CpG ODN as adjuvant in recombinant infectious pancreatic necrosis virus vaccines

A high priority target in current vaccinology is the identification of adjuvants that promote stable induction of Th1 immunity but with minimal side effects. Unmethylated CpG oligodinucleotides (ODNs) within specific flanking bases (referred to as CpG mot if) are relatively abundant in bacterial DNA and are known to stimulate innate immune responses. CpG motif show promise as vaccine adjuvant in animal models, and studies by our group have shown that certain CpG motifs induce cytokine expression and stimul ate cell proliferation of Atlantic salmon leucocytes. Infectious pancreatic necrosis (IPN) is one of the most threatening viral diseases in Norwegian aquaculture, and development of effective vaccines, as well as knowledge of protective mechanisms against this disease are important research issues to be addr essed. We have shown that particular CpG motifs induce protection against IPN virus. The aim of this project is to study efficacy of CpG motif as adjuvant for recombinant IPNV vaccines in Atlantic salmon, and to develop delivery system that enhances its i mmunostimulatory effects. Knowledge about the various processes explaining how the immune system becomes activated by the adjuvant will also be addressed.
