Development of new analytical and preparative techniques for enantioselective resolution of Stiripentol

The main goal of the work is the biocatalyzed kinetic resolution of stiripentol through three ways 1- Lipase mediated reaction (esterification or hydrolysis) 2- biocatalyzed reduction of the prochiral ketone 3- biocatalyzed oxidation of the racemic alc ohol. A mandatory prerequisite for the biocatalyzed enantioselective reactions is the analytical chiral separation of the enantiomers of the reaction substrate and product. This could be performed using chiral chromatography and/or electrophoresis. So a t the inception of the work analytical chiral separation techniques for the reactions substrates and products will be optimized. Then many trials will be performed using different lipases (for esterification or hydrolysis of the racemic alcohol), differen t alcohol dehydrogenases (for reduction of the prochiral ketone) and different reaction conditions (solvent, catalyst concentration) till achieving the most promising results which will be used on gram scale resolution of the drug.
