The present project concerns a research visit of a young researcher working at Belgrade University to the Norwegian University of Life Sciences in Ås. The visiting researcher is working in an allergology centered laboratory, and is working on recombinant allergens. The host lab in Norway has extensive experience in using food-grade lactic acid bacteria for display of antigens. The goal of the exchange project is to develop new desensitization strategies by combining the visiting researchers knowledge with the innovative lactic acid bacteria-based display technologies of the host laboratory. The host laboratory is already conducting research on lactic acid bacteria-based vaccines, while use of these bacteria for desensitization (instead of vaccination) in fact may be the most promising application of the novel display technology. Through the project, the host laboratory will get access to valuable new application routes for their display technology, learn more about allergy and desensitization strategies, and gain access to relevant mouse models. The visiting scientist will be trained in advanced display technologies and molecular microbiology in general. The experimental work will of three parts: (1) Secreting and anchoring relevant antigens in Lactobacil lus using technology developed in the host laboratory, (2) Analyze antigen expression and localization in recombinant Lactobacillus strains and select optimal constructs (optimal expression levels and/or optimal targeting efficiency), and (3) Analyzing t he in vivo functionality of the newly developed Lactobacillus-based vectors in mouse models using both oral and nasal administration routes (to be done mainly in the home lab of the visiting scientist, after her return).
