Development of a cDNA based reverse genetics system for infectious pancreatic necrosis virus (IPNV)

Principle objectives: The principal objectives are to establish a cDNA based reverse genetics system and use this system to verify or map viral factors of importance for pathogenicity in IPNV-infections Project sub-goals 1. Map the extreme 3' and 5' ends of the genomic segments of the chosen IPNV isolates 2. Construction of the plasmids that contain the genomic segments of IPNV 3. Transfection of these plasmids in cells expressing T7 polymerase, for production of infective IPNV particles 4. Manipulation of the VP2 gene (or other gene candidates) by in vitro mutagenesis
