskende lite er kjent om hvordan autofagi skrus av, spesielt i en flercellet organisme. Dette prosjektet tar mål av å forstå mekanismene for hvordan autofagi skrus av, ved å bruke bananfluer.

For å nå vårt mål om å forstå hvordan autofagi skrus av kommer vi til å slå ned genuttrykk og studere effekten av dette på bananflueceller i cellekultur når de skrur av autofagi. Slik vil vi finne nye gener som regulerer hvordan autofagi skrus av. De mest spennende genene fra disse forsøkene vil bli studert videre i bananfluelarver. Disse larvene vil ha noen celler hvor genuttrykk er skrudd av, omgitt av mange normale celler. Vi vil følge larvene når de går gjennom perioder med sult og perioder hvor de fråtser i fersk mat. Vi ønsker spesifikt å studere den fysiologiske effekten av at autofagi skrus av på korrekt eller feilaktig måte. 

To postdoktorer har nå jobbet tre år på prosjektet, hvor den ene har optimalisert betingelser for å slå ned genuttrykk i bananflueceller i cellekultur og studere effekten av dette når cellene skrur av autofagi. Den andre postdoktoren har etablert metodene for å studere de samme genene i fettvevet til bananfluelarver. I begge systemer har vi nå etablert hvor raskt autofagi normalt skrus av når mat igjen er tilgjengelig. Vi har også optimalisert bildeanalysemetodene for å ekstrahere informasjon om autofagiprosessen fra bilder av bananflueceller i kultur og fra bilder av ulike bananfluevev under sult og tilbakeføring av mat. Vi har identifisert noen potensielle regulatorer av hvordan autofagi skrus av og har delvis identifisert mekanismene for hvordan disse fungerer. Vi jobber også med å forstå rollen til disse regulatorene under fysiologiske betingelser og i kreftutvikling.

Funn fra prosjektet har vært presentert på internasjonale konferanser i 2021 (Nordic Autophagy Society møte i Tromsø, Norge), 2022 (EMBO Autophagy konferanse i Eger, Ungarn), 2023 (European Drosophila Research Conference i Lyon, Frankrike).

I 2021 publiserte vi en artikkel om hvordan autofagi kan være en prosess som kan utnyttes i behandling av en form for kreft i hjernen. Dette er funn fra bananfluemodeller for denne krefttypen.
I 2023 publiserte vi en artikkel om en bananfluemodell for MLL-rearrrangert leukemi. Dette er en av modellene vi bruker videre for å se på rollen til gener som regulerer hvordan autofagi skrus av.
I 2023 publiserte vi også en artikkel med et datasett av annoterte bilder av Drosophila S2 celler i ulike stadier av autofagi. Disse kan brukes feltet til å utvikle analysemetoder, slik som med maskinlæring.

Basert på funn fra dette prosjektet har prosjektleder søkt om og fått tildelt et ERC Starting Grant som startet i 2023. Dette er en meget prestisjefull tildeling fra det Europeiske forskningsrådet. 

Konklusjonene fra dette prosjektet vil ikke bare være relevant for å forstå denne viktige prosessen i bananfluer, men for å forstå denne overlevelsesprosessen i alle levende organismer.

Cells in a living organism need to be able to readily respond to changes in nutritional status. Upon starvation, certain tissues respond by activating catabolic processes, such as autophagy, to provide energy and building blocks to sustain essential cellular functions. Importantly, these cells must also turn off autophagy when nutrient supplies are replenished, because unrestricted autophagy is harmful to cellular fitness. Surprisingly little is known about how autophagy is terminated, especially in the context of a multicellular organism. This project aims at uncovering novel mechanisms of autophagy termination, using fruit flies as a model. 

To reach this goal, my team will first identify novel regulators of autophagy termination among transcription factors and signaling pathway components through a RNAi-mediated high-content imaging-based screen in fly cells in culture. Top candidates from this screen will be further evaluated in vivo in fly larvae using clonal experiments where wild-type and mutant cells are juxtaposed within the same tissue. To obtain a detailed mechanistic understanding of one or two identified novel regulators of autophagy termination, my team will use epistasis analysis, evaluate their relationship to conserved signaling pathways, study morphological changes in autophagy-related membranes and investigate their subcellular localization. Finally, we will analyze the impact of defects in autophagy termination on developmental timing of fly larvae, fitness of adult flies and a cancer cachexia phenotype. 

I envisage that the proposed screening strategy and follow-up experiments will lead to fundamental breakthroughs in our understanding of autophagy termination. In addition, this “Young Research Talents” project will allow my transition to a project group leader and propel my career as an independent researcher.

Turning off autophagy in a multicellular organism

Ras er en liten GTPase som ofte er mutert i ulike kreftformer. All kunnskap om hvordan Ras påvirker cellene våre kan potensielt komme til nytte for å bekjempe kreft. Flere av signalveiene
nedstrøms for Ras er godt kjent, mens andre er mindre kartlagt. En av de sistnevnte involverer Ral, som selv er en liten GTPase. Prosjektets hovedmål er å beskrive Ras-Ral signaleringsaksen
på molekylært nivå. Strategien vi har valgt er å gjøre et screen i bananfluer for gener som påvirker en fenotype indusert av aktiv Ral. I den første delen av prosjektet har jeg uttrykt en aktiv form av Ral i ulike fluevev for å karakterisere fenotypen. Jeg har også uttrykt dominant negativ Ral eller satt Ral ut av funksjon ved hjelp av RNAi og analysert fenotypene som da oppstår. Videre har jeg verifisert at disse fenotypene avhenger av Ral og proteiner som allerede er kjent å interagere med Ral. I tillegg til genetiske verktøy har jeg også tatt i bruk biokjemiske verktøy med kultur av Drosophila S2 celler for å kunne svare på mer spesifikke spørsmål om hvordan Ral aktiveres. Gjennom mitt screen i har jeg funnet en tidligere ukjent rolle for Ral i normal utvikling av bananfluens blodsystem, noe som kan hjelpe oss til å forstå hvordan utviklingen av spesifikke celletyper foregår. I løpet av 2016 har disse funnene blitt presentert på en nasjonal konferanse og lokalt på Oslo Universitetssykehus med svært gode tilbakemeldinger. Disse resultatene klargjøres nå for publikasjon i et internasjonalt anerkjent tidsskrift. Prosjektet hadde også et delmål som går på å karakterisere proteinene CNK, Rap1 og Ral i utviklingen av øyet til bananfluen. I dette systemet kan vi undersøke effekten disse proteinene har på cellenes form, polaritet og utvikling. I juni 2014 publiserte vi resultater fra et screen for gener som påvirker en fenotype i flueøyet indusert av CNK. Genet som koder for Rap1 var en av kandidatene fra screenet og vi kunne vise at Rap1 aktivitet trengs for å vedlikeholde det apikale domenet av fotoreseptorceller under utvikling. Disse og andre relaterte resultater vil følges opp i fremtidige prosjekter.

Ras mutations are found in more than 30 % of human cancers. Therefore an urgent need persists for  identification and functional understanding of modulators of Ras signaling, since they are highly relevant as therapeutic targets. Ras signals through sever al downstream pathways that have already been delineated, including the Raf-MAPK, the PI3K-AKT and the Ral axis. Of these, the Ral pathway is the least studied, but emerging evidence for a role of Ral in Ras-induced oncogenesis has spurred a recent intere st in this pathway. To understand how the Ras-Ral signaling axis contributes to normal and malignant growth it is important to understand the spatiotemporal regulation of this pathway. This can be achieved by identification of genetic interactors of Ral t hat will bring new functional knowledge of the pathway and uncover potential therapeutic targets.
Drosophila is a powerful system to identify conserved genes that functionally interact with specific human oncogenes. As a homologue of Ral is also found in  flies, we anticipate that expression of Ral during development affects some of the same protein networks that are perturbed in human cancers. Therefore we will use the phenotype induced by expression of constitutively active Ral during eye development to  isolate genetic modifiers of this phenotype. We expect the identification and characterization of these modifiers will lead to an enhanced understanding of the spatiotemporal regulation of the Ras-Ral signaling axis, potentially uncovering new therapeutic  targets to the benefit of cancer patients.

Functional understanding of Ras-Ral signaling in development and cancer

The aim of my exchange in Tom Neufelds lab is to study the role of the SNX18 fly homolog dSH3PX1 in vivo as part of my PhD thesis. The exchange period is scheduled from August 1st 2011 to December 31st 2011 (5 months). The main goal of my PhD thesis is to  identify novel PI3P binding proteins required for autophagy. To this end, I have performed a high-trough-put siRNA screen, using a siRNA library designed to individually deplete 80 putative PI3P-binding proteins containing PX or FYVE domains. Since the s uccessful completion of the screen, I am now at the stage of characterizing candidates from the screen in mammalian cell culture experiments. One of the best candidates from the screen is SNX18, a PX and BAR domain containing protein. siRNA-mediated deple tion of this protein strongly inhibits autophagy. It cellular localization is reminiscent of the core autophagic protein mAtg9 and our results show that mAtg9 and SNX18 colocalize. In mammals SNX18 belongs to a family with two closely related family membe rs, SNX9 and SNX33. SNX9 has a well established function in endocytosis. In Drosophila melanogaster, there is only one homolog of the SNX9/18/33 family, called dSH3PX1, which has been shown to function in endocytosis similarly to mammalian SNX9, but its r ole in autophagy has not been studied. This project aims at dissecting the role of dSH3PX1 in autophagy by employing dSH3PX1 mutant fly lines in fat body assays, neurodegeneration assays and by monitoring lifespan. The lab of Tom Neufeld provides the opti mal environment for performing these experiments as it has been one of the major contributors to the field using the fly as a model organism. A stay in his lab gives access to all the best established autophagy assays in flies as well as relevant fly line s and top-end facilities.
