The focus of the proposed research is to study mechanisms governing the pathogenicity of VHS virus infections. Our proposal is to employ a reverse genetics approach and make an infectious clone encoding the entire genome of VHSV and thereby create a basis for a detailed examination of the virulence traits and pathogenicity of VHS virus. During the pre-proposal period the main aim will be to carry out the work to show that an infectious clone can be made for VHS virus, as has been done for infectious hemat opoietic necrosis virus by the same partner (P2). This will create a basis for further studies into basic pathogenic mechanisms of VHS virus. It will also create a sound basis for development of immunoprophylactic measures to control one of the most devas tating diseases in a number of fish species worldwide.
