This proposed, integrated project is centred on understanding the neurobiology of Attention-Deficit/Hyperactivity Disorder (ADHD). Recent advances in modern neuroscience make it possible to make a focused attack on the topic. The proposal contains a multi disciplinary approach with different levels of basic neuroscientific analysis (genetics, neurophysiology, neurochemistry, behaviour, neurotoxicology). Existing animal strains and strains developed by gene-targeting techniques will be deployed to reveal in sights into how dopaminergic malfunction may represent a central factor in ADHD. In addition, at a translational level we will work through a network collaboration with a planned longitudinal human case-control study and a clinical neuropsychological stud y. The objectives are: 1. Strengthen the scientific bases for understanding brain functions underlying ADHD including factors that may increase its prevalence. Dopamine hypofunctioning is currently believed to be the major factor causing ADHD. We will th erefore investigate ways dopamine, and candidate genes affecting dopamine function, alter signal transmission. 2. Acquire knowledge about neurotoxins that affect dopamine functioning and thereby may increase the prevalence of ADHD. 3. Investigate fundame ntal behavioural processes, especially reinforcement (reward) and extinction, which, if deficient, may lead to development of ADHD symptoms. Data and insights obtained in objectives 1-4 will be critical for future scientifically-based prevention strategie s and treatments of ADHD. In order to obtain these long-term objectives, we want to: 5. Establish the core of a basic, cross-disciplinary research group on ADHD at the University of Oslo. 6. Develop translational neurobiological and behavioural strategie s for such research.
